    The effect of 12 weeks Anethum graveolens (dill) on metabolic markers in patients with metabolic syndrome; A randomized double blind controlled trial

    Background: The clustering of metabolic abnormalities defined as metabolic syndrome is now both a public health and a clinical problem .While interest in herbal medicine has greatly increased, lack of human evidence to support efficacies shown in animals does exist. This clinical trial study designed to investigate whether herbal medicine, Anethum graveolens (dill) extract, could improve metabolic components in patients with metabolic syndrome. Methods: A double-blind, randomized, placebo-controlled trial using a parallel design was conducted. 24 subjects who had metabolic syndrome diagnostic criteria (update of ATP III) were randomly assigned to either dill extract (n = 12) or placebo (n = 12) for 3 months. Results: Across lipid component of metabolic syndrome, no significant differences in triglyceride (TG) concentration and high density lipoprotein cholesterol were seen between the two groups. However TG improved significantly from baseline (257.0 vs. 201.5p = 0.01) with dill treatment but such a significant effect was not observed in placebo group. Moreover, no significant differences in waist circumference, blood pressure and fasting blood sugar were seen between two groups after 3 months follow up period. Conclusion: In this small clinical trial in patients with metabolic syndrome, 12 weeks of dill extract treatment had a beneficial effect in terms of reducing TG from baseline. However dill treatment was not associated with a significant improvement in metabolic syndrome related markers compared to control group. Larger studies might be required to prove the efficacy and safety of long-Term administration of dill to resolve metabolic syndrome components. © 2012 Mansouri et al.; licensee BioMed Central Ltd

    Trophic level, food preference and feeding ecology of Rachycentron canadum (Linnaeus, 1766), in Hormuzgan Province waters (northern Persian Gulf and Oman Sea)

    This study examined the ecological factors associated with feeding in Rachycentron canadum (cobia) in the waters of Persian Gulf and Oman Sea. Seasonally variation of stomach condition of cobia male, females, pooled and size wise were analyzed. In general, 577 cobia were collected (340 males, 237 females) (20–161 cm Total length) from Sept. 2014 to Sept. 2015 by the Ferdows-1 survey ship and from the landing areas in Bandar Abbas harbor seasonally. Our results provide the first estimates of trophic level (Tr.L) (4.32±0.76, mean±Se), consumption per biomass (Q/B) (6.13), and aspect ratio of the caudal fin (ARc.f) (3.1) for R. canadum in the Hormuzgan Province. We also found that bony fish made up more than 80 percent of the relative importance of particular prey (IRI %), indicating R. canadum primarily preys on these species. The immature group (20–40 cm, T.L), tended to feed on Penaeidae family, small crabs, and mollusks. Hierarchical clustering revealed that the frequencies of different types of prey could be applied to categorize the length classes into three levels. The result of this study revealed that the immature group and the 20-40 cm length class tended to feed on Penaeidae family, crabs, and mollusks. Also, we found that males generally fed more than females in all seasons and that R. canadum had an average dietary intake. The findings of this study can be used to prepare a food formulation for cobia that resembles the native diet

    A Role for IFITM Proteins in Restriction of Mycobacterium tuberculosis Infection

    SummaryThe interferon (IFN)-induced transmembrane (IFITM) proteins are critical mediators of the host antiviral response. Here, we expand the role of IFITM proteins to host defense against intracellular bacterial infection by demonstrating that they restrict Mycobacterium tuberculosis (MTb) intracellular growth. Simultaneous knockdown of IFITM1, IFITM2, and IFITM3 by RNAi significantly enhances MTb growth in human monocytic and alveolar/epithelial cells, whereas individual overexpression of each IFITM impairs MTb growth in these cell types. Furthermore, MTb infection, Toll-like receptor 2 and 4 ligands, and several proinflammatory cytokines induce IFITM1–3 gene expression in human myeloid cells. We find that IFITM3 co-localizes with early and, in particular, late MTb phagosomes, and overexpression of IFITM3 enhances endosomal acidification in MTb-infected monocytic cells. These findings provide evidence that the antiviral IFITMs participate in the restriction of mycobacterial growth, and they implicate IFITM-mediated endosomal maturation in its antimycobacterial activity

    Modelling the extremes of seasonal viruses and hospital congestion: The example of flu in a Swiss hospital

    Viruses causing flu or milder coronavirus colds are often referred to as ‘seasonal viruses’ as they tend to subside in warmer months. In other words, meteorological conditions tend to impact the activity of viruses, and this information can be exploited for the operational management of hospitals. In this study, we use 3 years of daily data from one of the biggest hospitals in Switzerland and focus on modelling the extremes of hospital visits from patients showing flu-like symptoms and the number of positive flu cases. We propose employing a discrete generalized Pareto distribution for the number of positive and negative cases. Our modelling framework allows for the parameters of these distributions to be linked to covariate effects, and for outlying observations to be dealt with via a robust estimation approach. Because meteorological conditions may vary over time, we use meteorological and not calendar variations to explain hospital charge extremes, and our empirical findings highlight their significance. We propose a measure of hospital congestion and a related tool to estimate the resulting CaRe (Charge-at-Risk-estimation) under different meteorological conditions. The relevant numerical computations can be easily carried out using the freely available GJRM R package. The empirical effectiveness of the proposed method is assessed through a simulation study

    HIV-1 Replication Is Differentially Regulated by Distinct Clinical Strains of Mycobacterium tuberculosis

    (MTb), the causative agent of TB, are known to modify the host immune response in a strain-specific manner. However, a MTb strain-specific impact upon the regulation of HIV-1 replication has not previously been established.. Furthermore, we show that the distinct pattern of CDC1551 or HN878 induced HIV-1 replication is associated with significantly increased levels of TNF and IL-6, and of the transcription and nuclear translocation of the p65 subunit of the transcription factor NF-κB, by CDC1551 relative to HN878.These results provide a precedent for TB strain-specific effects upon HIV-1 replication and thus for TB strain-specific pathogenesis in the outcome of HIV-1/TB co-infection. MTb strain-specific factors and mechanisms involved in the regulation of HIV-1 during co-infection will be of importance in understanding the basic pathogenesis of HIV-1/TB co-infection

    An image-based modeling framework for predicting spatiotemporal brain cancer biology within individual patients

    Imaging is central to the clinical surveillance of brain tumors yet it provides limited insight into a tumor\u27s underlying biology. Machine learning and other mathematical modeling approaches can leverage paired magnetic resonance images and image-localized tissue samples to predict almost any characteristic of a tumor. Image-based modeling takes advantage of the spatial resolution of routine clinical scans and can be applied to measure biological differences within a tumor, changes over time, as well as the variance between patients. This approach is non-invasive and circumvents the intrinsic challenges of inter- and intratumoral heterogeneity that have historically hindered the complete assessment of tumor biology and treatment responsiveness. It can also reveal tumor characteristics that may guide both surgical and medical decision-making in real-time. Here we describe a general framework for the acquisition of image-localized biopsies and the construction of spatiotemporal radiomics models, as well as case examples of how this approach may be used to address clinically relevant questions
