The following specific topics were addressed during the reporting period: a method for stabilizing the subsatellite against the rotational effects of atmospheric perturbation was developed; a variety of analytic studies of tether dynamics aimed at elucidating dynamic noise processes were performed; a novel mechanism for coupling longitudinal and latitudinal oscillations of the tether was discovered, and random vibration analysis for modeling the tethered subsatellite under atmospheric perturbation were studied

    Measurement of the gradient of the gravitational acceleration from a satellite platform is likely to provide the next improvement in knowledge of the Earth's gravity field after the upcoming Geopotential Research Mission. Observations from the subsatellite of a tethered satellite system (TSS) would increase sensitivity and resolution due to the low altitude possible. However, the TSS is a dynamically noisy system and would be perturbed by atmospheric drag fluctuations. The dynamic noise is being modeled in order to evaluate the feasibility of TSS gradiometry and to design methods of abating the error caused by this noise. The demonstration flights of the TSS are to provide an opportunity to directly observe the dynamical environment and refine modeling techniques.     The deployment maneuver of three axis vertical constellations with elastic tethers is analyzed. The deployment strategy devised previously was improved. Dampers were added to the system. Effective algorithms for damping out the fundamental vibrational modes of the system were implemented. Simulations of a complete deployment and a subsequent station keeping phase of a three mass constellation is shown

    The g-tuning maneuvers of a 3-mass, vertical tethered system are considered. In particular, the case of reaching a zero-g acceleration level on board the middle mass from a non-zero initial condition is analyzed. A control law that provides a satisfactory transient response is derived. The constellation dynamics in the case of the middle mass travelling from one tether tip to the other is also investigated. Instabilities that take place at the end of the maneuver are analyzed and accommodated by devising suitable damping algorithms

    Nicotine Acts on Growth Plate Chondrocytes to Delay Skeletal Growth through the α7 Neuronal Nicotinic Acetylcholine Receptor

    BACKGROUND: Cigarette smoking adversely affects endochondral ossification during the course of skeletal growth. Among a plethora of cigarette chemicals, nicotine is one of the primary candidate compounds responsible for the cause of smoking-induced delayed skeletal growth. However, the possible mechanism of delayed skeletal growth caused by nicotine remains unclarified. In the last decade, localization of neuronal nicotinic acetylcholine receptor (nAChR), a specific receptor of nicotine, has been widely detected in non-excitable cells. Therefore, we hypothesized that nicotine affect growth plate chondrocytes directly and specifically through nAChR to delay skeletal growth. METHODOLOGY/PRINCIPAL FINDINGS: We investigated the effect of nicotine on human growth plate chondrocytes, a major component of endochondral ossification. The chondrocytes were derived from extra human fingers. Nicotine inhibited matrix synthesis and hypertrophic differentiation in human growth plate chondrocytes in suspension culture in a concentration-dependent manner. Both human and murine growth plate chondrocytes expressed alpha7 nAChR, which constitutes functional homopentameric receptors. Methyllycaconitine (MLA), a specific antagonist of alpha7 nAChR, reversed the inhibition of matrix synthesis and functional calcium signal by nicotine in human growth plate chondrocytes in vitro. To study the effect of nicotine on growth plate in vivo, ovulation-controlled pregnant alpha7 nAChR +/- mice were given drinking water with or without nicotine during pregnancy, and skeletal growth of their fetuses was observed. Maternal nicotine exposure resulted in delayed skeletal growth of alpha7 nAChR +/+ fetuses but not in alpha7 nAChR -/- fetuses, implying that skeletal growth retardation by nicotine is specifically mediated via fetal alpha7 nAChR. CONCLUSIONS/SIGNIFICANCE: These results suggest that nicotine, from cigarette smoking, acts directly on growth plate chondrocytes to decrease matrix synthesis, suppress hypertrophic differentiation via alpha7 nAChR, leading to delayed skeletal growth

    Adverse effects of the antimalaria drug, mefloquine: due to primary liver damage with secondary thyroid involvement?

