    Marked attenuation of inflammatory mediator-induced C-fiber sensitization for mechanical and hypotonic stimuli in TRPV4-/- mice

    Inflammatory mediators can directly sensitize primary afferent nociceptors to mechanical and osmotic stimuli. Sensitized nociceptors have a lowered threshold of activation and increased spontaneous activity, which result in symptoms of hyperalgesia and pain, respectively. The transient receptor potential vanilloid 4 (TRPV4) ligand-gated ion channel has been implicated in the hyperalgesia for mechanical and osmotic stimuli associated with inflammatory states. To investigate whether TRPV4 directly contributes to the mechanisms of inflammatory mediator sensitization of C-fiber nociceptors, we compared the effect of the injection of simplified inflammatory soup (prostaglandin E2 and serotonin) into the mechanical receptive fields of C-fibers in TRPV4+/+ and TRPV4-/- mice in vivo. Following the injection of the soup, the percentage of C-fibers responding to a hypotonic stimulus and the magnitude of the response was significantly greater in TRPV4+/+ mice compared to TRPV4-/- mice. Moreover, in response to simplified inflammatory soup only C-fibers from TRPV4+/+ mice exhibited increased spontaneous activity and decreased mechanical threshold. These marked impairments in the response of C-fibers in TRPV4-/- mice demonstrate the importance of TRPV4 in nociceptor sensitization; we suggest that TRPV4, as TRPV1, underlies the nociceptive effects of multiple inflammatory mediators on primary afferent

    Moving Horizon Trend Identification Based on Switching Models for Data Driven Decomposition of Fluid Flows

    Modal decomposition is pretty popular in fluid mechanics, especially for data-driven analysis. Dynamic mode decomposition (DMD) allows to identify the modes that describe complex phenomenona such as those physically modelled by the Navier-Stokes equation. The identified modes are associated with residuals, which can be used to detect a meaningful change of regime, e.g., the formation of a vortex. Toward this end, moving horizon estimation (MHE) is applied to identify the trend of the norm of the residuals that result from the application of DMD for the purpose to automatically classify the time evolution of fluid flows. The trend dynamics is modelled as a switching nonlinear system and hence an MHE problem is solved in such a way to monitor the time behavior of the fluid and quickly identify changes of regime. The stability of the estimation error given by MHE is proved. The combination of DMD and MHE provide successful results as shown by processing experimental datasets of the velocity field of fluid flows obtained by a particle image velocimetry

    A comprehensive analysis of the psychometric properties of the contingencies of self-worth scale (CSWS)

    The Contingencies of Self-Worth Scale (CSWS) is a widely used personality self-report questionnaire developed for measuring the domains in which self-esteem is sustained by successes and achievements as well as threatened by obstacles and failures. Two studies (N study1 = 453, N study2 = 293) aimed to further refine our knowledge of its psychometric properties. Results attested that, at the first-order level, the originally hypothesized seven-factor model proved to be the best-fitting one, but the inclusion of a method factor significantly improved the fit to the data. At the second-order level, the model with two higher-order variables representing private sphere and public sphere of CSW fit better than alternative models. Finally, there was evidence that first- and second-order domains had a good degree of construct and discriminant validity. Overall, these studies provided a step forward in refining the psychometric structure of the CSWS

    The synergistic effect between positivity, socio-demographic factors and smoking cessation: results of a cohort study

    OBJECTIVE: To examine the extent to which a effect does exist between Positivity (POS), smoking and socio-demographic factors in determining quitting smoking in subjects participating in a Group Counselling Program (GCP) for smoking cessation.METHODS: 481 subjects were contacted through a telephone call. A logistic regression analysis was carried out. Possible interaction between sociodemographic variables and POS level was tested using the Synergism Index (SI).RESULTS: For individuals with a POS level over or equal to 3.4 the odds of being smoker was significantly higher among females (OR = 1.55), who smoked at home (OR = 2.16) and lower if there had children at home (OR = 0.53). For individuals with a POS level under 3.4, the only significant variable associated with smoking was beinga female (OR = 2.58). As far concerns the synergistic effect between the variables considered does exist between POS levels and having children at home (SI=1.13) and female gender (SI = 2.8).CONCLUSIONS: The synergistic effect between POS and sociodemographic factors adds evidence on the use of POS as possible determinants of individual happiness

    Consumers on critique: a survey of classical music listeners’ engagement with professional music reviews

    Music criticism has a long tradition as a leading agent in the classical music discourse. However, some people question its function in the contemporary music market. We explored the topicality of classical music critique by asking: Who reads professional reviews today? And what do readers expect from review? Through an online survey (English/German), we profiled the listening habits of classical music listeners (N = 1200) and their engagement with professional reviews. Our participants were more actively engaged with music, but contrary to the ‘highbrow’ stereotype, not more highly musically trained than the general population. They consumed music and opinion sources in a variety of ways. Approximately two-thirds (n = 741) of the participants had recently engaged with professional reviews, which were perceived as the most useful form of opinion, followed by short written commentaries and, lastly, ratings. A multiple logistic regression model suggested that the typical consumer of professional music critique was older with higher levels of musical engagement and education, had a higher inclination to purchase music and lower usage of streaming services, and had a preference for detailed reviews from traditional sources (e.g. newspapers). According to review readers, reviews should cover a variety of topics and offer evaluations underpinned with reasons. Reviewers should be constructive, open-minded, respectful, and well informed; their professional background was less relevant. Professional reviews should not necessarily provide a recommendation on what to buy, but rather guide listeners’ musical appreciation and understanding. Professional criticism still has an audience, although more so among older, musically educated listeners. Critics need to explore various channels in order to connect to a new generation of classical music listeners

    Genetic diversity of castanea sativa Mill. accessions from the Tuscan-Emilian Apennines and Emilia Romagna region (italy)

    This work investigated the genetic diversity of 134 Castanea sativa Mill. accessions present in the Italian region of Emilia-Romagna. Samples were taken from three collection fields (Granaglione, Zocca and Paloneta) in the Tuscan-Emilian Apennines. The accessions were analyzed by using 16 microsatellite markers (SSR). Genetic distances among accessions, calculated through the DICE coefficient, were used to construct an UPGMA cluster analysis. One major genotype (named "Marroni") was identified across the three investigated collection fields; this variety corresponds to a sweet chestnut cultivar that has been propagated and widely diffused in the Emilia-Romagna region. Other genotypes were represented by different varieties of Italian chestnuts. The results of this study will be used to define and share guidelines for the characterization and varietal certification of the chestnut varieties in the Emilia-Romagna region
