Triterpene Saponins from the Aerial Parts of Trifolium medium L. var. sarosiense
Seven previously unreported triterpene glycosides (1−7) were isolated from methanol extract of the aerial parts of
Trifolium medium var. sarosiense (zigzag clover). Their structures were established by the extensive use of 1D and 2D NMR experiments along with ESI-MS and HRMS analyses. Compounds 1−7 are oleanane derivatives characterized by the presence of a keto group at C-22 of an aglycone and a primary alcoholic function at C-24 and differing functions at C-30. Among these, compounds 1−3 and 6 showed a secondary alcoholic function at C-11, which is methoxylated in compounds 4 and 7. Compound 5 was shown to possess a known aglycone, wistariasapogenol A; however, it is described here for the first time as a saponin constituent of the Trifolium genus. Some aspects of taxonomic classification of zigzag clover are also discussed
Antifungal [Gaeumannomyces graminis var. tritici] activity of various glycosides of medicagenic acid
Different concentrations of medicagenic acid and five glycosides of this acid isolated from alfalfa (Medicago sativa) were added to agar medium (com meal agar, CMA) inoculated with cultures of Gaeumannomyces graminis var. tritici (Ggt). After 7 days of incubation at 25°C colony radius was measured and % of inhibition calculated in relation to the control medium (CMA enriched with the solvent of the tested compounds). Within the tested concentrations, only 3-O-β -D -glucopiranoside medicagenate (monoglucoside) significantly reduced the growth of Ggt on CMA medium. This compound at 0.05 mM concentration completely inhibited the development of the fungus and the effect was shown to be fungi-toxic.Różne koncentracje kwasu medikagenowego (KM) oraz pięciu glikozydów tego kwasu wyodrębnionych z lucerny siewnej (Medicago sativa) dodawano do pożywki agarowej (corn meal agar, CMA) zaszczepionej kulturą Gaeumannomyces graminis var. tritici (Ggt). Po różnych okresach inkubacji mierzono promień kolonii badanego grzyba i wyliczano % zahamowania jego wzrostu w stosunku do kontroli (CMA z dodatkiem rozpuszczalnika testowanych związków). Spośród analizowanych związków tylko monoglukozyd KM (3-O-β -D -glukopiranozyd KM) istotnie hamował wzrost Ggt w zakresie użytych koncentracji. Związek ten w stężeniu 50 µM spowodował zupełne zahamowanie rozwoju badanego grzyba, a po przełożeniu inokulum na pożywkę czystą, grzyb ten nie był zdolny do ponownego wzrostu. Wskazuje to, że w powyższej koncentracji monoglukozyd KM wykazuje efekt grzybobójczy w stosunku do Ggt
Calcium-Magnesium salt of saponins from bird's foot trefoil seeds (Lotus corniculatus L.)
A calcium-magnesium salt of saponins was isolated from bird's foot trefoil seeds with a yield of 0.04%. The salt contained: 1.37% - Ca and 0.04% - Mg. The saponin salts were deionized on IR-120 Amberlite and free crystalline saponin was obtained. In both cases the presence of one major component and of three trace ones was found, using TLC. In acid hydrolysates of the saponin salts and of the free saponin the following sugars were identified: rhamaose, xylose, glucose, galactose, glucuronic acid. From among aglycones soyasapogenols B, C, traces of E, and of one unidentified in both cases were found. The Ca-Mg salt obtained seems to be a natural form of saponins in bird's foot trefoil seeds
Oddziaływanie saponin z Medicago arabica, M. hybrida i M. sativa na wzrost i rozwój Fusarium oxysporum f. sp. tulipae
In the present work it was shown that total saponins
originated from M. hybrida and M. sativa substantially limited
mycelium growth of F. oxysporum f. sp. tulipae and symptoms
of fusariosis on tulip bulbs. Out of 15 individual tested saponins
originated from M. arabica, M. hybrida and M. sativa, four compounds:
3-O-[β-D-glucopyranosyl (1→2)α-L-arabinopyranosyl]
hederagenin, hederagenin 3-O-β-D-glucopyranoside, medicagenic
acid, medicagenic acid 3-O-β-D-glucopyranoside had
the strongest inhibitory effect on mycelium growth of Fusarium
oxysporum f. sp. tulipae on PDA medium. The total saponins
from M. arabica, M. hybrida and M. sativa inhibited the number
of colony forming units of Fusarium oxysporum f. sp. tulipae
in artifi cially infested substrate. The use of saponins originated
from Medicago as a fungicide is suggested.Wykazano, że suma saponin pozyskana z korzeni

0,05% i 0,1% oddziaływała fungitoksycznie na wzrost
grzybni 4 izolatów Fusarium oxysporum f. sp. tulipae
(F.ox.t.) w warunkach in vitro na pożywce PDA. Najwyższe
stężenie saponin (0,1%) najsilniej ograniczało
wzrost grzybni badanych izolatów F.ox.t.; inhibicja
wynosiła w granicach 73-80% dla saponin izolowanych
z M. hybrida i 46–65% dla saponin z M. sativa.
