    Cerebrospinal Fluid Tau Protein Levels and F-18-Fluorodeoxyglucose Positron Emission Tomography in the Differential Diagnosis of Alzheimer's Disease

    Aims: In this study, we aimed to compare cerebrospinal fluid (CSF) levels of total tau (t-tau), phosphorylated tau (p-tau(181)) and positron emission tomography with F-18-fluorodeoxyglucose (FDG-PET) in the differential diagnosis of Alzheimer's disease (AD) under clinical conditions. Method: In a cross-sectional, blinded, single-center study, we examined a sample of 75 unselected memory clinic patients with clinical diagnoses of dementia of Alzheimer type (DAT; n = 24), amnestic mild cognitive impairment (MCI; n = 16), other dementias (n = 13) and nondemented controls (n = 22). Discriminative accuracy, sensitivity and specificity were calculated and compared using ROC analyses. Results: p-tau(181) and FDG-PET were comparable in separating DAT from controls (sensitivity: 67 vs. 79%; specificity: 91% for both) and patients with other dementias (sensitivity: 71 vs. 79%; specificity: 100% for both). The sensitivity of p-tau 181 in differentiating MCI patients from controls was significantly (p < 0.05) superior to that of FDG-PET (75 vs. 44%) at a comparably high specificity (82 vs. 91%); t-tau measures were less accurate in all analyses. Conclusions: FDG-PET and CSF p-tau(181) levels are able to discriminate DAT in heterogeneous and unselected samples with a high accuracy.     Biogeochemical structure of the Laptev Sea in 2015-2020 associated with the River Lena plume

    The discharge of rivers and the subsequent dispersion of their plumes play a pivotal role in the biogeochemical cycling of the Arctic Ocean. Based on the data collected during annual transects conducted in the autumn period (September-October) from 2015-2020, this study explores the effect of River Lena plume dispersion on the seasonal and interannual changes in the hydrophysical and biogeochemical structure of the southeastern Laptev Sea. The temperature-salinity relationship (T-S), Redfield ratio and multiparameter cluster analysis were used to investigate variations in the water mass structure along the transect. The results revealed that the plume’s interannual and seasonal spreading patterns play a crucial role in regulating the local physical, biogeochemical, and biological processes in the southern Laptev Sea. During September-October, the hydrochemical water mass structure along the transects shifted from highly stratified to unstratified as the plume’s mixing intensity increased. Anomalous hydrochemical distributions were observed due to coastal upwelling, which was primarily characterized by high total alkalinity and nitrate levels, and low organic phosphorus, nitrite, and ammonia levels in the seawater. Wind and cold weather conditions drive deep vertical mixing of seawater, causing the resuspension of bottom sediment and the subsequent enrichment of bottom water by nutrients. Multi-parameter cluster analysis is used to describe the details of water mass structures in the highly dynamic southern Laptev Sea, with water mass structures typically undergoing significant changes within two weeks between September and October. The migration and transformation of water masses throughout the seasons are influenced by the volume of river discharge, fall-winter cooling, and atmospheric circulation patterns. Furthermore, the general atmospheric circulation is confirmed to be the primary cause of the interannual variation in the spread of the Lena River plume over the southeast Laptev Sea

    Cognitive reserve impacts on inter-individual variability in resting-state cerebral metabolism in normal aging

    peer reviewedThere is a great deal of heterogeneity in the impact of aging on cognition and cerebral functioning. One potential factor contributing to individual differences among the elders is the cognitive reserve, which designates the partial protection from the deleterious effects of aging that lifetime experience provides. Neuroimaging studies examining task-related activation in elderly people suggested that cognitive reserve takes the form of more efficient use of brain networks and/or greater ability to recruit alternative networks to compensate for age-related cerebral changes. In this multi-centre study, we examined the relationships between cognitive reserve, as measured by education and verbal intelligence, and cerebral metabolism at rest (FDG-PET) in a sample of 74 healthy older participants. Higher degree of education and verbal intelligence was associated with less metabolic activity in the right posterior temporoparietal cortex and the left anterior intraparietal sulcus. Functional connectivity analyses of resting-state fMRI images in a subset of 41 participants indicated that these regions belong to the default mode network and the dorsal attention network respectively. Lower metabolism in the temporoparietal cortex was also associated with better memory abilities. The findings provide evidence for an inverse relationship between cognitive reserve and resting-state activity in key regions of two functional networks respectively involved in internal mentation and goal-directed attention

