The Quest for Orthologs orthology benchmark service in 2022
The Orthology Benchmark Service (https://orthology.benchmarkservice.org) is the gold standard for orthology inference evaluation, supported and maintained by the Quest for Orthologs consortium. It is an essential resource to compare existing and new methods of orthology inference (the bedrock for many comparative genomics and phylogenetic analysis) over a standard dataset and through common procedures. The Quest for Orthologs Consortium is dedicated to maintaining the resource up to date, through regular updates of the Reference Proteomes and increasingly accessible data through the OpenEBench platform. MicroRNA-mediated down-regulation of NKG2D ligands contributes to glioma immune escape
Malignant gliomas are intrinsic brain tumors with a dismal prognosis. They are well-adapted to hypoxic conditions and poorly immunogenic. NKG2D is one of the major activating receptors of natural killer (NK) cells and binds to several ligands (NKG2DL). Here we evaluated the impact of miRNA on the expression of NKG2DL in glioma cells including stem-like glioma cells. Three of the candidate miRNA predicted to target NKG2DL were expressed in various glioma cell lines as well as in glioblastomas in vivo: miR-20a, miR-93 and miR-106b. LNA inhibitor-mediated miRNA silencing up-regulated cell surface NKG2DL expression, which translated into increased susceptibility to NK cell-mediated lysis. This effect was reversed by neutralizing NKG2D antibodies, confirming that enhanced lysis upon miRNA silencing was mediated through the NKG2D system. Hypoxia, a hallmark of glioblastomas in vivo, down-regulated the expression of NKG2DL on glioma cells, associated with reduced susceptibility to NK cell-mediated lysis. This process, however, was not mediated through any of the examined miRNA. Accordingly, both hypoxia and the expression of miRNA targeting NKG2DL may contribute to the immune evasion of glioma cells at the level of the NKG2D recognition pathway. Targeting miRNA may therefore represent a novel approach to increase the immunogenicity of glioblastoma
Cell-derived extracellular vesicles can be used as a biomarker reservoir for glioblastoma tumor subtyping
Glioblastoma (GBM) is one of the most aggressive solid tumors for which treatment options and biomarkers are limited. Small extracellular vesicles (sEVs) produced by both GBM and stromal cells are central in the inter-cellular communication that is taking place in the tumor bulk. As tumor sEVs are accessible in biofluids, recent reports have suggested that sEVs contain valuable biomarkers for GBM patient diagnosis and follow-up. The aim of the current study was to describe the protein content of sEVs produced by different GBM cell lines and patient-derived stem cells. Our results reveal that the content of the sEVs mirrors the phenotypic signature of the respective GBM cells, leading to the description of potential informative sEV-associated biomarkers for GBM subtyping, such as CD44. Overall, these data could assist future GBM in vitro studies and provide insights for the development of new diagnostic and therapeutic methods as well as personalized treatment strategies
Prazer e dor na docência: revisão bibliográfica sobre a Síndrome de Burnout
Os transtornos mentais, atualmente, são os principais responsáveis pelo afastamento do trabalho por longos períodos de tempo. São eles que conferem riscos para a manutenção da saúde mental, através do comportamento e da emoção. Burnout é o resultado ao estresse crônico, incrementado na interação com outras pessoas. Esta pesquisa objetiva apresentar algumas reflexões acerca dos principais fatores de risco para o desenvolvimento da Síndrome de Burnout entre os docentes, a fim de compreendê-los dentro de um processo de desgaste físico-emocional em decorrência do trabalho. Para tanto, realizou-se uma revisão bibliográfica, de natureza qualitativa, na base de dados da Biblioteca Virtual em Saúde (BVS/BIREME), de artigos sobre a temática em questão. Os resultados deste estudo demonstram a carência de pesquisas nessa área. É necessário aprofundar o conhecimento sobre a manifestação do estresse ocupacional entre os docentes, a fim de se compreender e elucidar alguns problemas enfrentados por essa atividade, como a insatisfação profissional, o baixo rendimento no trabalho, o absenteísmo e algumas doenças ocupacionais, dentre elas o Burnout.Today, the mental disorders are mainly responsible ones for the absence from work for long periods of time. They are who confer risks for the maintenance of the mental health, through the behavior and of the emotion. Burnout is a reply of one stress chronic, caused by the interaction with other people. This research aims to present some reflections on the main risk factors for the development of Syndrome of Burnout between teachers, in order to understand them within a physical and emotional wear as a result of the work. For such, a bibliographic search, of qualitative nature, was made in the Virtual Health Library (BVS/BIREME), articles on the topic in question. The results of this study demonstrate the lack of research in this area. It is necessaryto increase knowledge about the manifestation occupational stress between teachers, in order to understand and elucidate some problems faced by this activity, such as dissatisfaction, low work performance, absenteeism and some occupational diseases, among them Burnout
