    Monoamine oxidase-A modulates apoptotic cell death induced by staurosporine in human neuroblastoma cells

    Monoamine oxidases (MAOs) are mitochondrial enzymes which control the levels of neurotransmitters in the brain and dietary amines in peripheral tissues via oxidative deamination. MAO has also been implicated in cell signalling. In this study, we describe the MAO-A isoform as functional in apoptosis induced by staurosporine (STS) in human dopaminergic neuroblastoma cells (SH-SY5Y). Increased levels of MAO-A activity were induced by STS, accompanied by increased MAO-A protein and activation of the initiator of the intrinsic pathway, caspase 9, and the executioner caspase 3. MAO-A mRNA levels were unaffected by STS, suggesting that changes in MAO-A protein are due to post-transcriptional events. Two unrelated MAO-A inhibitors reduced caspase activation. STS treatment resulted in sustained activation of the mitogen-activated protein kinase pathway enzymes extracellular regulated kinase, c-jun terminal kinase and p38, and depletion of the anti-apoptotic protein Bcl-2. These changes were significantly reversed by MAO inhibition. Production of reactive oxygen species was increased following STS exposure, which was blocked by both MAO inhibition and the antioxidant N-acetylcysteine. Therefore our data provide evidence that MAO-A, through its production of reactive oxygen species as a by-product of its catalytic activity on the mitochondrial surface, is recruited by the cell to enhance apoptotic signalling

    Cryogenic microstripline-on-Kapton microwave interconnects

    Simple broadband microwave interconnects are needed for increasing the size of focal plane heterodyne radiometer arrays. We have measured loss and cross-talk for arrays of microstrip transmission lines in flex circuit technology at 297 and 77 K, finding good performance to at least 20 GHz. The dielectric constant of Kapton substrates changes very little from 297 to 77 K, and the electrical loss drops. The small cross-sectional area of metal in a printed circuit structure yields overall thermal conductivities similar to stainless steel coaxial cable. Operationally, the main performance tradeoffs are between crosstalk and thermal conductivity.     Ketogenic Diet Alters Dopaminergic Activity in the Mouse Cortex [post-print]

    The present study was conducted to determine if the ketogenic diet altered basal levels of monoamineneurotransmitters in mice. The catecholamines dopamine (DA) and norephinephrine (NE) and the indolamine serotonin (5HT) were quantified postmortem in six different brain regions of adult mice fed a ketogenic diet for 3 weeks. The dopamine metabolites 3,4-dihydroxyphenylacetic acid (DOPAC) and homovanillic acid (HVA) and the serotonin metabolite 5-hydroxyindole acetic acid (5HIAA) were also measured. Tissue punches were collected bilaterally from the motor cortex, somatosensory cortex,nucleus accumbens, anterior caudate–putamen, posterior caudate–putamen and the midbrain. Dopaminergic activity, as measured by the dopamine metabolites to dopamine content ratio – ([DOPAC] + [HVA])/[DA] – was significantly increased in the motor and somatosensory cortex regions of mice fed the ketogenic diet when compared to those same areas in brains of mice fed a normal diet. These results indicate that the ketogenic diet alters the activity of the meso-cortical dopaminergic system, which may contribute to the diet\u27s therapeutic effect in reducing epileptic seizure activity

    ASMs and Operational Algorithmic Completeness of Lambda Calculus

    We show that lambda calculus is a computation model which can step by step simulate any sequential deterministic algorithm for any computable function over integers or words or any datatype. More formally, given an algorithm above a family of computable functions (taken as primitive tools, i.e., kind of oracle functions for the algorithm), for every constant K big enough, each computation step of the algorithm can be simulated by exactly K successive reductions in a natural extension of lambda calculus with constants for functions in the above considered family. The proof is based on a fixed point technique in lambda calculus and on Gurevich sequential Thesis which allows to identify sequential deterministic algorithms with Abstract State Machines. This extends to algorithms for partial computable functions in such a way that finite computations ending with exceptions are associated to finite reductions leading to terms with a particular very simple feature.Comment: 37 page

    An X-Ray Dip in the X-Ray Transient 4U 1630-47

    An x-ray dip was observed during a 1996 Rossi X-Ray Timing Explorer observation of the recurrent x-ray transient 4U 1630-47. During the dip, the 2-60 keV x-ray flux drops by a factor of about three, and, at the lowest point of the dip, the x-ray spectrum is considerably softer than at non-dip times. We find that the 4U 1630-47 dip is best explained by absorption of the inner part of an accretion disk, while the outer part of the disk is unaffected. The spectral evolution during the dip is adequately described by the variation of a single parameter, the column density obscuring the inner disk.Comment: 13 pages, 4 figures, Accepted for publication in Ap

    The Impact of Atmospheric Fluctuations on Degree-scale Imaging of the Cosmic Microwave Background

    Fluctuations in the brightness of the Earth's atmosphere originating from water vapor are an important source of noise for ground-based instruments attempting to measure anisotropy in the Cosmic Microwave Background. This paper presents a model for the atmospheric fluctuations and derives simple expressions to predict the contribution of the atmosphere to experimental measurements.     Temporal and spatial variations in freshwater 14C reservoir effects: Lake Myvatn, Northern Iceland

    Lake Mývatn is an interior highland lake in northern Iceland that forms a unique ecosystem of international scientific importance and is surrounded by a landscape rich in archaeological and palaeoenvironmental sites. A significant Freshwater 14C Reservoir Effect (FRE) has been identified in carbon from the lake at some Norse (c.870-1000 AD) archaeological sites in the wider region (Mývatnssveit). Previous AMS measurements indicated this FRE was ~1500-1900 14C years. Here we present the results of a study using stable isotope and 14C measurements to quantify the Mývatn FRE for both the Norse and modern periods. This work has identified a temporally variable FRE that is greatly in excess of previous assessments. New, paired samples of contemporaneous bone from terrestrial herbivores and omnivores (including humans) from Norse sites demonstrate at least some omnivore diets incorporated sufficient freshwater resources to result in a herbivore-omnivore age offset of up to 400 14C yrs. Modern samples of benthic detritus, aquatic plants, zooplankton, invertebrates and freshwater fish indicate an FRE in excess of 5000 14C yrs in some species.
