    The relationship between the educational background and managerial experience of senior Navy Medical Service Corps executives (Health Care Administrators) and their perceived current and required management capabilities

    The purpose of this thesis is to examine the reported educational attainment and management experience of senior Navy medical department executives in an attempt to isolate those variables that affect their perceived management capabilities. The data used for this thesis were taken from the results of a joint Bureau of Medicine and Surgery (BUMED) and Naval Postgraduate School (NPS) survey. which was designed to asses the unique educational needs of Navy medical department executives. This thesis explores one of the findings of the survey which was that the Medical Service Corps Health Care Administrator (HCA) cohort perceived their current skills to be higher than the skill levels required for a majority of the management skill categories listed in the survey. This thesis isolates certain management education and experience variables to determine why this group is so confident in their skill levels. The findings indicate that the HCA cohort has more management education and experience than the other medical communities. Additionally, management experience had more of an impact than management education on the HCA responses.http://archive.org/details/therelationshipb109453086

    Protocol: High-throughput and quantitative assays of auxin and auxin precursors from minute tissue samples

    Background: The plant hormone auxin, indole-3-acetic acid (IAA), plays important roles in plant growth and development. The signaling response to IAA is largely dependent on the local concentration of IAA, and this concentration is regulated by multiple mechanisms in plants. Therefore, the precise quantification of local IAA concentration provides insights into the regulation of IAA and its biological roles. Meanwhile, pathways and genes involved in IAA biosynthesis are not fully understood, so it is necessary to analyze the production of IAA at the metabolite level for unbiased studies of IAA biosynthesis. Results: We have developed high-throughput methods to quantify plant endogenous IAA and its biosynthetic precursors including indole, tryptophan, indole-3-pyruvic acid (IPyA), and indole-3-butyric acid (IBA). The protocol starts with homogenizing plant tissues with stable-labeled internal standards added, followed by analyte purification using solid phase extraction (SPE) tips and analyte derivatization. The derivatized analytes are finally analyzed by selected reaction monitoring on a gas chromatograph-mass spectrometer (GC-MS/MS) to determine the precise abundance of analytes. The amount of plant tissue required for the assay is small (typically 2–10 mg fresh weight), and the use of SPE tips is simple and convenient, which allows preparation of large sets of samples within reasonable time periods. Conclusions: The SPE tips and GC-MS/MS based method enables high-throughput and accurate quantification of IAA and its biosynthetic precursors from minute plant tissue samples. The protocol can be used for measurement of these endogenous compounds using isotope dilution, and it can also be applied to analyze IAA biosynthesis and biosynthetic pathways using stable isotope labeling. The method will potentially advance knowledge of the role and regulation of IAA

    Determining subpopulation methylation profiles from bisulfite sequencing data of heterogeneous samples using DXM

    Epigenetic changes, such as aberrant DNA methylation, contribute to cancer clonal expansion and disease progression. However, identifying subpopulation-level changes in a heterogeneous sample remains challenging. Thus, we have developed a computational approach, DXM, to deconvolve the methylation profiles of major allelic subpopulations from the bisulfite sequencing data of a heterogeneous sample. DXM does not require prior knowledge of the number of subpopulations or types of cells to expect. We benchmark DXM\u27s performance and demonstrate improvement over existing methods. We further experimentally validate DXM predicted allelic subpopulation-methylation profiles in four Diffuse Large B-Cell Lymphomas (DLBCLs). Lastly, as proof-of-concept, we apply DXM to a cohort of 31 DLBCLs and relate allelic subpopulation methylation profiles to relapse. We thus demonstrate that DXM can robustly find allelic subpopulation methylation profiles that may contribute to disease progression using bisulfite sequencing data of any heterogeneous sample

