    Varicose: a MAGUK required for the maturation and function of Drosophila septate junctions

    <p>Abstract</p> <p>Background</p> <p>Scaffolding proteins belonging to the membrane associated guanylate kinase (MAGUK) superfamily function as adapters linking cytoplasmic and cell surface proteins to the cytoskeleton to regulate cell-cell adhesion, cell-cell communication and signal transduction. We characterize here a <it>Drosophila </it>MAGUK member, Varicose (Vari), the homologue of vertebrate scaffolding protein PALS2.</p> <p>Results</p> <p>Varicose localizes to pleated septate junctions (pSJs) of all embryonic, ectodermally-derived epithelia and peripheral glia. In <it>vari </it>mutants, essential SJ proteins NeurexinIV and FasciclinIII are mislocalized basally and epithelia develop a leaky paracellular seal. In addition, <it>vari </it>mutants display irregular tracheal tube diameters and have reduced lumenal protein accumulation, suggesting involvement in tracheal morphogenesis. We found that Vari is distributed in the cytoplasm of the optic lobe neuroepithelium, as well as in a subset of neuroblasts and differentiated neurons of the nervous system. We reduced <it>vari </it>function during the development of adult epithelia with a partial rescue, RNA interference and generation of genetically mosaic tissue. All three approaches demonstrate that <it>vari </it>is required for the patterning and morphogenesis of adult epithelial hairs and bristles.</p> <p>Conclusion</p> <p>Varicose is involved in scaffold assembly at the SJ and has a role in patterning and morphogenesis of adult epithelia.</p

    An African Salmonella Typhimurium ST313 sublineage with extensive drug-resistance and signatures of host adaptation

    Abstract: Bloodstream infections by Salmonella enterica serovar Typhimurium constitute a major health burden in sub-Saharan Africa (SSA). These invasive non-typhoidal (iNTS) infections are dominated by isolates of the antibiotic resistance-associated sequence type (ST) 313. Here, we report emergence of ST313 sublineage II.1 in the Democratic Republic of the Congo. Sublineage II.1 exhibits extensive drug resistance, involving a combination of multidrug resistance, extended spectrum β-lactamase production and azithromycin resistance. ST313 lineage II.1 isolates harbour an IncHI2 plasmid we name pSTm-ST313-II.1, with one isolate also exhibiting decreased ciprofloxacin susceptibility. Whole genome sequencing reveals that ST313 II.1 isolates have accumulated genetic signatures potentially associated with altered pathogenicity and host adaptation, related to changes observed in biofilm formation and metabolic capacity. Sublineage II.1 emerged at the beginning of the 21st century and is involved in on-going outbreaks. Our data provide evidence of further evolution within the ST313 clade associated with iNTS in SSA

    Making it last: Combating hedonic adaptation in romantic relationships

    Is the waning of passion and satisfaction in romantic relationships inevitable, or can the honeymoon period be sustained? The Hedonic Adaptation Prevention model, which describes the mechanisms by which people adapt to positive life changes, posits that hedonic adaptation is a powerful barrier to sustained relationship well-being and suggests how to thwart it. In this paper, we apply the model to a new area of study - namely, intimate relationships. We explore the practices, habits, and activities that can increase the number of positive events and emotions in relationships, boost their variety, lower a couple's entitled aspirations, and build their appreciation - all variables that can serve to slow adaptation and increase well-being. Additionally, we discuss types of romantic relationships (e.g. long-distance relationships and unhealthy relationships) that may be relatively less susceptible to hedonic adaptation. © 2013 Copyright Taylor and Francis Group, LLC

    Servicio Social ComunitarioInvestigación realizada tuvo como objetivo la realización de una animación sociocultural en el establecimiento carcelario la Modelo ubicado en la ciudad de Bogotá- Colombia, específicamente en la unidad de salud mental, a través de metodologías participativas y alternativas de intervención como el Aprendizaje Servicio Solidario (ASS); de esta manera fue posible identificar en un primer momento algunas de las necesidades sentidas, percibidas e inferidas, tanto de la institución, como de las personas privadas de la libertad (PPL), a través del arte y el deporte. En un segundo momento se desarrolló una intervención a través de las metodologías ya mencionadas, utilizando el deporte como herramienta principal, buscando potencializar en los internos diferentes alternativas para la solución de problemas y la transformación de las necesidades evaluadas. En cuanto a este último objetivo de la intervención no todas las necesidades pudieron tener alguna modificación, puesto que algunas de ellas se encuentran vinculadas con lineamientos institucionales fuera del alcance de esta investigación. Para este estudio no se contó con una muestra estable, ya que para cada sesión los PPL podían elegir su participación de manera voluntaria y en casos particulares se evidenciaban traslados de patio o de establecimiento carcelario y, además, se presentaron situaciones de inseguridad en el patio, que no permitieron el desarrollo adecuado de las últimas sesiones de intervención.140 p.1.     Taenia larvae possess distinct acetylcholinesterase profiles with implications for host cholinergic signalling

