    VIVA, VLA Imaging of Virgo spirals in Atomic gas: I. The Atlas & The HI Properties

    We present the result of a new VLA HI Imaging survey of Virgo galaxies, VIVA (the VLA Imaging survey of Virgo galaxies in Atomic gas). The survey includes high resolution HI data of 53 carefully selected late type galaxies (48 spirals and 5 irregular systems). The goal is to study environmental effects on HI gas properties of cluster galaxies to understand which physical mechanisms affect galaxy evolution in different density regions, and to establish how far out the impact of the cluster reaches. As a dynamically young cluster, Virgo contains examples of galaxies experiencing a variety of environmental effects. Its nearness allows us to study each galaxy in great detail. We have selected Virgo galaxies with a range of star formation properties in low to high density regions (at the projected distance from M87, d_87=0.3-3.3 Mpc). Contrary to pr evious studies, more than half of the galaxies in the sample (~60%) are fainter than 12 mag in B_T. Overall, the selected galaxies represent the late type Virgo galaxies (S0/a to Sd/Irr) down to m_p<~14.6 fairly well in morphological type, systemic velocity, subcluster membership, HI mass and deficiency. In this paper (VIVA I: the atlas and the HI properties), we present HI maps and properties, and describe the HI morphology and kinematics of individual galaxies in detail (abbreviated).Comment: K band magnitudes for 6 galaxies in Table 3 have been corrected.     Virgo Galaxies with Long One-Sided HI Tails

    In a new HI imaging survey of Virgo galaxies (VIVA: VLA Imaging of Virgo galaxies in Atomic gas), we find 7 spiral galaxies with long HI tails. The morphology varies but all the tails are extended well beyond the optical radii on one side. These galaxies are found in intermediate-low density regions (0.6-1 Mpc in projection from M87). The tails are all pointing roughly away from M87, suggesting that these tails may have been created by a global cluster mechanism. While the tidal effects of the cluster potential are too small, a rough estimate suggests that simple ram-pressure stripping indeed could have formed the tails in all but two cases. At least three systems show HI truncation to within the stellar disk, providing evidence for a gas-gas interaction. Although most of these galaxies do not appear disturbed optically, some have close neighbors, suggesting that tidal interactions may have moved gas outwards making it more susceptible to the ICM ram-pressure or viscosity. Indeed, a simulation study of one of the tail galaxies, NGC 4654, suggests that the galaxy is most likely affected by the combined effect of a gravitational interaction and ram-pressure stripping. We conclude that these one-sided HI tail galaxies have recently arrived in the cluster, falling in on highly radial orbits. It appears that galaxies begin to lose their gas already at intermediate distances from the cluster center through ram-pressure or turbulent viscous stripping and tidal interactions with neighbours, or a combination of both.Comment: 4 pages, 3 figures (including 1 plate), accepted for accepted for publication in ApJ Letter (vol. 659, L115), a version with full resolution Plate 1 is available from http://www.astro.umass.edu/~achung/astro-ph/viva_tail.pd

    Event Signaling Within Higher Performance Network Systems

    The afterburner ATM link Adapter has allowed us to evaluate three event-signaling schemes: polling, traditional interrupts and the clocked interrupts first investigated in our operating system work in AURORA. The schemes are evaluated in the context of a single-copy TCP/IP stack. The experimental results indicate that clocked interrupts can provide throughput comparable with traditional interrupts for dedicated machines (up to over 144 Mbps, the highest TCP/IP/ATM throughput reported), and better performance when the machines are loaded with an artificial workload. Polling, implemented to be used with an unmodified netperf measurement tool, was competitive for small TCP/IP socket buffer sizes (¡32KB). We concluded that clocked interrupts may be preferable for applications requiring high throughput on systems with heavy processing workloads, such as servers

    Population Structure of the Blue Crab Callinectes sapidus in the Maryland Coastal Bays

    The population structure of the blue crab Callinectes sapidus was examined in the Maryland Coastal Bays (MCB) from 2014 to 2016. Crabs were sampled from April to December of each year. Size–frequency distributions showed a strong seasonal cycle, with small crabs being abundant in April, increasing in size through September, with adult crabs observed in the highest proportions from August through October of each year. A subsample of crabs was assayed for hemolymph ecdysone concentrations to examine molting patterns in field-collected blue crabs. Molting was observed throughout the sampling season, peaking in April for immature crabs, with lows in all size classes occurring in October. The mean size at maturity (L50) for females collected in this study was 116mmcarapace width (CW), which is comparable to that reported for the lower Chesapeake Bay (CB) and suggests crabs in the MCB are not significantly smaller as previously thought; however, large crabs (\u3e127 mmCW) appear to make up a smaller proportion of the total population in the MCB than in CB. Ovigerous females were observed at two distinct locations depending on the season, with 13/15 (86.7%) in southern Chincoteague Bay in April and May and 24/41 (58.5%) nearest to the Ocean City Inlet in July and August, indicating two potentially distinct spawning grounds and periods. This work suggests that blue crab reproductive success and general population trends are similar across both systems, with fishing pressure or disease in the MCB potentially explaining the low abundance of adult male crabs

    Structural and Functional Analysis of a β2-Adrenergic Receptor Complex with GRK5.

