Clothes for the Emperor or Can Graduate Schools Learn From Undergraduate Macroeconomics?
The current crisis is not only one of financial markets, but also of macroeconomics. Leading scholars call for a paradigm shift away from dynamic general equilibrium models, though some argue that the profession's arsenal already contains the tools and historical lessons needed to deal with such crises. Taking this view to the limit, this note demonstrates that the workhorse models of undergraduate macroeconomics not only permit a refined view and classification of financial crises. These models also identify scenarios under which either policymakers would be ill advised to follow conventional prescriptions, or full-scale depressions loom that cannot be fought by means of fiscal or monetary policy alone.Teaching macroeconomics, lessons, graduate, undergraduate, financial crisis, liquidity trap, risk premium
The macroeconomics of financial crises: How risk premiums, liquidity traps and perfect traps affect policy options
The paper shows that structural models of the IS-LM and Mundell-Fleming variety have a lot to tell about the macroeconomics of the current global crisis. PIGS or Lambs? The European Sovereign Debt Crisis and the Role of Rating Agencies
This paper asks whether rating agencies played a passive role or were an active driving force during Europe's sovereign debt crisis. We address this by estimating relationships between sovereign debt ratings and macroeconomic and structural variables. We then use these equ-ations to decompose actual ratings into systematic and arbitrary components that are not explained by observed previous procedures of rating agencies. Next, we check whether both systematic and arbitrary parts of credit ratings affect credit spreads. We find that both do, which opens the possibility that arbitrary rating downgrades trigger processes of self-fulfilling prophecy that may drive even relatively healthy countries towards default.Sovereign debt ratings, sovereign default, debt crisis, budget deficit, rating agencies, PIGS, risk premiums, government bond spreads.
Teaching Macroeconomics after the Crisis: A Survey among Undergraduate Instructors in Europe and the U.S.
An online survey among undergraduate macroeconomics instructors reveals that roughly half of them were scared when the crisis erupted and remain wary that more may be in the offing. As regards teaching, courses feature much the same lineups of models as they did before the crisis. A striking change concerns public debt dynamics, which receives much more emphasis. Regarding the finer fabric of undergraduate macro teaching, exciting things are going on. G-Protein-gekoppelten Endothelin-Rezeptor-Subtypen A und B die Aktivierung der
Phospholipase C (PLC) und somit einen Anstieg der intrazellulÀren Ca2+-Konzentration
induzieren. G-Protein-gekoppelte Rezeptoren (G protein coupled receptors, GPCRs)
unterliegen einer fein abgestimmten Regulation, um einer Ăberstimulation der Zelle
vorzubeugen. Der klassische Desensibilisierungsmechanismus beinhaltet die Phosphorylierung
des aktivierten Rezeptors durch GPCR-Kinasen (GRKs). Eine Phosphorylierung
durch diese Serin/Threonin-Kinasen erhöht die AffinitÀt des Rezeptors zu Arrestin,
welches daraufhin bindet und zu einer sterischen Blockade und somit zur Entkopplung
des Rezeptors vom G-Protein fĂŒhrt. Die Rezeptor-Phosphorylierung spielt fĂŒr viele GPCRs
ebenfalls eine Rolle in der Agonisten-induzierten Rezeptor-Endozytose (Internalisierung).
Im Endothelin A (ETA)-Rezeptor sind 15 Phosphorylierungsstellen (13 hiervon
Serin/Threonin-Reste) bekannt, die sich hauptsÀchlich in der Sequenz, die der
C-terminalen Palmitoylierungsstelle folgt, befinden. Diese Sequenz (im Folgenden als
C-terminale ExtremitÀt (CTE) bezeichnet) enthÀlt drei Serin- und sieben Threonin-Reste.
Sechs dieser zehn Phosphorylierungsstellen befinden sich in einem proximalen, vier in
einem distalen Cluster.
