    Modifications of Hyaluronan Influence the Interaction with Human Bone Morphogenetic Protein-4 (hBMP-4).

    n this study, we have demonstrated that the modification of hyaluronan (hyaluronic acid; Hya) with sulfate groups led to different binding affinities for recombinant human bone morphogenetic protein-4 (rhBMP-4). The high-sulfated sHya2.8 (average degree of sulfation (D.S.) 2.8) exhibited the tightest interaction with rhBMP-4, followed by the low-sulfated sHya1.0, as determined with surface plasmon resonance (SPR), ELISA, and competition ELISA. Unmodified Hya, chondroitin-sulfate (CS), and heparan sulfate (HS) showed significantly less binding affinity. SPR data could be fitted to an A + B = AB Langmuir model and binding constants were evaluated ranging from 13 pM to 5.45 microM. The interaction characteristics of the differentially sulfated Hyas are promising for the incorporation of these modified polysaccharides in bioengineered coatings of biomaterials for medical applications

    A probabilistic evolutionary optimization approach to compute quasiparticle braids

    Topological quantum computing is an alternative framework for avoiding the quantum decoherence problem in quantum computation. The problem of executing a gate in this framework can be posed as the problem of braiding quasiparticles. Because these are not Abelian, the problem can be reduced to finding an optimal product of braid generators where the optimality is defined in terms of the gate approximation and the braid's length. In this paper we propose the use of different variants of estimation of distribution algorithms to deal with the problem. Furthermore, we investigate how the regularities of the braid optimization problem can be translated into statistical regularities by means of the Boltzmann distribution. We show that our best algorithm is able to produce many solutions that approximates the target gate with an accuracy in the order of 10610^{-6}, and have lengths up to 9 times shorter than those expected from braids of the same accuracy obtained with other methods.Comment: 9 pages,7 figures.     Left ventricular assist device implantation augments nitric oxide dependent control of mitochondrial respiration in failing human hearts

    AbstractOBJECTIVESThe objective of the study was to evaluate nitric oxide (NO) mediated regulation of mitochondrial respiration after implantation of a mechanical assist device in end-stage heart failure.BACKGROUNDVentricular unloading using a left ventricular assist device (LVAD) can improve mitochondrial function in end-stage heart failure. Nitric oxide modulates the activity of the mitochondrial electron transport chain to regulate myocardial oxygen consumption (MVO2).METHODSMyocardial oxygen consumption was measured polarographically using a Clark-type oxygen electrode in isolated left ventricular myocardium from 26 explanted failing human hearts obtained at the time of heart transplantation.RESULTSThe rate of decrease in oxygen concentration was expressed as a percentage of baseline. Results of the highest dose of drug are shown. Decrease in MVO2 was greater in LVAD hearts (n = 8) compared with heart failure controls (n = 18) in response to the following drugs: bradykinin (−34 ± 3% vs. −24 ± 5%), enalaprilat (−37 ± 5% vs. −23 ± 5%) and amlodipine (−43 ± 13% vs. −16 ± 5%; p < 0.05 from controls). The decrease in MVO2 in LVAD hearts was not significantly different from controls in response to diltiazem (−22 ± 5% in both groups) and exogenous NO donor, nitroglycerin (−33 ± 7% vs. −30 ± 3%). Nw-nitro-L-arginine methyl ester, inhibitor of NO synthase, attenuated the response to bradykinin, enalaprilat and amlodipine. Reductions in MVO2 in response to diltiazem and nitroglycerin were not altered by inhibiting NO.CONCLUSIONSChronic LVAD support potentiates endogenous NO-mediated regulation of mitochondrial respiration. Use of medical or surgical interventions that augment NO bioavailability may promote myocardial recovery in end-stage heart failure

    The evolution of representation in simple cognitive networks

    Representations are internal models of the environment that can provide guidance to a behaving agent, even in the absence of sensory information. It is not clear how representations are developed and whether or not they are necessary or even essential for intelligent behavior. We argue here that the ability to represent relevant features of the environment is the expected consequence of an adaptive process, give a formal definition of representation based on information theory, and quantify it with a measure R. To measure how R changes over time, we evolve two types of networks---an artificial neural network and a network of hidden Markov gates---to solve a categorization task using a genetic algorithm. We find that the capacity to represent increases during evolutionary adaptation, and that agents form representations of their environment during their lifetime. This ability allows the agents to act on sensorial inputs in the context of their acquired representations and enables complex and context-dependent behavior. We examine which concepts (features of the environment) our networks are representing, how the representations are logically encoded in the networks, and how they form as an agent behaves to solve a task. We conclude that R should be able to quantify the representations within any cognitive system, and should be predictive of an agent's long-term adaptive success.Comment: 36 pages, 10 figures, one Tabl

    An in vitro comparison between two methods of electrical resistance measurement for occlusal caries detection

    Because of different measurement techniques and the easier design of the CRM prototype, this in vitro study aimed to compare the diagnostic performance and reproducibility of two electrical methods (Electronic Caries Monitor III, ECM and Cariometer 800, CRM) for occlusal caries detection, and to evaluate the effect of staining/ discoloration of fissures on diagnostic performance. Hundred and seventeen third molars with no apparent occlusal cavitation were selected. Six examiners inspected all specimens independently, using the CRM, and a subgroup of 4 using the ECM. Histological validation using a stereomicroscope was performed after hemisectioning. Intra- and interexaminer reproducibility was assessed by Lin's concordance correlation coefficient (CCC) and Bland and Altman analysis. Diagnostic performance parameters included sensitivity (SE), specificity (SP) and area under the ROC curve (A(z)). The CCC yielded an intra- and interexaminer reproducibility of 0.69/0.62 (ECM) and of 0.79/0.74 (CRM). The mean intra- and interexaminer 95% range of measurements (range between Bland and Altman limits of agreement) given in percentages of the instrument reading were 67%/65% for the ECM and 28%/33% for the CRM. A(z) at the D3-4 level was 0.74 (ECM) and 0.78 (CRM). The CRM showed at least equivalent diagnostic performance to the ECM. However, improvement is still desirable. Diagnostic performance appeared to be enhanced in discolored lesions; however, this may be related to sample lesion distribution characteristics. Copyright (C) 2006 S. Karger AG, Basel

    AVGS, AR and D for Satellites, ISS, the Moon, Mars and Beyond

    With the continuous need to rotate crew and re-supply the International Space Station (ISS) and the desire to return humans to the Moon and for the first time, place humans on Mars, NASA must develop a more robust and highly reliable capability to perform Autonomous Rendezvous and Capture (AR&C) because, unlike the Apollo missions, NASA plans to send the entire crew to the Lunar or Martian surface and must be able to dock with the Orion spacecraft upon return. In 1997, NASA developed the Video Guidance Sensor (VGS) which was flown and tested on STS-87 and STS-95. In 2001, NASA designed and built a more enhanced version of the VGS, called the Advanced Video Guidance Sensor (AVGS). The AVGS offered significant technology improvements to the precursor VGS design. This paper will describe the AVGS as it was in the DART mission of 2005 and the Orbital Express mission of 2007. The paper will describe the capabilities and design concepts of the AVGS as it was flown on the DART 2005 Mission and the DARPA Orbital Express Mission slated to fly in 2007. The paper will cover the Flight Software, problems encountered, testing for Orbital Express and where NASA is going in the future
