    This thesis sets out to explore the influence of Nietzsche upon Hesse throughout the latter's lifetime. It concentrates mainly on the major novels, although there have been one or two divergences from these where space has permitted. During his literary career, Hesse also made many references to Nietzsche in letters, essays and reviews, and these have been incorporated as much as possible into this text. At the beginning of his career, Hesse was drawn to Nietzsche both by his theory of aesthetics, an attraction which soon faded, and by his use of language, for which he retained life-long admiration. Hesse replaced his enthusiasm for Nietzsche's aesthetics with a more general interest in Nietzsche the man and poet. In the following years his attention turned to Nietzsche's theories of the artist (a perennial theme in his own works). After a lapse in interest came the period of his most intense involvement with Nietzsche during the latter half of, and immediately following the First World War, for it was at this time that he felt the greatest personal empathy with Nietzsche. The next years show Hesse incorporating various aspects of Nietzsche's . philosophy, especially his theories of cultural disease and decadence into his 'outsider' novels. As Hesse aged, he grew away from Nietzsche, seeing Jakob Burckhardt as the major influence upon the second half of his life, but signs of Nietzschean influence are still evident in his last two major novels, and he retained his regard for Nietzsche until the end of his life. This thesis traces in chronological order the various phases of Hesse's interest in Nietzsche and attempts to assess the influence asserted by Nietzsche in both the literary and the personal spheres of Hesse's life

    Understanding One’s Own Teaching Experiences: New Family and Consumer Sciences Teachers Reflect on Their First-Year of Teaching Through Video-Recorded Classroom Observations and Interviews

    The purpose of this study was to examine the experiences of new Family and Consumer Sciences (FCS) teachers and to describe their perceptions of classroom teaching readiness. The researcher completed a series of three classroom observations and three interviews of five first-year FCS teachers. The Danielson’s Framework for Teaching instrument was used for all observations and two sections of interviews used Interview Questions for Family and Consumer Sciences Educators instrument, which was taken in part, with permission, by Horizon Research, incorporated. All observations and interviews were completed over a time span of two to three weeks and examined essential teaching skills and Family and Consumer Sciences pedagogical content knowledge. In addition, this study examined their content knowledge and perceived quality of their student teaching experience during their teacher preparation semester. This study was completed during the spring of 2020 with five participants who had graduated from Pittsburg State University in Pittsburg, Kansas. This case study was qualitative in design with the purpose to learn more about a unique phenomenon: first-year FCS teachers’ readiness. While the majority of participants indicated they felt prepared to teach in the examined content areas and felt comfortable with the progression of their essential teaching skills, they recognized that some aspects of their teaching needed improvement, particularly: (a) resolving lesson planning to reflect higher-order thinking, (b) implementation of routines and procedures, (c) engaging students in learning, and (d) applying more accurate ways of reflecting on teaching. Although the majority of participants described their student teaching as positive, they mentioned there were improvements to be made in teacher education curriculum at the post-secondary level. Although there was much literature regarding teacher effectiveness, there was a lack of literature concerning Family and Consumer Sciences teachers. This study was created to add to existing research, to give voice to those individuals in their first-year of teaching, and to assess content and pedagogical areas of strengths and weaknesses. The results of this study will provide educators, mentors, administrators, and researchers with a better understanding of how new Family and Consumer Sciences teachers were prepared to teach their content areas and where they need to improve in essential teaching skills

The focus of this guide is to provide insight and recommendations to the undocumented population in relation to graduate school and life after graduation. Extensive guides and resources currently exist for undocumented students who have received DACA protection, equivalent resources and guides are scarce for undocumented graduate students who do not have DACA protection.     An EIAV field isolate reveals much higher levels of subtype variability than currently reported for the equine lentivirus family

    <p>Abstract</p> <p>Background</p> <p>Equine infectious anemia virus (EIAV), a lentivirus that infects horses, has been utilized as an animal model for the study of HIV. Furthermore, the disease associated with the equine lentivirus poses a significant challenge to veterinary medicine around the world. As with all lentiviruses, EIAV has been shown to have a high propensity for genomic sequence and antigenic variation, especially in its envelope (Env) proteins. Recent studies have demonstrated Env variation to be a major determinant of vaccine efficacy, emphasizing the importance of defining natural variation among field isolates of EIAV. To date, however, published EIAV sequences have been reported only for cell-adapted strains of virus, predominantly derived from a single primary virus isolate, EIAV<sub>Wyoming </sub>(EIAV<sub>WY</sub>).</p> <p>Results</p> <p>We present here the first characterization of the Env protein of a natural primary isolate from Pennsylvania (EIAV<sub>PA</sub>) since the widely utilized and referenced EIAV<sub>WY </sub>strain. The data demonstrated that the level of EIAV<sub>PA </sub>Env amino acid sequence variation, approximately 40% as compared to EIAV<sub>WY</sub>, is much greater than current perceptions or published reports of natural EIAV variation between field isolates. This variation did not appear to give rise to changes in the predicted secondary structure of the proteins. While the EIAV<sub>PA </sub>Env was serologically cross reactive with the Env proteins of the cell-adapted reference strain, EIAV<sub>PV </sub>(derivative of EIAV<sub>WY</sub>), the two variant Envs were shown to lack any cross neutralization by immune serum from horses infected with the respective virus strains.</p> <p>Conclusion</p> <p>Taking into account the significance of serum neutralization to universal vaccine efficacy, these findings are crucial considerations towards successful EIAV vaccine development and the potential inclusion of field isolate Envs in vaccine candidates.</p