In der vorliegenden Arbeit wurde die Desensibilisierung des ETA-Rezeptors untersucht. Ob
die GRK2, deren Beteiligung an diesem Prozess bekannt ist, hier die Funktion einer Kinase
einnimmt und welche Phosphorylierungsstellen gegebenenfalls involviert sind, ist nicht
eindeutig geklÀrt. Daher wurden durch den Austausch von Serin und Threonin gegen
Alanin phosphorylierungsdefiziente ETA-Rezeptoren hergestellt und in der Zelllinie
HEK 293 transient exprimiert. WĂ€hrend kontinuierlicher Endothelin-1 (ET-1)-Stimulation
wurde die PLC-AktivitĂ€t analysiert, um Aufschluss ĂŒber die Regulation der phosphorylierungsdefizienten
ETA-Rezeptor-Mutanten zu erhalten.
Die kombinierte Substitution der in der CTE befindlichen Serin/Threonin-Reste, jedoch
nicht die Substitution aller proximalen oder aller distalen Phosphorylierungsstellen, fĂŒhrte
zu einer gestörten Desensibilisierung, was sich in einer Erhöhung des PLC-Signals unter
kontinuierlicher ET-1-Stimulation manifestierte. FĂŒr eine physiologische Desensibilisierung
schien das Vorhandensein mindestens eines intakten Serin/Threonin-Clusters
obligatorisch zu sein, denn die PLC-AktivitÀt jeder untersuchten Mutante mit einer
Kombination aus proximalen und distalen Substitutionen war gegenĂŒber der PLC-AktivitĂ€t
des Wildtyps signifikant erhöht.
Des Weiteren wurde die ET-1-induzierte PLC-AktivitÀt von Zellen untersucht, die den
ETA-Rezeptor und GRK2 transient koexprimierten. Neben der wildtypischen Kinase
(GRK2-WT) kamen die beiden Mutanten GRK2-D110A und GRK2-D110A/K220R zum
Einsatz. Die Mutation K220R wurde gewÀhlt, da sie in einer katalytisch inaktiven GRK2
resultiert. Die Mutation D110A beeintrÀchtigt hingegen die Bindung von GRK2 an Gαq. Die
rekombinante Expression von GRK2-WT fĂŒhrte sowohl beim wildtypischen ETA-Rezeptor
(ETA-WT) als auch bei der phosphorylierungsdefizienten Mutante ETA-6PD (mit einer
Kombination aus proximalen und distalen Substitutionen) zu einer Reduktion der
PLC-AktivitÀt. Durch eine Expression von GRK2-D110A wurde die ETA-WT-, jedoch nicht
die ETA-6PD-vermittelte PLC-AktivitÀt inhibiert. Beide Signale waren von einer GRK2-
D110A/K220R-Expression unbeeintrÀchtigt. Diese Ergebnisse belegen, dass in die
Desensibilisierung des ETA-Rezeptors mindestens zwei unabhÀngige GRK2-vermittelte
Komponenten involviert sind: 1) ein phosphorylierungsunabhÀngiger Mechanismus, der
durch die Bindung der Kinase an Gαq realisiert wird und 2) ein Mechanismus, der die
Phosphorylierung von Serin/Threonin-Resten in der CTE des Rezeptors beinhaltet und in
redundanter Weise in der Lage ist, entweder proximale oder distale Phosphorylierungsstellen
zu involvieren. Das Vorhandensein eines phosphorylierungsabhÀngigen Desensibilisierungsmechanismus
konnte durch GRK2-Knockdown-Experimente bestÀtigt werden.
In HEK 293-Zellen wurde zusÀtzlich die Internalisierung von ETA-WT und einer Mutante,
bei der alle bekannten Serin- und Threonin-Phosphorylierungsstellen entfernt wurden,
untersucht. HierfĂŒr wurden die auf der ZelloberflĂ€che exprimierten Rezeptoren mit HiLyte
Fluor 488-gekoppeltem ET-1 markiert. Die Internalisierung der fluoreszenzmarkierten
Rezeptoren wurde im konfokalen Laser Scanning-Mikroskop beobachtet. Es wurden keine
Unterschiede in der Endozytosegeschwindigkeit des wildtypischen und des phosphorylierungsdefizienten
Rezeptors festgestellt. Dies lÀsst den Schluss zu, dass die
Internalisierung des ETA-Rezeptors phosphorylierungsunabhÀngig geschieht.