Spośród badanych 15 poszczególnych pojedyńczych
saponin otrzymanych z M. arabica, M. hybrida
i M. sativa 4 następujące saponiny wykazały silne haO-[β-D-glukopiranozylo(1→2)α-L-arabinopiranozyd]
hederageniny (M. arabica) – w stężeniu 140 μgxcm־³
70% hamowanie, 3-O-β-D-glukopiranozyd hederageniny
( M. hybrida) – w stężeniu 100 μgxcm־³ 80%
hamowanie, kwas medikagenowy (M. sativa) – w stężeniu
60 μgxcm־³ 85% hamowanie, 3-O-β-D-glukopiranozyd
kwasu medikagenowego (M. hybrida, M. sativa)
– w stężeniu 115 μgxcm־³ 65% hamowanie.
Suma saponin pozyskana z M. hybrida i M. sativa
zastosowana profi laktycznie do zaprawiania cebul
tulipana silnie ograniczała rozwój fuzariozy. W stężeniu
0,1% suma saponin z każdego badanego gatunku
lucerny ograniczała rozwój choroby w około 80%,
oceniany długością i głębokością plam infekcyjnych
i hamowała tworzenie się gum. Zastosowane sumy
saponin w wyższym stężeniu 0,5% silniej ograniczały
rozwój patogena.
Badane sumy saponin, zastosowane w stężeniu
5%, do podlewania zakażonego przez F.ox.t. 218
podłoża, wykazały inhibicyjny, jednakże zróżnicowany
wpływ na rozwój patogena w podłożu. Najsilniej
ograniczała liczbę jednostek tworzących kolonie F.ox.
t 218, suma saponin z M. sativa (około 92%), słabsze
wyniszczające oddziaływanie na patogena wykazała
M. hybrida (około 60%) a najsłabiej ograniczała rozwój
patogena suma saponin z M. arabica (około 40%)
– w porównaniu do liczebności jednostek tworzących
kolonie F.ox.t. 218 w podłożu kontrolnym nie traktowanym
Flavonoids from the bird's foot trefoil seeds (Lotus corniculatus L.)
In the seeds of bird's foot trefoil (Lotus corniculatus L.) the presence of quercetin- and kaempferol heterosides was found. Eight crystalline compounds and one chromatographically homogeneous fraction were obtained from the flavonoid mixture using column chromatography on polyamide and cellulose. By means of TL-chromatography, elemental analysis and UV-spectrometry seven of the isolated compounds were identified to be quercetin, quercetin 3-arabopyranoside (guajaverin), quercetin 3-galactoside, quercetin 3-rhamnoside, kaempferol 3,7-diglucoside, kaempferol 3,7-dirhamnoside kaempferol 3-glucosyl-7-rhamnoside and two remaining ones were only partly characterized a snearer indefinite kaempferol heterosides

Antifungal activity of total saponins originated from roots of Medicago hybrida
(Pourret) Trautv. were evaluated in vitro against six pathogenic fungi and eight
individual major saponin glycosides were tested against one of the most susceptible
fungi. The total saponins showed fungitoxic effect at all investigated concentrations
(0.01%, 0.05% and 0.1%) but their potency was different for individual fungi. The
highest saponin concentration (0.1%) was the most effective and the inhibition of
Fusarium oxysporum f. sp. callistephi, Botrytis cinerea, Botrytis tulipae, Phoma narcissi,
Fusarium oxysporum f. sp. narcissi was 84.4%, 69.9%, 68.6%, 57.2%, 55.0%,
respectively. While Fusarium oxysporum Schlecht., a pathogen of Muscari armeniacum,
was inhibited by 9.5% only. Eight major saponin glycosides isolated from the
total saponins of M. hybrida roots were tested against the mycelium growth of Botrytis
