Tacaribe virus (TacV) is the prototype of the New World group of arenaviruses. The TacV genome encodes four proteins: the nucleoprotein (N), the glycoprotein precursor, the polymerase (L), and a RING finger protein (Z). Using a reverse genetics system, we demonstrated that TacV N and L are sufficient to drive transcription and replication mediated by TacV-like RNAs and that Z is a powerful inhibitor of these processes (Lopez et al., J. Virol. 65:12241-12251, 2001). More recently, we provided the first evidence of an interaction between Z and L and showed that Z's inhibitory activity was dependent on its ability to bind to L (Jácamo et al., J. Virol. 77:10383-10393, 2003). In the present study, we mapped the TacV Z-binding sites on the 2,210-amino-acid L polymerase. To that end, we performed deletion analysis and point mutations of L and studied the Z-L interaction by coimmunoprecipitation with specific sera. We found that the C-terminal region of L was not essential for the interaction and identified two noncontiguous regions that were critical for binding: one at the N-terminus of L between residues 156 and 292 and a second one in the polymerase domain (domain III). The importance of domain III in binding was revealed by substitutions in D1188 and H1189 within motif A and in each residue of the conserved SDD sequence (residues 1328, 1329, and 1330) within motif C. Our results showed that of the substituted residues, only H1189 and D1329 appeared to be critically involved in binding Z.Fil: Wilda, Maximiliano.

Differential Flight Muscle Development in Workers, Queens and Males of the Eusocial Bees, Apis mellifera and Scaptotrigona postica

    Get PDF
The flight capability of the adult eusocial bees, Apis mellifera L. and Scaptotrigona postica Latreille (Hymenoptera: Apidae), is intrinsically linked to their colonial functions, such as the nuptial flight for mating in the case of queens and males, and the exploration of new habitats for nesting and food sources in the case of workers. Flight is achieved by the contraction of indirect flight muscles that produce changes in thoracic volume and, therefore, wing movement. The purpose of this work is to examine possible differences in muscle development that may be associated with the flying activity of individuals in a given life stage considering the behavioral and physiological differences among the stages and between the two species studied. Measurements of the muscle fibers obtained from light microscopy preparations of muscle were submitted to statistical analysis in order to detect the differences at a given time, or throughout the life of the individual. The results show that muscle morphology is similar in both species, but in A. mellifera the muscle fibers are thicker and more numerous than in S. postica. Differences in the fiber thickness according to life stage in all classes of individuals of both species were detected. These results are discussed in relation to the need for flying in each life stage

Hidden in Plain Sight: Health Discrimination & Avoidance Among the SGM Population

Despite the advancements the American health care system has made to increase access, there still remains the prominent issue of healthcare avoidance. Specifically, the sexual and gender minority (SGM) population remains incredibly vulnerable. This is due to the stigma that those who are not visibly or socially presenting themselves as gender confirming or hetero-normative; they may be denied access to healthcare or may face discrimination in pursuit of proper health care. Current research aims at identifying potential deterrents to receiving health care amongst an emerging and increasing population. Data was collected through an online-survey and distributed through Reddit threads in which participants could respond anonymously to questions regarding SGM issues, but more specifically healthcare avoidance and fear of discrimination. A total of 1316 respondents composed the sample size in this SGM study. Measured variables in this study included: age, education, income, gender, sex assigned at birth, partner status, race, ACES score, and overall healthcare avoidance. Within this study, it was found that ACE scores, gender minority status, income, and fear of discrimination were positively correlated. The results were statistically significant (p \u3c 0.005). The dilemma of health care avoidance among the SGM population is not only evident in research, but also in practice. This community faces some of the highest rates of discrimination inherently and is one of the groups with the largest needs for timely and efficient health care regardless of their sexual preferences or gender expression. It is a primary objective of health care to help those in need, and that includes those who may not be visually conforming or fall within heteronormativity. Further research should be conducted to be more representative of the population, as well as help in reducing further discrimination or fear of discrimination in clinical settings

Tycho: Facilitation Support for Groupware User Tests

    Get PDF
Running user tests for groupware requires tracking the progress of multiple users performing co-dependent tasks, while facilitating and observing their coordinated actions. This requirement negatively impacts the main objective of user testing, which is detecting usability flaws. User testing of groupware becomes more challenging when run remotely. Even if there are tools for remote user testing that can get the job done for a single user, they are not prepared for collaborative scenarios. In this paper we argue that tool support for the facilitation of user tests of groupware is missing. Consequently, we propose a method for user tests with tool assistance that makes it possible to automate task synchronization, especially for different workflows that must be run concurrently. We evaluated our proposal by comparing it to a manually facilitated approach during on-site coordinated user tests. The results indicate that, while the task of designing and running user tests with tool support takes more time, it allows the testers to better focus on detecting usability problems.Fil: Grigera, Julián. Metallopanstimulin as a marker for head and neck cancer

    Get PDF
BACKGROUND: Metallopanstimulin (MPS-1) is a ribosomal protein that is found in elevated amounts in the sera of patients with head and neck squamous cell carcinoma (HNSCC). We used a test, denoted MPS-H, which detects MPS-1 and MPS-1-like proteins, to determine the relationship between MPS-H serum levels and clinical status of patients with, or at risk for, HNSCC. PATIENTS AND METHODS: A total of 125 patients were prospectively enrolled from a university head and neck oncology clinic. Participants included only newly diagnosed HNSCC patients. Two control groups, including 25 non-smokers and 64 smokers, were studied for comparison. A total of 821 serum samples collected over a twenty-four month period were analyzed by the MPS-H radioimmunoassay. RESULTS: HNSCC, non-smokers, and smokers had average MPS-H values of 41.5 ng/mL, 10.2 ng/mL, and 12.8 ng/mL, respectively (p = 0.0001). CONCLUSION: We conclude that MPS-1 and MPS-1-like proteins are elevated in patients with HNSCC, and that MPS-H appears to be a promising marker of presence of disease and response to treatment in HNSCC patients