Die in dieser Arbeit erzielten Ergebnisse deuten auf eine komplexe ETA-Rezeptor-
Regulation hin. Die gleichzeitige Beteiligung von phosphorylierungsabhÀngigen
und -unabhÀngigen Komponenten an der Desensibilisierung und Internalisierung geht
ĂŒber das klassische Paradigma der GPCR-Regulation hinaus und könnte im Zusammenhang
mit einer Zelltyp-spezifischen Feinabstimmung des Endothelin-Signals stehen.The vasoactive peptide hormones of the endothelin family activate phospholipase C (PLC)
via stimulation of the G protein coupled endothelin A and B receptors to induce an
increase of the intracellular Ca2+ concentration. G protein coupled receptors (GPCRs) are
tightly regulated to prevent cellular overstimulation. A well-known desensitization
mechanism involves the phosphorylation of the activated receptor by GPCR kinases
(GRKs). Phosphorylation of the receptor by these serine/threonine kinases promotes
binding of arrestin to the receptor which subsequently is uncoupled from its G protein by
steric inhibition. Receptor phosphorylation is also involved in the agonist induced
receptor endocytosis (internalization) of many GPCRs. The endothelin A (ETA) receptor
contains 15 confirmed phosphorylation sites (including 13 serine/threonine residues)
which are essentially located in the sequence following the C-terminal palmitoylation site.
This sequence is subsequently referred to as C-terminal extremity (CTE). It contains three
serine and seven threonine residues which are located in a proximal cluster (including six
phosphorylation sites) and a distal cluster (including four phosphorylation sites).
In this work the desensitization of the ETA receptor was analyzed. Involvement of GRK2 in
this process was shown in previous studies. Unclear however is whether GRK2 acts as a
kinase in this process or rather mediates ETA receptor desensitization in a
phosphorylation independent way. Furthermore, in the case of a phosphorylation
dependent desensitization mechanism, the phosphorylation sites are unknown.
Therefore, various phosphorylation deficient ETA receptor mutants were created by
substitution of serine/threonine-phosphoacceptor sites by alanine in different
combinations. The phospholipase C (PLC) activity of mutant receptors transiently
expressed in HEK-293 cells was analyzed during continuous endothelin-1 (ET-1)
stimulation to gain information about their regulation and an involvement of
phosphorylation in this process.
The total deletion of phosphoacceptor sites in the CTE affected receptor regulation
resulting in an increased PLC activity during continuous ET-1 stimulation. However,
proximal as well as distal Serine/Threonine-phosphoacceptor sites both turned out to be
sufficient to induce wild type-like desensitization. In this context, the integrity of either
the proximal or the distal phosphoacceptor cluster seemed to be mandatory for the wild
type-like desensitization because the second messenger production of any investigated
ETA receptor containing combined substitutions of proximal and distal Serine/Threoninephosphoacceptor
sites was significantly increased.
Additionally, the ET-1 induced PLC activity of cells transiently coexpressing the ETA
receptor and GRK2 was analyzed. Besides the wild type GRK2 (GRK2-WT) the mutants
GRK2-D110A and GRK2-D110A/K220R were used. The mutation K220R was chosen
because it results in the loss of kinase activity. The mutation D110A affects GRK2âs
coupling to Gαq. Recombinantly expressed GRK2-WT decreased the PLC activity mediated
by both the wild type ETA receptor (ETA-WT) and the phosphorylation deficient mutant
ETA-6PD containing proximal and distal substitutions. The expression of GRK2-D110A
suppressed ETA-WT signaling but did not impact PLC activity mediated by the
phosphorylation-deficient mutant ETA-6PD. Moreover, GRK2-D110A/K220R failed to
inhibit signaling of ETA-WT and ETA-6PD. This demonstrates that ETA desensitization
involves at least two autonomous GRK2-mediated components: 1) a phosphorylationindependent
signal decrease mediated by blocking of Gαq and 2) a mechanism involving
phosphorylation of serine and threonine residues in the CTE of the receptor in a
redundant fashion, able to incorporate either proximal or distal phosphoacceptor sites.
The existence of a phosphorylation dependent mechanism of desensitization was
confirmed by GRK2 knockdown experiments.