    Direct comparison of activation maps during galvanic vestibular stimulation: A hybrid H-2[(15) O] PET-BOLD MRI activation study

    Previous unimodal PET and fMRI studies in humans revealed a reproducible vestibular brain activation pattern, but with variations in its weighting and expansiveness. Hybrid studies minimizing methodological variations at baseline conditions are rare and still lacking for task-based designs. Thus, we applied for the first time hybrid 3T PET-MRI scanning (Siemens mMR) in healthy volunteers using galvanic vestibular stimulation (GVS) in healthy volunteers in order to directly compare (H2O)-O-15-PET and BOLD MRI responses. List mode PET acquisition started with the injection of 750 MBq (H2O)-O-15 simultaneously to MRI EPI sequences. Group-level statistical parametric maps were generated for GVS vs. rest contrasts of PET, MR-onset (event-related), and MR-block. All contrasts showed a similar bilateral vestibular activation pattern with remarkable proximity of activation foci. Both BOLD contrasts gave more bilateral wide-spread activation clusters than PET;no area showed contradictory signal responses. PET still confirmed the right-hemispheric lateralization of the vestibular system, whereas BOLD-onset revealed only a tendency. The reciprocal inhibitory visual-vestibular interaction concept was confirmed by PET signal decreases in primary and secondary visual cortices, and BOLD-block decreases in secondary visual areas. In conclusion, MRI activation maps contained a mixture of CBF measured using (H2O)-O-15-PET and additional non-CBF effects, and the activation-deactivation pattern of the BOLD-block appears to be more similar to the (H2O)-O-15-PET than the BOLD-onset

    Similarity between structural and proxy estimates of brain connectivity.

    peer reviewedFunctional magnetic resonance and diffusion weighted imaging have so far made a major contribution to delineation of the brain connectome at the macroscale. While functional connectivity (FC) was shown to be related to structural connectivity (SC) to a certain degree, their spatial overlap is unknown. Even less clear are relations of SC with estimates of connectivity from inter-subject covariance of regional F18-fluorodeoxyglucose uptake (FDGcov) and grey matter volume (GMVcov). Here, we asked to what extent SC underlies three proxy estimates of brain connectivity: FC, FDGcov and GMVcov. Simultaneous PET/MR acquisitions were performed in 56 healthy middle-aged individuals. Similarity between four networks was assessed using Spearman correlation and convergence ratio (CR), a measure of spatial overlap. Spearman correlation coefficient was 0.27 for SC-FC, 0.40 for SC-FDGcov, and 0.15 for SC-GMVcov. Mean CRs were 51% for SC-FC, 48% for SC-FDGcov, and 37% for SC-GMVcov. These results proved to be reproducible and robust against image processing steps. In sum, we found a relevant similarity of SC with FC and FDGcov, while GMVcov consistently showed the weakest similarity. These findings indicate that white matter tracts underlie FDGcov to a similar degree as FC, supporting FDGcov as estimate of functional brain connectivity

    Mercury in the Barents region – River fluxes, sources, and environmental concentrations

    Arctic rivers are receiving increased attention for their contributing of mercury (Hg) to the Arctic Ocean. Despite this, the knowledge on both the terrestrial release sources and the levels of Hg in the rivers are limited. Within the Arctic, the Barents region has a high industrial development, including multiple potential Hg release sources. This study presents the first overview of potential Hg release sources on Norwegian and Russian mainland draining to the Barents Sea. Source categories cover mining and metallurgy industry; historical pulp and paper production; municipal and industrial solid waste handling; fossil fuel combustion; and past military activities. Available data on Hg in freshwater bodies near the identified potential release sources are reviewed. Levels of Hg were occasionally exceeding the national pollution control limits, thereby posing concern to the local human population and wildlife. However, the studies were sparse and often unsystematic. Finally, we present new data of Hg measured in five Barents rivers. These data reveal strong seasonality in the Hg levels, with a total annual flux constituting 2% of the panarctic total. With this new insight we aspire to contribute to the international efforts of reducing Hg pollution, such as through the effective implementation of the Minamata Convention. Future studies documenting Hg in exposed Barents freshwater bodies are warranted.publishedVersio
