    Transient apoptosis inhibition in donor stem cells improves hematopoietic stem cell transplantation

    During hematopoietic stem cell transplantation, a substantial number of donor cells are lost because of apoptotic cell death. Transplantation-associated apoptosis is mediated mainly by the proapoptotic BCL-2 family proteins BIM and BMF, and their proapoptotic function is conserved between mouse and human stem and progenitor cells. Permanent inhibition of apoptosis in donor cells caused by the loss of these BH3-only proteins improves transplantation outcome, but recipients might be exposed to increased risk of lymphomagenesis or autoimmunity. Here, we address whether transient inhibition of apoptosis can serve as a safe but efficient alternative to improve the outcome of stem cell transplantation. We show that transient apoptosis inhibition by short-term overexpression of prosurvival BCL-XL, known to block BIM and BMF, is not only sufficient to increase the viability of hematopoietic stem and progenitor cells during engraftment but also improves transplantation outcome without signs of adverse pathologies. Hence, this strategy represents a promising and novel therapeutic approach, particularly under conditions of limited donor stem cell availability

    Durability Enhancement Of SCC With Waste Glass Powder

    Self compacting concrete (SCC) requires a large quantity of fine materials compared to common concrete. In this work waste glass powder was used to replace (50%) of filler required. Two types of SCC were manufactured, a control SCC type (CTL) using cement and limestone filler and another with glass powder replacing 50% of the filler. Mechanical and durability properties, namely, compressive strength, resistivity, chloride ion penetration, carbonation, capillary water absorption and oxygen permeability were assessed on both SCC types. It could be concluded that waste glass powder can be used successfully in SCC improving chloride penetration and water absorption by capillarity and maintaining strength levels. The potential risk of alkali-silica observed on mortar was mitigated when incorporating glass powder

    DNA polymerase requirements for parvovirus H-1 DNA replication in vitro

    An in vitro system using nuclei from parvovirus H-1-infected cells was used to characterize the influence of inhibitors of mammalian DNA polymerases on viral DNA synthesis. The experiments tested the effects of aphidicolin, which is highly specific for DNA polymerase alpha, and 2',3'-dideoxythymidine-5'-triphosphate (ddTTP), which inhibits cellular DNA polymerases in the order gamma greater than beta greater than alpha. Both aphidicolin and ddTTP were inhibitory, indicating that both polymerase alpha and a ddttp-sensitive enzyme are required for viral DNA synthesis. This was seen more clearly in kinetic measurements, which indicated an initial period of rapid DNA synthesis with the participation of polymerase alpha, followed by a period of less rapid, but more sustained, rate of DNA synthesis carried out by a ddTTP-sensitive enzyme, probably polymerase gamma. One interpretation of the results is that polymerase alpha functions in a strand displacement stage of the viral DNA replication mechanism, whereas polymerase gamma serves to convert the displaced single strands back to double-strand replicative form.</jats:p

    This paper presents selected applications of the miniaturized hydraulic components offered by specialized manufacturers and some results of the authors' own research on microflows, including results of hydraulic microfeeder vibration measurements performed by the touchless method using a laser vibrometer. The latter was chosen in order to eliminate measuring instrument influence on the investigated microhydraulic object. Special attention was focused on acoustic problems: noise sources and methods of noise reduction.W pracy przedstawiono wybrane przykłady zastosowań zminiaturyzowanych elementów hydraulicznych proponowanych przez wyspecjalizowanych producentów oraz podano wyniki niektórych prac własnych nad zjawiskami związanymi z mikroprzepływami. W artykule prezentuje się m.in. wyniki pomiarów drgań zaworu przelewowego mikrozasilacza hydraulicznego wykonane za pomocą wibrometru laserowego - metodą bezdotykową. Wykorzystanie do pomiarów w obszarze mikrohydrauliki wibrometru laserowego podyktowane było koniecznością wyeliminowania wpływu przyrządów pomiarowych na badany obiekt. Szczególną uwagę skupiono na problemach akustycznych: źródłach hałasu i metodach jego redukcji
