    Efeito da casca de Stryphnodendron adstringens (barbatimão) em modelos de nocicepção animais

    The antinociceptive activity of the crude extract and fractions of Stryphnodendron adstringens (Martius) Coville (barbatimão) was evaluated. Three experimental models of pain induction were used: abdominal writhing, formalin, and hot plate. The results demonstrated an antinociceptive effect of barbatimão in the experimental models of writhing induced by acetic acid and pain induced by formalin. However, the extracts did not significantly alter latency time on the hot plate in mice. These results suggest that barbatimão extract has a peripheral antinociceptive effect.A atividade antinociceptiva do extrato bruto e frações do Stryphnodendron adstringens (Mart.) Coville (barbatimão) foi avaliada. Foram usados três modelos experimentais para avaliação da dor: teste de contorção abdominal induzida pelo ácido acético, teste da formalina e teste da placa quente. Os resultados mostraram um efeito antinociceptivo evidente do barbatimão nos modelos experimentais de contorção induzida por ácido acético e dor induzida pela formalina. Por outro lado, o barbatimão não modificou significativamente o tempo de latência dos animais no teste da placa quente. Estes resultados sugerem que o extrato de barbatimão apresenta efeito antinociceptivo por mecanismos periféricos

    Inhibitory Effect of Helicteres gardneriana Ethanol Extract on Acute Inflammation

    The anti-inflammatory effect of an ethanol extract of Helicteres gardneriana (Nees) Castiglioni was assayed in experimental models of pleurisy and microcirculation in situ. Treatment of animals with 500 mg/kg body weight reduced the exudate volume (35% reduction) induced by intrapleural injection of carrageenan and the migration of polymorphonuclear cells into the inflamed pleural cavity of rats (40%). Additionally, rolling and adhesion of leukocytes and the number of leukocytes that migrated toward the perivascular space in response to the carrageenan injection were decreased by the extract (500 mg/kg). These data demonstrate the anti-inflammatory effect of the ethanol extract of Helicteres gardneriana and imply that inhibition of leukocyte-endothelial interactions is important in the extract's mechanism of action

    Antiinflammatory activity of Cayaponia podantha crude extract and fractions

    The effects of the crude extract and fractions of Cayaponia podantha (Cp) on experimental inflammation models were investigated. Paw edema induced by carrageenan (Cg) and peritonitis induced by Cg, LPS, and LTB4 were evaluated in rats treated orally with different doses of extract. Croton oil (CO) induced ear edema and the determination of MPO activity were evaluated in mice. Crude Cp extract and hexane (HF), ethyl-acetate (AF) and hidromethanol (MF) fractions were topically applied immediately after the application of the CO. Four hours after Cg injection, animals treated with crude extract (250 and 500 mg/kg) displayed significantly decreased paw edema. The Cp extract (250, 500, and 750 mg/kg) decreased vascular permeability and leukocyte migration in the peritonitis model in the 3rd h after induction of the inflammatory reaction. Furthermore, the 500 mg/kg dose of Cp extract also reduced LPS- and LTB4-induced migration. Crude extract and hexane and ethyl-acetate fractions (5.0 mg) significantly inhibited ear edema and MPO activity. Our results showed that Cp crude extract and fractions exhibited anti-inflammatory effects when they are administered orally or topically in animals.Colegio de Farmacéuticos de la Provincia de Buenos Aire

    Use of Propolis Hydroalcoholic Extract to Treat Colitis Experimentally Induced in Rats by 2,4,6-Trinitrobenzenesulfonic Acid

    This study focused on the therapeutic effect of a propolis SLNC 106PI extract on experimental colitis. Wistar adult rats received 0.8 mL rectal dose of one of the following solutions: saline (group S), 20 mg TNBS in 50% ethanol (group TNBS), 20 mg TNBS in 50% ethanol and propolis extract in saline (group TNBS-P), propolis extract in saline (group SP), and 20 mg TNBS in 50% ethanol and 50 mg/kg mesalazine (group TNBS-M). The animals were euthanized 7 or 14 days after the colitis induction. Samples of the distal colon were harvested for the analysis of myeloperoxidase (MPO) enzyme activity and for morphometric analysis in paraffin-embedded histological sections with hematoxylin-eosin or histochemical staining. The animals treated with TNBS exhibited the typical clinical signs of colitis. Increased MPO activity confirmed the presence of inflammation. TNBS induced the development of megacolon, ulceration, transmural inflammatory infiltrate, and thickened bowel walls. Treatment with propolis moderately reduced the inflammatory response, decreased the number of cysts and abscesses, inhibited epithelial proliferation, and increased the number of goblet cells. The anti-inflammatory activity of the propolis SLNC 106 extract was confirmed by the reductions in both the inflammatory infiltrate and the number of cysts and abscesses in the colon mucosa

    Effect of crude extracts of leaves of Smallanthus sonchifolius (yacon) on glycemia in diabetic rats