Furthermore, the internalization of ETA-WT and a mutant lacking all known
serine/threonine phosphorylation sites were analyzed. For this purpose, receptors
expressed on the cell surface were labeled by the addition of HiLyte Fluor 488 coupled
ET-1. The internalization of fluorescently labeled receptors was observed by confocal
laser scanning microscopy. No differences in the kinetics of endocytosis of the wild type
and the phosphorylation deficient receptors were detected. This suggests a
phosphorylation independent internalization mechanism for the ETA receptor.
The results of this work hint at a complex regulation of ETA receptor signaling. The
coexistence of diverse phosphorylation-dependent and -independent components of
desensitization and internalization exceeds the classical paradigm of GPCR regulation and
may be associated with cell specific fine-tuning of endothelin signaling
Untersuchungen ĂŒber die endogene Expression und Regulation des Natrium-Iodid-Symporters (NIS) in extrathyreoidalen Tumoren in vitro und in vivo
Aufgrund der Expression des Natrium-Iodid-Symporters (NIS) besitzt die SchilddrĂŒse die FĂ€higkeit zur Iodidanreicherung. Dieser Mechanismus ermöglicht die Radioiodtherapie benigner und maligner SchilddrĂŒsenerkrankungen. Da die Radioiodtherapie eine effektive und nebenwirkungsarme TherapiemodalitĂ€t der differenzierten SchilddrĂŒsenkarzinome ist, stellt sich die Frage, ob NIS auch in extrathyreoidalen Tumoren exprimiert ist und somit die Radioiodtherapie auch in diesen FĂ€llen eine Option darstellen kann.
In extrathyreoidalen Geweben findet sich eine physiologische Expression des NIS in erster Linie in der BrustdrĂŒse, im Magen, in den SpeicheldrĂŒsen und im Plexus choroideus. Extrathyreoidale Tumore betreffend wurden bisher einige Studien ĂŒber die AktivitĂ€t des NIS bei Mammakarzinomen veröffentlicht: NIS-Ăberexpression mit konsekutiver Iodidaufnahme wurde in Zelllinien humaner Mammakarzinome und auch bei Patienten nachgewiesen. Ăber die funktionelle Expression des NIS in weiteren extrathyreoidalen TumorentitĂ€ten ist jedoch deutlich weniger bekannt.
In der vorliegenden Arbeit wurde eine endogene Expression des NIS mit daraus resultierender Iodidaufnahme in murinen Zelllinien aus Mamma-, Rektum- und Lungenkarzinomen nachgewiesen. Die Expression des NIS wurde funktionell durch Iodid- bzw. Pertechnetataufnahme in vitro und in vivo in prĂ€klinischen Tumormodellen dokumentiert. Molekularbiologisch konnte NIS auf mRNA- und auf Proteinebene mittels real-time RT-PCR, ImmunfluoreszenzfĂ€rbungen und Immunoblots nachgewiesen werden. Es konnte in vitro eine Modulation der funktionellen Iodidaufnahme in AbhĂ€ngigkeit von der Tyrosinphosphorylierung und des cAMP-Spiegels demonstriert werden. In vivo konnte in subkutanen allotransplantierten Tumoren dieser Zelllinien eine Steigerung der NIS-AktivitĂ€t durch Inhibition von Tyrosinkinasen induziert werden. Des Weiteren wurden mehrere murine transgene Modelle kolorektaler Karzinome auf NIS-Ăberexpression untersucht: in 14 % der Tumorgewebsproben konnte eine erhöhte Expression der NIS-mRNA dokumentiert werden.
Zusammenfassend wurde erstmals eine aktive endogene Iodidaufnahme in Tumoren kolorektalen und bronchialen Ursprungs dokumentiert und der Nachweis erbracht, dass diese Iodidaufnahme auf eine endogene Ăberexpression des NIS zurĂŒckzufĂŒhren ist. Des Weiteren wurde eine Steigerung der Iodidaufnahme in diesen Tumoren durch Tyrosinkinaseinhibition demonstriert. Weitere Versuche sind erforderlich um diese Ergebnisse auf humane Gewebe zu ĂŒbertragen, mit dem langfristigen Ziel der Entwicklung neuer Anwendungsmöglichkeiten der Radionuklidtherapie in der Onkologie
