    New application of decomposition of U(1) gauge potential:Aharonov-Bohm effect and Anderson-Higgs mechanism

    In this paper we study the Aharonov-Bohm (A-B) effect and Anderson-Higgs mechanism in Ginzburg-Landau model of superconductors from the perspective of the decomposition of U(1) gauge potential. By the Helmholtz theorem, we derive exactly the expression of the transverse gauge potential A\vec{A}_\perp in A-B experiment, which is gauge-invariant and physical. For the case of a bulk superconductor, we find that the gradient of the total phase field θ\theta provides the longitudinal component A{\vec A}_{\parallel}, which reflects the Anderson-Higgs mechanism. For the case of a superconductor ring, the gradient of the longitudinal phase field θ1\theta_1 provides the longitudinal component A{\vec A}_{\parallel}, while the transverse phase field θ2\theta_2 produces new physical effects such as the flux quantization inside a superconducting ring.Comment: 6 pages, no figures, final version to appear in Modern Physics Letters

    Preparation and biological application of antibodies against leucoanthocyanidin reductase and anthocyanidin reductase from grape berry

    Proanthocyanidins (PAs) endow wine with the flavor of bitterness and astringency. Both leucoanthocyanidin reductase (LAR) and anthocyanidin reductase (ANR) are two key enzymes of PA biosynthesis in grape berries, but the previous studies on these enzymes only focused on the transcriptional expression of these genes. Here, the full-length cDNAs of VvLAR1, VvLAR2 and VvANR, respectively, were cloned from wine grape berries and were then introduced into the pGEX-4T-1 expression vectors, which were highly expressed in Escherichia coli DH5α cells with the induction of the isopropyl-β-D-thiogalactoside (IPTG). The purified fusion proteins were used as the antigens to immunize rabbits, separately. The obtained antiserums were further purified to obtain the immunoglobulin G (Ig G) fractions, which were demonstrated to be capable of specifically immuno-recognizing the VvLAR1, VvLAR2 and VvANR from the crude protein extracts from grape berries with weight masses of approximately 43 kD. The analyses of translational expression of these enzyme genes during berry development and immunohistochemical localization of these proteins, by using the obtained antibodies, showed that a high amount of VvLAR1, VvLAR2 or VvANR was present at the pre-veraison stage and these enzyme proteins were all localized on the outer layer of the berry skin and the vascular bundle, as well as in the inner layer of the seed coat. This work provides an important basis for further studies on PA biosynthesis in grape berries.

    Observations on Copy Number Variations in a Kidney-yang Deficiency Syndrome Family

    We have performed an analysis of a family with kidney-yang deficiency syndrome (KDS) in order to determine the structural genomic variations through a novel approach designated as “copy number variants” (CNVs). Twelve KDS subjects and three healthy spouses from this family were included in this study. Genomic DNA samples were genotyped utilizing an Affymetrix 100 K single nucleotide polymorphism array, and CNVs were identified by Copy Number Algorithm (CNAT4.0, Affymetrix). Our results demonstrate that 447 deleted and 476 duplicated CNVs are shared among KDS subjects within the family. The homologus ratio of deleted CNVs was as high as 99.78%. One-copy-duplicated CNVs display mid-range homology. For two copies of duplicated CNVs (CNV4), a markedly heterologous ratio was observed. Therefore, with the important exception of CNV4, our data shows that CNVs shared among KDS subjects display typical Mendelian inheritance. A total of 113 genes with established functions were identified from the CNV flanks; significantly enriched genes surrounding CNVs may contribute to certain adaptive benefit. These genes could be classified into categories including: binding and transporter, cell cycle, signal transduction, biogenesis, nerve development, metabolism regulation and immune response. They can also be included into three pathways, that is, signal transduction, metabolic processes and immunological networks. Particularly, the results reported here are consistent with the extensive impairments observed in KDS patients, involving the mass-energy-information-carrying network. In conclusion, this article provides the first set of CNVs from KDS patients that will facilitate our further understanding of the genetic basis of KDS and will allow novel strategies for a rational therapy of this disease

    A case-control study on risk factors of breast cancer in China

    Introduction: To screen the risk factors associated with breast cancer among Chinese women in order to evaluate the individual risk of developing breast cancer among women in China. Material and methods: A case-control study on 416 breast cancer patients and 1156 matched controls was conducted in 14 hospitals in 8 provinces of China in 2008. Controls were age- and region-matched to the cases. Clinicians conducted in-person interviews with the subjects to collect information on demographics and suspected risk factors for breast cancer that are known worldwide. Conditional logistic regression was used to derive odds ratios (OR) and 95% confidence intervals (CI) for the associations between risk factors and breast cancer. Results: Compared with matched controls, women with breast cancer were significantly more likely to have higher body mass index (BMI, OR = 4.07, 95% CI; 2.98-5.55), history of benign breast disease (BBD) biopsy (OR = 1.68, 95% CI; 1.19-2.38), older age of menarche (AOM) (OR = 1.41, 95% CI: 107-187), stress anticipation (SA), for grade 1-4, OR = 2.15, 95% CI; 1.26-3.66; for grade 5-9, OR = 3.48, 95% CI; 2.03-5.95) and menopause (OR = 2.22, 95% CI: 1.50-3.282) at the level of p < 0.05. Family history of breast cancer (FHBC) in first-degree relatives (OR = 1.66, 95% CI; 0.77-3.59) and use of oral contraceptives (OC) (OR = 1.59, 95% CI; 0.83-3.05) were associated with an increased risk of breast cancer at the level of p < 0.20. Conclusions: Our results showed that BMI, history of BBD biopsy, older AOM, SA and menopause were associated with increased risk of breast cancer among Chinese women. The findings derived from the study provided some suggestions for population-based prevention and control of breast cancer in China.Medicine, General & InternalSCI(E)15ARTICLE2303-309

    Epidemiological and genomic analyses of human isolates of Streptococcus suis between 2005 and 2021 in Shenzhen, China

    Streptococcus suis (S. suis) is an important food-borne zoonotic pathogen that causes swine streptococcosis, which threatens human health and brings economic loss to the swine industry. Three-quarters of human S. suis infections are caused by serotype 2. A retrospective analysis of human S. suis cases in Shenzhen, a megacity in China, with high pork consumption, between 2005 and 2021 was conducted to understand its genomic epidemiology, pathogen virulence, and drug resistance characteristics. The epidemiological investigation showed that human cases of S. suis in Shenzhen were mainly associated with people who had been in close contact with raw pork or other swine products. Whole-genome sequence analysis showed that 33 human isolates in Shenzhen were dominated by serotype 2 (75.76%), followed by serotype 14 (24.24%), and the most prevalent sequence types (STs) were ST7 (48.48%) and ST1 (39.40%). ST242 (9.09%) and ST25 (3.03%), which were rarely reported, were also found. Phylogenetic analysis showed that the Shenzhen human isolates had close genetic relatedness to isolates from Guangxi (China), Sichuan (China), and Vietnam. We found a new 82 KB pathogenicity island (PAI) in the serotype 2 isolate that may play a role in sepsis. Similarly, a serotype 14 isolate, containing 78 KB PAI, was isolated from a patient presenting with streptococcal toxic shock syndrome (STSLS) who subsequently died. Multi-drug resistance (MDR) was high in human isolates of S. suis from Shenzhen. Most human isolates were resistant to tetracycline, streptomycin, erythromycin, and clindamycin, and 13 isolates had intermediate resistance to penicillin. In conclusion, swine importation from Guangxi, Sichuan, and Vietnam should be more closely monitored, and the use of antibiotics limited to reduce the potential for antimicrobial resistance (AMR)
