<p>Abstract</p> <p>Background</p> <p>Quercetin derivatives in onions have been regarded as the most important flavonoids to improve diabetic status in cells and animal models. The present study was aimed to examine the hypoglycemic and insulin-sensitizing capacity of onion peel extract (OPE) containing high quercetin in high fat diet/streptozotocin-induced diabetic rats and to elucidate the mechanism of its insulin-sensitizing effect.</p> <p>Methods</p> <p>Male Sprague-Dawley rats were fed the AIN-93G diet modified to contain 41.2% fat and intraperitoneally injected with a single dose of streptozotocin (40 mg/kg body weight). One week after injection, the rats with fasting blood glucose levels above 126 mg/dL were randomly divided into 4 groups to treat with high fat diet containing 0 (diabetic control), 0.5, or 1% of OPE or 0.1% quercetin (quercetin equivalent to 1% of OPE) for 8 weeks. To investigate the mechanism for the effects of OPE, we examined biochemical parameters (insulin sensitivity and oxidative stresses) and protein and gene expressions (pro-inflammatory cytokines and receptors).</p> <p>Results</p> <p>Compared to the diabetic control, hypoglycemic and insulin-sensitizing capability of 1% OPE were demonstrated by significant improvement of glucose tolerance as expressed in incremental area under the curve (<it>P </it>= 0.0148). The insulin-sensitizing effect of OPE was further supported by increased glycogen levels in liver and skeletal muscle (<it>P </it>< 0.0001 and <it>P </it>= 0.0089, respectively). Quantitative RT-PCR analysis showed increased expression of insulin receptor (<it>P </it>= 0.0408) and GLUT4 (<it>P </it>= 0.0346) in muscle tissues. The oxidative stress, as assessed by superoxide dismutase activity and malondialdehyde formation, plasma free fatty acids, and hepatic protein expressions of IL-6 were significantly reduced by 1% OPE administration (<it>P </it>= 0.0393, 0.0237, 0.0148 and 0.0025, respectively).</p> <p>Conclusion</p> <p>OPE might improve glucose response and insulin resistance associated with type 2 diabetes by alleviating metabolic dysregulation of free fatty acids, suppressing oxidative stress, up-regulating glucose uptake at peripheral tissues, and/or down-regulating inflammatory gene expression in liver. Moreover, in most cases, OPE showed greater potency than pure quercetin equivalent. These findings provide a basis for the use of onion peel to improve insulin insensitivity in type 2 diabetes.</p

AIM: To evaluate the efficacy and safety of three different concentrations of diluted atropine for the control of myopia in Korean children, and to assess the risk factors associated with rapid myopia progression. METHODS: A total of 285 children, with refractive errors within the range of -6 diopters (D) between 5 and 14 years of age were included. After using 0.01%, or 0.025%, or 0.05% atropine, for about 1y, changes in refraction, axial lengths and frequency of adverse events were analyzed. Logistic regression analyses were performed to evaluate the risk factors associated with rapid myopia progression. RESULTS: The changes in the mean spherical equivalent values were -0.134 D/mo in the before atropine group, -0.070 D/mo in the 0.01% atropine group, -0.047 D/mo in the 0.025% atropine group, and -0.019 D/mo in the 0.05% atropine group, with significant differences between the groups (P<0.001). The axial elongation was 0.046 mm/mo, 0.037 mm/mo, 0.025 mm/mo, and 0.019 mm/mo respectively, with significant differences between the groups (P=0.003). The incidence of photophobia and near vision difficulty was not different among the three atropine groups (P=0.425 and P=0.356, respectively). Multivariate logistic regression analyses showed that only highly myopic parents were a significant predictive factor of rapid myopia progression in Korean children (odds ratio, 8.155; 95% confidence interval, 3.626-18.342; P<0.001). CONCLUSION: Treatment with 0.01%, 0.025% and 0.05% atropine solution inhibits myopia progression in Korean children in a dose-dependent manner. Children with highly myopic parents preferentially shows a rapid myopia progression rate