    O presente estudo investigou o efeito do extrato bruto das folhas de yacon, obtido por extração a frio (Y C) ou a quente (Y H), ou por extração hidro-alcoólica (Y E), na glicemia de ratos diabéticos (induzidos por estreptozocina, 50 mg/kg, i.p.) e não diabéticos. Os animais foram tratados por via oral com os extratos (400 mg/kg) ou água por um período de 3, 7, 10 e 14 dias. Após o tratamento, os animais foram anestesiados e amostras de sangue foram coletadas para determinações dos níveis de glicose e creatinina plasmática e da atividade de enzimas (AST, ALT, ALP). Também foi avaliado o consumo de ração, água, volume de urina, peso corporal dos animais e a DL50 do Y E. O tratamento dos animais com Y E, por 14 dias, reduziu a glicemia de ratos diabéticos e não diabéticos. Adicionalmente, o Y E restaurou a atividade das enzimas que estavam alteradas, e melhorou o ganho de peso dos animais diabéticos. Estes resultados demonstraram que a efetividade do extrato de yacon está relacionada ao método de preparação do extrato e ao tempo de tratamento dos animais. O efeito do Y E na glicemia de ratos diabéticos não está relacionado à redução da ingesta ou a interferência do extrato na absorção intestinal de carboidratos.The present study investigated the effect of crude extracts from yacon leaves, obtained by aqueous cold (Y C) or hot (Y H) extraction, or hydro-ethanolic extraction (Y E), on glycemia of the diabetic (induced by streptozotocine, 50 mg/kg, ip) and nondiabetic rats. The animals were treated orally with the extracts (400 mg/kg) or water for 3, 7, 10, and 14 days. After this period, the animals were anesthetized and blood samples were collected for measurement of plasmatic glucose and creatinine and activity of the enzymes (AST, ALT, ALP). Also were evaluated the consumption of food, water, urine volume, body weight of the animals and LD50 of Y E. Treatment with Y E for 14 days reduced glycemia in diabetic and nondiabetic animals. No similar reduction was observed in animals treated with Y C or Y H. Additionally, Y E restored the activity of the plasmatic enzymes that were altered, and improved weight gain in the diabetic animals. These results showed that the effectiveness of the yacon extracts is related to the method of preparation and to the time of treatment. The effect of Y E on glycemia in diabetic rats is not related to reduction of food intake or to interference of the extract with intestinal absorption of carbohydrates

    Analysis of myosin-V immunoreactive myenteric neurons from arthritic rats

    CONTEXT: The inflammatory response itself and the consequent oxidative stress are able to promote neurodegeneration. So, it is possible that enteric nervous system is affected by inflammatory diseases threatening quality of life of patients. However, gastrointestinal symptoms of arthritis are usually attributed to anti-inflammatory drugs rather than neural damage. OBJECTIVE: To confirm if the general population of myenteric neurons from the ileum and jejunum of rats is affected by arthritis. METHODS: Twenty Holtzmann rats, 58-day-old male, were used and divided in four groups: control group (C30), arthritic group (Art30), older control group (C60) and older arthritic group (Art60). At 58 days old, the animals in groups Art30 and Art60 received an injection of the complete Freund's adjuvant in order to induce arthritis. The whole-mount preparations of ileum and jejunum were processed for myosin-V immunohistochemistry. Quantitative and morphometric analyses were performed. RESULTS: Groups Art30 and Art60 presented, respectively, a reduction of 2% and 6% in intestinal area when compared to their control groups. No significant differences were observed in general neuronal density among the four groups (P>0.05). Group C60 presented a reduction of 14.4% and 10.9% in mean neuronal cell body area when compared to group C30 (P0.05). CONCLUSION: Arthritis does not promote quantitative or morphological damages in general myenteric population. However, studies in progress have revealed some significant alterations in myenteric neurons subpopulations (nitrergic and VIP-ergic neurons)

    O objetivo deste trabalho foi estabelecer valores de referência de alguns parâmetros fisiológicos (bioquímicos, hematológicos e urinários) de ratos provenientes do Biotério Central da Universidade Estadual de Maringá, Estado do Paraná. Foram utilizados ratos machos, normais e saudáveis da linhagem Wistar, com peso de 200 a 220 g. Foiavaliado o perfil hematológico (leucócitos: totais, mononucleares e polimorfonucleares) e a determinação bioquímica de vários constituintes plasmáticos (aspartato aminotransferase, alanina aminotransferase, glicose, creatinina, fosfatase alcalina, fosfatase ácida, uréia, colesterol total, colesterol HDL, triglicerídeos, teste de tolerância à glicose GTT) e urinários (glicose, creatinina). É imprescindível que cada laboratório estabeleça seu conjunto devalores de referência dos animais normais, de acordo com a dieta, a linhagem, o sexo e a idade de cada espécie utilizada. Assim, através deste trabalho, o pesquisador pode identificar desvios dos parâmetros, facilitando a escolha dos animais para o estudo.<br><br>The present work was carried out to establish reference values for some physiological parameters (biochemical, hematological and urinary) in rats proceeding from the Central Biotery at the State University of Maringá, State of Paraná. Normal male Wistar rats weighing 200-220 g were used. The hematological profile (total leukocytes, mononuclear and polymorphonuclear cells count), theclinic chemistry (aspartate aminotransferase, alanine aminotransferase, glucose, creatinine, alkaline phosphatase, acid phosphatase, urea, cholesterol, HDL-c, triglycerides, glucose tolerance) and urinary parameters (glucose and creatinine) were determined. Each laboratory should establish a database of reference values for their animals, according to diet, strain, sex and age foreach species used. Thus, in this work, a database is provided regarding reference values of some physiological parameters in rats for use in experimental investigations
