    Limnology for the ornithologist: effects of Lake Maggiore water level on migratory flows

    Wetlands are widely recognised as the most critical stop-over locations along migration flyways. Wetland ecology is mostly influenced by water levels and water regimes. This research focuses on Bolle di Magadino (Switzerland), an important stop-over site on Lake Maggiore, artificially regulated by a dam. In this work we examined how the artificial flooding of a wetland affects the use of this stop-over site by migrating passerines during spring. Bird presence in the area was evaluated using both data collected at the ringing station located in the wetlands and the bird traffic rate (BTR) supplied by the BirdScan MR1, an avian vertical-looking radar (VLR) capable of automatically detecting and classifying birds in flight. In an attempt to shed light on the effect of lake level on stop- over quality, we i) simulated with GIS the extent of the flooded area and of the different habitat categories as the lake level changes; ii) calculated the relationship between lake level and the ability of stop-overing birds to acquire trophic resources; iii) verified that the flux of passerines below 500 m above ground level measured by radar could be used as a proxy for the number of stop-overing birds; iv) calculated the relationship between the number of birds leaving the stop-over and the lake level. While the number of ringed passerines has proven to be representative of the migratory flow below 500 meters of altitude at the site of interest, a high lake level seems to have a negative impact on the use by some species of the Bolle di Magadino area as a stop-over site during spring. In particular, two of the target species -the blackcap and the reed bunting- have proven to be sensitive to higher water levels. While taking into account the limitations and the relative nature of the results, could be necessary for the competent authorities to take these results into consideration in order to safeguard the Bolle di Magadino's role as an important stop- over area during spring

    Retarded germination of Nicotiana tabacum seeds following insertion of exogenous DNA mimics the seed persistent behavior

    Tobacco seeds show a coat-imposed dormancy in which the seed envelope tissues (testa and endosperm) impose a physical constraint on the radicle protrusion. The germination-limiting process is represented by the endosperm rupture which is induced by cell-wall weakening. Transgenic tobacco seeds, obtained by insertion of exogenous genes codifying for seed-based oral vaccines (F18 and VT2eB), showed retarded germination with respect to the wild type and modified the expression of endogenous proteins. Morphological and proteomic analyses of wild type and transgenic seeds revealed new insights into factors influencing seed germination. Our data showed that the interference of exogenous DNA influences the germination rather than the dormancy release, by modifying the maturation process. Dry seeds of F18 and VT2eB transgenic lines accumulated a higher amount of reserve and stressâ\u80\u93related proteins with respect to the wild type. Moreover, the storage proteins accumulated in tobacco F18 and VT2eB dry seeds have structural properties that do not enable the early limited proteolysis observed in the wild type. Morphological observations by electron and light microscopy revealed a retarded mobilization of the storage material from protein and lipid bodies in transgenic seeds, thus impairing water imbibition and embryo elongation. In addition, both F18 and VT2eB dry seeds are more rounded than the wild type. Both the morphological and biochemical characteristics of transgenic seeds mimic the seed persistent profile, in which their roundness enables them to be buried in the soil, while the higher content of storage material enables the hypocotyl to elongate more and the cotyledons to emerge

    The spreading of the invasive sacred ibis in Italy

    The spreading of invasive species in new continents can vary from slow and limited diffusion to fast colonisations over vast new areas. We studied the sacred ibis Threskiornis aethiopicus along a 31-year period, from 1989 to 2019, with particular attention to the first area of release in NW Italy. We collected data on species distribution through observations by citizen science projects, population density by transects with distance method, breeding censuses at colonies, and post breeding censuses at roosts. The birds counted at winter roosts in NW Italy increased from a few tens up to 10,880 individuals in 2019. Sacred ibises started breeding in 1989, with a single nest in north-western Italy. The number of breeders remained very low until 2006, when both overwintering and breeding sacred ibises started to increase exponentially and expand their range throughout northern Italy with isolated breeding cases in central Italy. In 2019, the number of nests had increased to 1249 nests in 31 colonies. In NW Italy, the density of foraging birds averaged 3.9 ind./km2 in winter and 1.5 ind./km2 in the breeding period, with a mean size of the foraging groups of 8.9 and 2.1 birds respectively. Direct field observations and species distribution models (SDM) showed that foraging habitats were mainly rice fields and wetlands. A SDM applied to the whole Italian peninsula plus Sardinia and Sicily showed that the variables best related to the SDM were land class (rice fields and wetlands), altitude, and the temperature seasonality. The areas favourable for species expansion encompass all the plains of Northern Italy, and several areas of Tuscany, Latium, Sardinia, and Apulia

    Drugs targeting the mitochondrial pore act as citotoxic and cytostatic agents in temozolomide-resistant glioma cells

    <p>Abstract</p> <p>Background</p> <p>High grade gliomas are one of the most difficult cancers to treat and despite surgery, radiotherapy and temozolomide-based chemotherapy, the prognosis of glioma patients is poor. Resistance to temozolomide is the major barrier to effective therapy. Alternative therapeutic approaches have been shown to be ineffective for the treatment of genetically unselected glioma patients. Thus, novel therapies are needed. Mitochondria-directed chemotherapy is an emerging tool to combat cancer, and inner mitochondrial permeability transition (MPT) represents a target for the development of cytotoxic drugs. A number of agents are able to induce MPT and some of them target MPT-pore (MPTP) components that are selectively up-regulated in cancer, making these agents putative cancer cell-specific drugs.</p> <p>Objective</p> <p>The aim of this paper is to report a comprehensive analysis of the effects produced by selected MPT-inducing drugs (Betulinic Acid, Lonidamine, CD437) in a temozolomide-resistant glioblastoma cell line (ADF cells).</p> <p>Methods</p> <p>EGFRvIII expression has been assayed by RT-PCR. EGFR amplification and PTEN deletion have been assayed by differential-PCR. Drugs effect on cell viability has been tested by crystal violet assay. MPT has been tested by JC1 staining. Drug cytostatic effect has been tested by mitotic index analysis. Drug cytotoxic effect has been tested by calcein AM staining. Apoptosis has been assayed by Hoechst incorporation and Annexine V binding assay. Authophagy has been tested by acridine orange staining.</p> <p>Results</p> <p>We performed a molecular and genetic characterization of ADF cells and demonstrated that this line does not express the EGFRvIII and does not show EGFR amplification. ADF cells do not show PTEN mutation but differential PCR data indicate a hemizygous deletion of PTEN gene. We analyzed the response of ADF cells to Betulinic Acid, Lonidamine, and CD437. Our data demonstrate that MPT-inducing agents produce concentration-dependent cytostatic and cytotoxic effects in parallel with MPT induction triggered through MPTP. CD437, Lonidamine and Betulinic acid trigger apoptosis as principal death modality.</p> <p>Conclusion</p> <p>The obtained data suggest that these pharmacological agents could be selected as adjuvant drugs for the treatment of high grade astrocytomas that resist conventional therapies or that do not show any peculiar genetic alteration that can be targeted by specific drugs.</p

    Novel laser-based techniques for monitoring of volcanoes

Their main advantages and drawbacks are discussed focusing on the achievable sensitivity and precision levels in analysis of gaseous species. Some of the most recent experimental results obtained in laboratory development as well as in field tests of home-built laser spectrometers are reported. New perspectives in optical devices aimed at geochemical and geophysical applications are also considered