    The phosphorylation of agonist-occupied G-protein-coupled receptors (GPCRs) by GPCR kinases (GRKs) functions to turn off G-protein signaling and turn on arrestin-mediated signaling. While a structural understanding of GPCR/G-protein and GPCR/arrestin complexes has emerged in recent years, the molecular architecture of a GPCR/GRK complex remains poorly defined. We used a comprehensive integrated approach of cross-linking, hydrogen-deuterium exchange mass spectrometry (MS), electron microscopy, mutagenesis, molecular dynamics simulations, and computational docking to analyze GRK5 interaction with the β2-adrenergic receptor (β2AR). These studies revealed a dynamic mechanism of complex formation that involves large conformational changes in the GRK5 RH/catalytic domain interface upon receptor binding. These changes facilitate contacts between intracellular loops 2 and 3 and the C terminus of the β2AR with the GRK5 RH bundle subdomain, membrane-binding surface, and kinase catalytic cleft, respectively. These studies significantly contribute to our understanding of the mechanism by which GRKs regulate the function of activated GPCRs. PAPERCLIP

Although the mtPTP is a potential therapeutic target for many human pathologies, its potential as a drug target is currently unrealized. Herein we describe an optimization effort initiated around hit 1, 5-(3-hydroxyphenyl)-N-(3,4,5-trimethoxyphenyl)isoxazole-3-carboxamide, which was found to possess promising inhibitory activity against mitochondrial swelling (EC50 100 µm). This enabled the construction of a series of picomolar mtPTP inhibitors that also potently increase the calcium retention capacity of the mitochondria. Finally, the therapeutic potential and in vivo efficacy of one of the most potent analogues, N-(3-chloro-2-methylphenyl)-5-(4-fluoro-3-hydroxyphenyl)isoxazole-3-carboxamide (60), was validated in a biologically relevant zebrafish model of collagen VI congenital muscular dystrophies

    Increased serum kallistatin levels in type 1 diabetes patients with vascular complications

    BACKGROUND: Kallistatin, a serpin widely produced throughout the body, has vasodilatory, anti-angiogenic, anti-oxidant, and anti-inflammatory effects. Effects of diabetes and its vascular complications on serum kallistatin levels are unknown. METHODS: Serum kallistatin was quantified by ELISA in a cross-sectional study of 116 Type 1 diabetic patients (including 50 with and 66 without complications) and 29 non-diabetic controls, and related to clinical status and measures of oxidative stress and inflammation. RESULTS: Kallistatin levels (mean(SD)) were increased in diabetic vs. control subjects (12.6(4.2) vs. 10.3(2.8) μg/ml, p = 0.007), and differed between diabetic patients with complications (13.4(4.9) μg/ml), complication-free patients (12.1(3.7) μg/ml), and controls; ANOVA, p = 0.007. Levels were higher in diabetic patients with complications vs. controls, p = 0.01, but did not differ between complication-free diabetic patients and controls, p > 0.05. On univariate analyses, in diabetes, kallistatin correlated with renal dysfunction (cystatin C, r = 0.28, p = 0.004; urinary albumin/creatinine, r = 0.34, p = 0.001; serum creatinine, r = 0.23, p = 0.01; serum urea, r = 0.33, p = 0.001; GFR, r = -0.25, p = 0.009), total cholesterol (r = 0.28, p = 0.004); LDL-cholesterol (r = 0.21, p = 0.03); gamma-glutamyltransferase (GGT) (r = 0.27, p = 0.04), and small artery elasticity, r = -0.23, p = 0.02, but not with HbA1c, other lipids, oxidative stress or inflammation. In diabetes, geometric mean (95%CI) kallistatin levels adjusted for covariates, including renal dysfunction, were higher in those with vs. without hypertension (13.6 (12.3-14.9) vs. 11.8 (10.5-13.0) μg/ml, p = 0.03). Statistically independent determinants of kallistatin levels in diabetes were age, serum urea, total cholesterol, SAE and GGT, adjusted r2 = 0.24, p < 0.00001. CONCLUSIONS: Serum kallistatin levels are increased in Type 1 diabetic patients with microvascular complications and with hypertension, and correlate with renal and vascular dysfunction