tulipae. The mycelium growth of the pathogen was greatly inhibited by hederagenin
3-O-β -D-glucopyranoside and medicagenic acid 3-O-β -D-glucopyranoside. Medicagenic
acid 3-O-β -D-glucuronopyranosyl-28-O-β -D-glucopyranoside and oleanolic
acid 3-O-[β -D-glucuronopyranosyl(1→2)-α -L-galactopyranosyl]-28-O-β -D-glucopyranoside
showed low fungitoxic activity. Medicagenic acid 3-O-β-D-glucopyranosyl-
28-O-β -D-glucopyranoside, hederagenin 3-O-[α -L-rhamnopyranosyl(1→-2)-D-glucopyranosyl(
1→2)-D-glucopyranosyl]- 28-O-β -D-glucopyranoside and hederagenin
3-O-α -D-glucuronopyranosyl-28-O-β -D-glucopyranoside did not limit or only
slightly inhibited growth of the tested pathogen. While 2β, 3β-dihydroxyolean-12
ene-23-al-28-oic acid 3-O-β -D-glucuronopyranosyl-28-O-β -D-glucopyranoside slightly

cych glikozydów saponinowych względem Botrytis cinerea. Suma saponin wykazała
fungistatyczny wpływ przy wszystkich badanych stężeniach (0,01%, 0,05% i 0,1%)
ale ich aktywnooeć była różna dla poszczególnych grzybów. Najwyższe stężenie (0.1%)
saponin było najbardziej efektywne i wyrażało się silnym hamowaniem Fusarium
oxysporum f. sp. callistephi (84,4%), Botrytis cinerea (69,9%), Botrytis tulipae (68,6%),
Phoma narcissi (57,2%), Fusarium oxysporum f. sp. narcissi (55,0%). Podczas, gdy
Fusarium oxysporum Schlecht, patogen pochodzący z Muscari armeniacum był zahamowany
zaledwie w 9,5%. Osiem dominujących glikozydów saponinowych wyodrębnionych
z sumy saponin testowano w stosunku do Botrytis tulipae. Wzrost liniowy
grzybni tego patogena był silnie zahamowany przez dwa glikozydy: 3-O-β-Dglukopyranozyd
hederageniny i 3-O-β -D-glukopyranozyd kwasu medikagenowego.
3-O-β-D-glukuronopyranozylo-28-O-β-D-glukopyranozyd kwasu medikagenowego
i 3-O-[-D-glukuronopyranozylo(1→2)- α -L-galaktopyranozylo]-28-O-β -D-glukopyranozyd
kwasu oleanolowego wykazywały niską fungitoksycznooeć. 3-O-β -D-glukopyranozylo-
28-O-β -D-glukopyranozyd kwasu medikagenowego, 3-O-[α-L-rhamnopyranozylo(
1→2)-β -D-glukopyranozylo(1→2)-D-glukopyranozylo]-28-O-β-Dglukopyranozyd

Evaluation of nematicidal properties of saponins from Medicago spp
The nematicidal activity of saponins from Medicago arborea (tops), M. arabica (tops and roots) and M. sativa (tops and roots) against the plant-parasitic nematode Xiphinema index was investigated. Nematicidal activity of related prosapogenins and sapogenins on X. index is also described. Saponins from Medicago spp. at different concentrations were all nematicidal, those from M. arborea tops being the less effective. In general, saponins induced 100% mortality at 500 mu g ml(-1) between 8 and 48 h, while prosapogenins resulted in toxicity starting at 125 mu g ml(-1). Differences in the effects on X. index induced by prosapogenins and sapogenins were less pronounced, although prosapogenins displayed a larger range of activity. Assays with purified sapogenins demonstrated the relationship of the observed nematicidal activity of M. sativa and M. arborea to the content of the main aglycones (medicagenic acid and hederagenin, respectively) in the saponin extracts. Hederagenin displayed the highest bioactivity, giving 38% mortality after 1 h at 125 mu g ml(-1)