    Dentes intencionalmente modificados e etnicidade em cemitérios do Brasil Colônia e Império

    A modificação intencional dos dentes foi muito difundida na África, vindo com escravos para a América. No Brasil foram encontradas em alguns sítios arqueológicos, sendo aqui estudadas em dois cemitérios históricos do Brasil: Pretos Novos, no Rio de Janeiro (único cemitério de um mercado de escravos na América), e cemitérios da antiga Sé de Salvador. Entre 570 dentes (30 indivíduos) dos Pretos Novos foram encontrados 13 dentes modificados. Entre 3181 dentes da Sé (62 enterros primários do séc. XVIII, e número indeterminado de re-deposições de outros períodos) foram encontrados 122 dentes modificados. Sua análise resultou em 13 tipos de modificações, a maior parte nos incisivos centrais superiores, e 10 tipos de arcos dentários modificados, evidenciando-se a técnica de percussão e lascamento dos dentes na maior parte dos casos. O achado de diferentes estilos de modificações sugere diferentes origens geográficas para os escravos do Rio e Salvador, o que é consistente com os dados históricos. No adro da Sé, a proximidade de enterros com modificações dentárias semelhantes sugere laços étnicos ou sociais, talvez de parentesco, entre os mortos.Intentional modification of the teeth was very common in Africa, coming to America with the slaves. In two historical cemeteries of Brazil, named Pretos Novos, in Rio de Janeiro (the only burial ground from a slave market ever known in America) and the burial ground of the old Sé, the more important church of Salvador.13 modified teeth were identified among 570 teeth (30 individuals) from Pretos Novos. 122 modified teeth were identified among 3181 teeth from Sé (62 individuals, primary burials, 18th century and non estimated number of comingled skeletons from other periods). The result of the analysis revealed 13 different modification styles, and 10 different compositions of modified dental arches. The differences are suggestive of different geographical origins for the slaves in both cemeteries, and this result is consistent with historical data. In the Sé burial ground, the proximity of burials of individuals with similar dental modifications is suggestive of ethnic, or social, may be familial links of the dead

    Envelope determinants of equine lentiviral vaccine protection

    Lentiviral envelope (Env) antigenic variation and associated immune evasion present major obstacles to vaccine development. The concept that Env is a critical determinant for vaccine efficacy is well accepted, however defined correlates of protection associated with Env variation have yet to be determined. We reported an attenuated equine infectious anemia virus (EIAV) vaccine study that directly examined the effect of lentiviral Env sequence variation on vaccine efficacy. The study identified a significant, inverse, linear correlation between vaccine efficacy and increasing divergence of the challenge virus Env gp90 protein compared to the vaccine virus gp90. The report demonstrated approximately 100% protection of immunized ponies from disease after challenge by virus with a homologous gp90 (EV0), and roughly 40% protection against challenge by virus (EV13) with a gp90 13% divergent from the vaccine strain. In the current study we examine whether the protection observed when challenging with the EV0 strain could be conferred to animals via chimeric challenge viruses between the EV0 and EV13 strains, allowing for mapping of protection to specific Env sequences. Viruses containing the EV13 proviral backbone and selected domains of the EV0 gp90 were constructed and in vitro and in vivo infectivity examined. Vaccine efficacy studies indicated that homology between the vaccine strain gp90 and the N-terminus of the challenge strain gp90 was capable of inducing immunity that resulted in significantly lower levels of post-challenge virus and significantly delayed the onset of disease. However, a homologous N-terminal region alone inserted in the EV13 backbone could not impart the 100% protection observed with the EV0 strain. Data presented here denote the complicated and potentially contradictory relationship between in vitro virulence and in vivo pathogenicity. The study highlights the importance of structural conformation for immunogens and emphasizes the need for antibody binding, not neutralizing, assays that correlate with vaccine protection