    Applications for Gradient Metal Alloys Fabricated Using Additive Manufacturing

    Recently, additive manufacturing (AM) techniques have been developed that may shift the paradigm of traditional metal production by allowing complex net-shaped hardware to be built up layer-by-layer, rather than being machined from a billet. The AM process is ubiquitous with polymers due to their low melting temperatures, fast curing, and controllable viscosity, and 3D printers are widely available as commercial or consumer products. 3D printing with metals is inherently more complicated than with polymers due to their higher melting temperatures and reactivity with air, particularly when heated or molten. The process generally requires a high-power laser or other focused heat source, like an electron beam, for precise melting and deposition. Several promising metal AM techniques have been developed, including laser deposition (also called laser engineered net shaping or LENS and laser deposition technology (LDT)), direct metal laser sintering (DMLS), and electron beam free-form (EBF). These machines typically use powders or wire feedstock that are melted and deposited using a laser or electron beam. Complex net-shape parts have been widely demonstrated using these (and other) AM techniques and the process appears to be a promising alternative to machining in some cases. Rather than simply competing with traditional machining for cost and time savings, the true advantage of AM involves the fabrication of hardware that cannot be produced using other techniques. This could include parts with "blind" features (like foams or trusses), parts that are difficult to machine conventionally, or parts made from materials that do not exist in bulk forms. In this work, the inventors identify that several AM techniques can be used to develop metal parts that change composition from one location in the part to another, allowing for complete control over the mechanical or physical properties. This changes the paradigm for conventional metal fabrication, which relies on an assortment of "post-processing" methods to locally alter properties (such as coating, heat treating, work hardening, shot peening, etching, anodizing, among others). Building the final part in an additive process allows for the development of an entirely new class of metals, so-called "functionally graded metals" or "gradient alloys." By carefully blending feedstock materials with different properties in an AM process, hardware can be developed with properties that cannot be obtained using other techniques but with the added benefit of the net-shaped fabrication that AM allows

    COSEE OCEAN Inquiry Group Report: Opportunities for Creating Lifelong Ocean Science Literacy

    This Inquiry Group Report for the Centers for Ocean Sciences Education Excellence Ocean Communities in Education And Social Networks (COSEE OCEAN) provides a fresh look at how broader ocean science literacy can be developed, especially through less-recognized channels such as opportunistic learning, the private and “third” sectors, and the enormously varied activities under the heading of informal science education. The 10 authors of this report (see Contributors section) have been working together for two years to find and review a range of issues and resources for current and potential ocean science literacy providers, both professional and volunteer. Several chapters provide a survey of useful materials and websites, while others offer ideas that will be new for many readers, including opportunities and unfamiliar venues for carrying out this enterprise. Included are chapters depicting a perhaps surprising plethora of channels for increasing ocean science literacy, both in school and especially, out of school. Other chapters discuss the societal contexts for using these channels, which include ideas for potential funding sources and cultural partners. Not every reader will find every chapter useful so each chapter is written to stand alone. This requires redundancy of some material from chapter to chapter. The primary authors for each chapter have their own writing styles and voice. Finally, given the varied topics of each chapter, some are dense with annotated lists of existing materials, while others were written to analyze fresh opportunities for the creation of new materials and projects. This report was written with several audiences in mind: the professional educators in the Centers for Ocean Sciences Education Excellence (COSEE) network; formal and informal educators who are looking for ways to incorporate ocean sciences in the work they do; and policymakers in both science and education who are concerned with using all available channels to improve the public understanding of such major concerns as global climate change, energy sources and uses, the health of local and global ecosystems, and natural resources such as seafood

    Time esophageal pH < 4 overestimates the prevalence of pathologic esophageal reflux in subjects with gastroesophageal reflux disease treated with proton pump inhibitors

    <p>Abstract</p> <p>Background</p> <p>A Stanford University study reported that in asymptomatic GERD patients who were being treated with a proton pump inhibitor (PPI), 50% had pathologic esophageal acid exposure.</p> <p>Aim</p> <p>We considered the possibility that the high prevalence of pathologic esophageal reflux might simply have resulted from calculating acidity as time pH < 4.</p> <p>Methods</p> <p>We calculated integrated acidity and time pH < 4 from the 49 recordings of 24-hour gastric and esophageal pH from the Stanford study as well as from another study of 57 GERD subjects, 26 of whom were treated for 8 days with 20 mg omeprazole or 20 mg rabeprazole in a 2-way crossover fashion.</p> <p>Results</p> <p>The prevalence of pathologic 24-hour esophageal reflux in both studies was significantly higher when measured as time pH < 4 than when measured as integrated acidity. This difference was entirely attributable to a difference between the two measures during the nocturnal period. Nocturnal gastric acid breakthrough was not a useful predictor of pathologic nocturnal esophageal reflux.</p> <p>Conclusion</p> <p>In GERD subjects treated with a PPI, measuring time esophageal pH < 4 will significantly overestimate the prevalence of pathologic esophageal acid exposure over 24 hours and during the nocturnal period.</p
