    Statistical Analysis of Winter Sulphur Dioxide Concentration Data in Vienna

    The paper describes two nonlinear regression models, applied to winter daily SO2 concentration data and to the corresponding meteorological data from the metropolitan area of Vienna. The first model accounts for the role of wind speed and temperature (a proxy for emissions due to residential heating) on average SO2 concentration in the area. The second regression has an additional wind direction input and tries to point out the contribution by the industrial emissions (located primarily near the south-eastern border of the area) to concentration in the most polluted subarea. Both models offer a satisfactory fitting performance (e.g., correlations around 0.85 between observed and regression values). However, since model validation is a critical point for regressions, sensitivity tests of model fitting performance are carried out by using various data sets for the estimation of regression coefficients. One of such tests points out that there is an "optimal length" of the data set to be used, namely neither a too short set nor a set including "too past" data offer a satisfactory fitting quality

    The object of this overall research, which has an expected duration of two years is to set up a scheme for predicting ground-level pollutant concentrations for real-time control purposes (i.e. the action to be taken at the emission sources in the presence of forthcoming high pollutant events). The forecasting scheme is described, together with its application to sulphur dioxide pollution in the Venetian lagoon area. The scheme is based on a complex mathematical model to be run on a computer and receiving at the beginning of each interval of time all the information (about the meteorological and emission situation) required for the prediction. The type of control action which should be taken on the basis of such a forecast is the object of this part of the research

    Air-quality management problems fall into three main classes: it is difficult to obtain a reliable picture of all the physicochemical processes involved, comprehensive assessments of the costs and benefits of alternative control strategies are not easily made, and the technology for pollution abatement is not yet well established. Various mathematical or formal management models do exist but the overall impact of modeling on decision making has so far been relatively small. The first aim of the IIASA Workshop on which this volume is based was to bridge the gap between air-quality modeling and management. As described in the ten papers in Part One, Workshop participants examined the goals actually pursued by decision makers, the potential role of mathematical models in air-quality management, and the extent to which modeling has been used in real situations in a number of countries. The Workshop's second aim, reported in the eight papers in Part Two, was to consider the unusual strategy of real-time emission control. An extended description of the IIASA case study of the Venetian Lagoon area was presented, together with contributions on real-time forecast and control schemes in operation in Japan and Italy

    PRIMA-1 induces autophagy in cancer cells carrying mutant or wild type p53.

    PRIMA-1 is a chemical compound identified as a growth suppressor of tumor cells expressing mutant p53. We previously found that in the MDA-MB-231 cell line expressing high level of the mutant p53-R280K protein, PRIMA-1 induced p53 ubiquitination and degradation associated to cell death. In this study, we investigated the ability of PRIMA-1 to induce autophagy in cancer cells. In MDA-MB-231 and HCT116 cells, expressing mutant or wild type p53, respectively, autophagy occurred following exposure to PRIMA-1, as shown by acridine orange staining, anti-LC3 immunofluorescence and immunoblots, as well as by electron microscopy. Autophagy was triggered also in the derivative cell lines knocked-down for p53, although to a different extent than in the parental cells expressing mutant or wild type p53. In particular, while wild type p53 limited PRIMA-1 induced autophagy, mutant p53 conversely promoted autophagy, thus sustaining cell viability following PRIMA-1 treatment. Therefore, the autophagic potential of PRIMA-1, besides being cell context dependent, could be modulated in a different way by the presence of wild type or mutant p53. Furthermore, since both cell lines lacking p53 were more sensitive to the cytotoxic effect of PRIMA-1 than the parental ones, our findings suggest that a deregulated autophagy may favor cell death induced by this drug

    Controle da ferrugem asiática da soja com diferentes épocas de aplicação de fungicidas.