Comparison of Dst Forecast Models for Intense Geomagnetic Storms

We have compared six disturbance storm time (Dst) forecast models using 63 intense geomagnetic storms (Dst <=100 nT) that occurred from 1998 to 2006. For comparison, we estimated linear correlation coefficients and RMS errors between the observed Dst data and the predicted Dst during the geomagnetic storm period as well as the difference of the value of minimum Dst (Delta Dst(sub min)) and the difference in the absolute value of Dst minimum time (Delta t(sub Dst)) between the observed and the predicted. As a result, we found that the model by Temerin and Li gives the best prediction for all parameters when all 63 events are considered. The model gives the average values: the linear correlation coefficient of 0.94, the RMS error of 14.8 nT, the Delta Dst(sub min) of 7.7 nT, and the absolute value of Delta t(sub Dst) of 1.5 hour. For further comparison, we classified the storm events into two groups according to the magnitude of Dst. We found that the model of Temerin and Lee is better than the other models for the events having 100 <= Dst < 200 nT, and three recent models (the model of Wang et al., the model of Temerin and Li, and the model of Boynton et al.) are better than the other three models for the events having Dst <= 200 nT

The efficacy of the combination vaccine of the individual C-terminal fragments of ApxIA, ApxIIA and ApxIIIA of Actinobacillus pleuropneumoniae (APP) was evaluated in piglets. Twenty piglets were divided equally into 2 groups (n=10). All piglets were intramuscularly primed at 4 week-of-age (0 week post prime inoculation (WPPI)) and were intramuscularly boosted at 6 week-of-age (2 WPPI). Group A piglets were inoculated with sterile PBS and group B piglets were inoculated with the combination vaccine. Concentrations of each of the C-terminal fragment-specific IgG as determined by ELISA were significantly higher in group B than in group A from 2 WPPI until the end of this study. Clinical signs were observed from only 10% of group B piglets after the challenge with the mixture of APP serotypes 1, 2 and 5 at 4 WPPI, while 50% of group A piglets were protected against APP infections. Overall, intramuscular inoculation with the vaccine candidate can efficiently protect piglets against APP infection

KINEMATIC COMPARISONS OF KETTLEBELL TWO-ARM SWINGS BETWEEN EXPERTS AND BEGINNERS

The purpose of this study was to investigate kinematic comparisons of kettlebell two-arm swings between experts and beginners in order to find out biomechanical key points for preventing sports injuries and enhancing kettlebell performance. Four experts and three beginners performed kettlebell two-arm swings fifteen times with a 16 kg kettlebell. Experts demonstrated larger ranges of motions (ROM) of pelvic segment and hip joint than beginners, while beginners revealed larger ROM of shoulder joint than experts. Magnitudes and sequential orders of peak angular velocities of major joints were significantly different between two groups. Conclusively, the mobility of pelvic segment and hip joint are required, while the stability of the other joint are needed to produce appropriate kettlebell two-arm swings. The activation and strength of gluteus muscles would be key contributors

Dihydrotestosterone-Inducible IL-6 Inhibits Elongation of Human Hair Shafts by Suppressing Matrix Cell Proliferation and Promotes Regression of Hair Follicles in Mice

Autocrine and paracrine factors are produced by balding dermal papilla (DP) cells following dihydrotestosterone (DHT)-driven alterations and are believed to be key factors involved in male pattern baldness. Herein we report that the IL-6 is upregulated in balding DP cells compared with non-balding DP cells. IL-6 was upregulated 3hours after 10–100nM DHT treatment, and ELISA showed that IL-6 was secreted from balding DP cells in response to DHT. IL-6 receptor (IL-6R) and glycoprotein 130 (gp130) were expressed in follicular keratinocytes, including matrix cells. Recombinant human IL-6 (rhIL-6) inhibited hair shaft elongation and suppressed proliferation of matrix cells in cultured human hair follicles. Moreover, rhIL-6 injection into the hypodermis of mice during anagen caused premature onset of catagen. Taken together, our data strongly suggest that DHT-inducible IL-6 inhibits hair growth as a paracrine mediator from the DP

Downbeat, Positional, and Perverted Head-Shaking Nystagmus Associated with Lamotrigine Toxicity

Lamotrigine is an effective antiepileptic drug with few adverse effects. Nystagmus is one of the commonly observable signs of lamotrigine overdose, which may result from central nervous system involvement. However, the physiologic basis of lamotrigine-induced nystagmus is not fully understood. Here we report a patient who presented with lamotrigine-associated nystagmus that was probably related to cerebellar dysfunction
