    Determination of polyamines in Arabidopsis thaliana by capillary electrophoresis using salicylaldehyde-5-sulfonate as a derivatizing reagent

    Herein, we report a novel method for the determination of polyamines in a sample extracted from Arabidopsis thaliana by capillary electrophoresis (CE) using salicylaldehyde-5-sulfonate (SAS) as a derivatizing reagent. An aldehyde group of SAS forms a Schiff base with amino groups of aliphatic polyamines, resulting in an anionic species with an absorption band in the ultraviolet region. The derivatization method was straightforward since the derivatives were formed by mixing a sample with the derivatizing reagent at a neutral pH. In addition, the negative charges induced by SAS led to a high resolution with a short analysis time. This method permitted the separation of five polyamines, which play important roles in plants. However, further improvement in sensitivity was needed for the determination of the polyamines in plant samples. Therefore, the CE method was coupled with solid-phase extraction (SPE) using an ion-pairing formation with sodium dodecyl benzene sulfonate. The SPE method improved the concentration limits of detection to sub-μM levels, which corresponded with a 10-fold enhancement. The calibration curves for cadaverine, putrescine, and spermidine were linear with concentrations that ranged from 1 to 20 μM and correlation coefficients (R2) were greater than 0.998.     Genetic dissection of behavioral traits related to successful training of drug detection dogs

    Abstract Drug detection dogs play integral roles in society. However, the interplay between their behaviors and genetic characteristics underlying their performance remains uninvestigated. Herein, more than 120,000 genetic variants were evaluated in 326 German Shepherd or Labrador Retriever dogs to profile the genetic traits associated with various behavioral traits related to the successful training of drug detection dogs. Behavioral breed differences were observed in ‘friendliness to humans’ and ‘tolerance to dogs.’ A genome-wide association study within both breeds identified 11 regions potentially associated with drug detection dog characteristics as well as ‘interest in the target’ and ‘friendliness to humans,’ which are related to drug detection abilities. Among them, 63 protein-coding genes, including Atat1 and Pfn2 known to be associated with anxiety-related or exploration behavior in mice, respectively, were located surrounding the identified candidate polymorphisms. This study highlights genetic characteristics associated with behavioral traits that are important for the successful training of drug detection dogs. Thus, these findings may facilitate improved breeding and training of these dogs

    Classifying Water Quality of Wando and Tamari According to the Characterization of H2O Stable Isotope and Ionized Type in the Middle and Lower Kiso River*

    Wando and tamari are water bodies in the floodplain of a river and play an important role in maintaining valuable ecosystems. There are over 100 wando and tamari in the middle and lower basin of the Kiso River. An Acheilognathus longipinnis, Itasenpara Bitterling, which is designated as an endangered species, has been identified in these areas. The habitat of these valuable wando and tamari species is endangered by the development of large land plants around these reservoirs caused by river dredging for flood protection. Therefore, it is essential to collect detailed data about the habitat, hydraulic water flow, physical structures and landscape in order to preserve this species, but so far little attention has been given to water quality. The purpose of our research was to classify the water quality of wando and tamari water, especially ions, and to determine the water type based on its origin. Samples were collected at 10 wando sites and 15 tamari sites from August 2011 to February 2013 along the Kiso River. As our analysis by trilinear diagram shows, the waters of wando and tamari almost all belong to the category of Type I (Ca-HCO3), which forms in shallow aquifers. The two reservoirs that do not belong to Type I are presumably contaminated by drainage water from human activities