    O objetivo do trabalho foi avaliar a resposta de linhagens da cultura da soja a diferentes épocas de aplicação de fungicidas no controle da ferrugem asiática. O experimento foi conduzido em condições de campo no Instituto Federal do Triângulo Mineiro- campus Uberaba. Foram utilizadas 2 linhagens promissoras (resistentes a ferrugem asiática) do Programa de Melhoramento Genético da Embrapa, EPAMIG/Fundação Triângulo (BRSMG 771F e MGBR08-77511) além de uma cultivar suscetível (BRSMG Vencedora) do mesmo programa e quatro épocas de aplicação de fungicida (Piraclostrobina & Epoxiconazol 0,50+ óleo mineral 0,5% dose L p.c.ha-1): T1- testemunha (sem aplicação de fungicida) T2- aplicação na fase R1+ 20 dias + 20 dias, T3- aplicação primeiras pústulas + retorno das pústulas+ retorno das pústulas, T4- aplicação primeiras pústulas + 20 dias após primeiras pústulas + R5.1, T5- aplicação na fase R5.1. A aplicação dos fungicidas foram efetuadas utilizando um pulverizador costal. O delineamento experimental utilizado foi um fatorial em blocos ao acaso 3x5x4 (3 linhagens x 5 épocas de aplicação de fungicidas x 4 repetições) totalizando 15 tratamentos. As parcelas experimentais constam de quatro linhas com seis metros de comprimento por dois metros de largura e o espaçamento entrelinhas de 0,5 metros. Após duas avaliações, observa-se para os tratamentos com variedades resistentes a ferrugem, que ocorre um atraso no início da doença no campo. Porém, ainda faz-se necessário avaliar o comportamento de tais materiais em condições de alta pressão de inoculo, o que ocorrerá até o fim do ciclo da cultura

    A produção de semente de soja livre de sementes de outras cultivares ou outras sementes, dentro dos limites estabelecidos pela legislação, é facilitada pelo fato da soja ser espécie autógama. No caso específico das cultivares geneticamente modificadas (GM), a baixa taxa de fecundação cruzada que ainda pode existir é um dos pontos críticos de contaminação genética dos campos de sementes convencionais por transgênicos. O objetivo deste trabalho foi verificar a eficiência de dois métodos de detecção de misturas de semente GM em amostras de soja convencional, para avaliar lotes de sementes quanto aos limites exigidos pela legislação. Foram utilizadas amostras de semente da cultivar BRSMG 810C (convencional) e da cultivar BRSMG 850GRR. Foram avaliados dois tamanhos de amostras (200 e 400 sementes), com quatro níveis de contaminação (semente GM adicionada às amostras convencionais, para se obter 0%, 0,5%, 1% e 1,5%), e dois métodos de detecção: imunoensaio de fluxo lateral e bioensaio (pré-embebição em solução de herbicida a 0,6% do equivalente ácido, na proporção de 2,5 vezes o peso do substrato, por 16 horas no escuro a 25oC). O método de bioensaio é eficiente na detecção da presença de GM em amostras de semente convencional de soja, desde que as mesmas apresentem alta qualidade fisiológica. Existe correlação significativa entre os valores obtidos pelo método de detecção por bioensaio e os níveis de contaminação estabelecidos (r= 0,82; p< ou = 0,0001). O tamanho da amostra não influenciou a eficiência dos dois métodos na detecção da presença de sementes adventícias (GM)

    06N-P63\u3b1 and TA-P63\u3b1 exhibit intrinsic differences in transactivation specificities that depend on distinct features of DNA target sites.

    TP63 is a member of the TP53 gene family that encodes for up to ten different TA and 06N isoforms through alternative promoter usage and alternative splicing. Besides being a master regulator of gene expression for squamous epithelial proliferation, differentiation and maintenance, P63, through differential expression of its isoforms, plays important roles in tumorigenesis. All P63 isoforms share an immunoglobulin-like folded DNA binding domain responsible for binding to sequence-specific response elements (REs), whose overall consensus sequence is similar to that of the canonical p53 RE. Using a defined assay in yeast, where P63 isoforms and RE sequences are the only variables, and gene expression assays in human cell lines, we demonstrated that human TA- and 06N-P63\u3b1 proteins exhibited differences in transactivation specificity not observed with the corresponding P73 or P53 protein isoforms. These differences 1) were dependent on specific features of the RE sequence, 2) could be related to intrinsic differences in their oligomeric state and cooperative DNA binding, and 3) appeared to be conserved in evolution. Since genotoxic stress can change relative ratio of TA- and 06N-P63\u3b1 protein levels, the different transactivation specificity of each P63 isoform could potentially influence cellular responses to specific stresses