    Larvae of the cestodes Taenia solium and Taenia crassiceps infect the central nervous system of humans. Taenia solium larvae in the brain cause neurocysticercosis, the leading cause of adult-acquired epilepsy worldwide. Relatively little is understood about how cestode-derived products modulate host neural and immune signalling. Acetylcholinesterases, a class of enzyme that breaks down acetylcholine, are produced by a host of parasitic worms to aid their survival in the host. Acetylcholine is an important signalling molecule in both the human nervous and immune systems, with powerful modulatory effects on the excitability of cortical networks. Therefore, it is important to establish whether cestode derived acetylcholinesterases may alter host neuronal cholinergic signalling. Here we make use of multiple techniques to profile acetylcholinesterase activity in different extracts of both Taenia crassiceps and Taenia solium larvae. We find that the larvae of both species contain substantial acetylcholinesterase activity. However, acetylcholinesterase activity is lower in Taenia solium as compared to Taenia crassiceps larvae. Further, whilst we observed acetylcholinesterase activity in all fractions of Taenia crassiceps larvae, including on the membrane surface and in the excreted/secreted extracts, we could not identify acetylcholinesterases on the membrane surface or in the excreted/secreted extracts of Taenia solium larvae. Bioinformatic analysis revealed conservation of the functional protein domains in the Taenia solium acetylcholinesterases, when compared to the homologous human sequence. Finally, using whole-cell patch clamp recordings in rat hippocampal brain slice cultures, we demonstrate that Taenia larval derived acetylcholinesterases can break down acetylcholine at a concentration which induces changes in neuronal signalling. Together, these findings highlight the possibility that Taenia larval acetylcholinesterases can interfere with cholinergic signalling in the host, potentially contributing to pathogenesis in neurocysticercosis

    The Astropy Problem

    The Astropy Project (http://astropy.org) is, in its own words, "a community effort to develop a single core package for Astronomy in Python and foster interoperability between Python astronomy packages." For five years this project has been managed, written, and operated as a grassroots, self-organized, almost entirely volunteer effort while the software is used by the majority of the astronomical community. Despite this, the project has always been and remains to this day effectively unfunded. Further, contributors receive little or no formal recognition for creating and supporting what is now critical software. This paper explores the problem in detail, outlines possible solutions to correct this, and presents a few suggestions on how to address the sustainability of general purpose astronomical software

    Self-reported mindfulness and cortisol during a Shamatha meditation retreat.

    Objective: Cognitive perseverations that include worry and rumination over past or future events may prolong cortisol release, which in turn may contribute to predisease pathways and adversely affect physical health. Meditation training may increase self-reported mindfulness, which has been linked to reductions in cognitive perseverations. However, there are no reports that directly link self-reported mindfulness and resting cortisol output. Here, the authors investigate this link. Methods: In an observational study, we measured self-reported mindfulness and p.m. cortisol near the beginning and end of a 3-month meditation retreat (N = 57). Results: Mindfulness increased from pre- to post-retreat, F(1, 56) = 36.20, p < .001. Cortisol did not significantly change. However, mindfulness was inversely related to p.m. cortisol at pre-retreat, r(53) = −.31, p < .05, and post-retreat, r(53) = −.30, p < .05, controlling for age and body mass index. Pre to postchange in mindfulness was associated with pre to postchange in p.m. cortisol, β = −.37, t(49) = 2.30, p < .05: Larger increases in mindfulness were associated with decreases in p.m. cortisol, whereas smaller increases (or slight decreases) in mindfulness were associated with an increase in p.m. cortisol. Conclusions: These data suggest a relation between self-reported mindfulness and resting output of the hypothalamic-pituitary-adrenal system. Future work should aim to replicate this finding in a larger cohort and determine stronger inference about causality by using experimental designs that include control-group conditions