    BACKGROUND: Mefloquine is a clinically important antimalaria drug, which is often not well tolerated. We critically reviewed 516 published case reports of mefloquine adverse effects, to clarify the phenomenology of the harms associated with mefloquine, and to make recommendations for safer prescribing. PRESENTATION: We postulate that many of the adverse effects of mefloquine are a post-hepatic syndrome caused by primary liver damage. In some users we believe that symptomatic thyroid disturbance occurs, either independently or as a secondary consequence of the hepatocellular injury. The mefloquine syndrome presents in a variety of ways including headache, gastrointestinal disturbances, nervousness, fatigue, disorders of sleep, mood, memory and concentration, and occasionally frank psychosis. Previous liver or thyroid disease, and concurrent insults to the liver (such as from alcohol, dehydration, an oral contraceptive pill, recreational drugs, and other liver-damaging drugs) may be related to the development of severe or prolonged adverse reactions to mefloquine. IMPLICATIONS: We believe that people with active liver or thyroid disease should not take mefloquine, whereas those with fully resolved neuropsychiatric illness may do so safely. Mefloquine users should avoid alcohol, recreational drugs, hormonal contraception and co-medications known to cause liver damage or thyroid damage. With these caveats, we believe that mefloquine may be safely prescribed in pregnancy, and also to occupational groups who carry out safety-critical tasks. TESTING: Mefloquine's adverse effects need to be investigated through a multicentre cohort study, with small controlled studies testing specific elements of the hypothesis

    Smoking and primary total hip or knee replacement due to osteoarthritis in 54,288 elderly men and women

    BACKGROUND The reported association of smoking with risk of undergoing a total joint replacement (TJR) due to osteoarthritis (OA) is not consistent. We evaluated the independent association between smoking and primary TJR in a large cohort. METHODS The electronic records of 54,288 men and women, who were initially recruited for the Second Australian National Blood Pressure study, were linked to the Australian Orthopaedic Association National Joint Replacement Registry to detect total hip replacement (THR) or total knee replacement (TKR) due to osteoarthritis. Competing risk regressions that accounted for the competing risk of death estimated the subhazard ratios for TJR. One-way and probabilistic sensitivity analyses were undertaken to represent uncertainty in the classification of smoking exposure and socioeconomic disadvantage scores. RESULTS An independent inverse association was found between smoking and risk of THR and TKR observed in both men and women. Compared to non-smokers, male and female smokers were respectively 40% and 30% less likely to undergo a TJR. This significant association persisted after controlling for age, co-morbidities, body mass index (BMI), physical exercise, and socioeconomic disadvantage. The overweight and obese were significantly more likely to undergo TJR compared to those with normal weight. A dose–response relationship between BMI and TJR was observed (P < 0.001). Socioeconomic status was not independently associated with risk of either THR or TKR. CONCLUSION The strengths of the inverse association between smoking and TJR, the temporal relationship of the association, together with the consistency in the findings warrant further investigation about the role of smoking in the pathogenesis of osteoarthritis causing TJR.George Mnatzaganian, Philip Ryan, Christopher M Reid, David C Davidson and Janet E Hille

    Recent Developments in Gravity Gradiometry from the Space Shuttle-Borne Tethered Satellite System

    Abstract - This paper describes the current development of a noncryogenic gravity gradiometer for future use on board the tethered satellite system (TSS). The paper also proposes a way of testing a single\u2010axis prototype of such an instrument that makes use of the free\u2010fall technique in vacuum. This gravity gradiometer has a design sensitivity of the order of 10^-2 EU (1 EU =E\uf6tv\uf6s Unit=10^-9 ms-2/m) in 10\u2010s integration time. Because of the small size and light weight, a noncryogenic gravity gradiometer is a valid candidate to fit the limited accommodation capability of the TSS deployed from the shuttle and maintained, for several days, at an altitude of 120\u2013130 km. This unique capability improves the resolution and accuracy achievable in reconstructing the earth gravity field, in particular at short wavelengths. A preliminary analysis of the dynamics of the TSS satellite indicates the potential of such satellite as a low\u2010altitude research platform for gravity gradiometry
