    Identification des dermatophytes en cultures sur la base de leurs critères macroscopiques et microscopiques

    Contexte : Les dermatophytes sont des champignons filamenteux parasites spécialisés qui dégradent les tissus kératinisés. Ils sont responsables de la plupart des mycoses de la peau, du cuir chevelu et des cheveux, et des ongles. Le choix du traitement des dermatophytoses dépend des symptômes et du dermatophyte incriminé parmi une quinzaine d'espèces possibles. L'identification des dermatophytes se fait en général sur la base des caractères macroscopiques et microscopiques des cultures. L'identification est parfois difficile ou reste incertaine car il peut y avoir des variations d'un isolat à l'autre au sein d'une même espèce. Cependant, les espèces sont facilement identifiées sur la base de séquences d'ADN. En pratique, des séquences d'ADN ribosomique suffisamment polymorphes sont le plus souvent utilisées pour discriminer les espèces de dermatophytes. Des méthodes spécialisées et sophistiquées telles que les séquences d'ADN et la spectrométrie de masse sont de plus en plus proposées dans la littérature pour identifier les dermatophytes. Toutefois, ces méthodes ne peuvent pas être utilisées directement par un médecin dans un cabinet médical. C'est pourquoi des méthodes plus simples basées sur l'observation de caractères phénotypiques des champignons en culture ne devraient pas être abandonnées. Objectif : Etablir une clé d'identification dichotomique se basant sur des caractères macroscopiques et microscopiques permettant une identification fiable du dermatophyte par la culture. Des clés d'identification des espèces seront élaborées et testées pour leur validation en parallèle avec leur identification par des méthodes de Biologie Moléculaire. Créer un outil simple qui pourra être utilisé au laboratoire par des médecins ou des biologistes non spécialisés en mycologie pour identifier les dermatophytes sans avoir recours à une technologie sophistiquée. Méthodes : Inventaire des espèces isolées de 2001 à 2012 au laboratoire de dermatologie du CHUV. Inventaire des caractères phénotypiques permettant de caractériser chaque espèce. Création d'un système dichotomique sur la base des caractères phénotypiques pour séparer et identifier les espèces (clé d'identification des espèces). Résultats attendus : Les résultats attendus sont définis au niveau des objectifs. L'outil doit être accessible pour des personnes inexpérimentées qui pourront alors identifier les dermatophytes. Plus-value : Les dermatophytoses sont fréquemment diagnostiquées. Cet outil est destiné à tous les dermatologues installés et au personnel de laboratoire qui ne sont pas nécessairement spécialisés en la matière

    COORDINATION: HOW ELITE SWIMMERS DIFFER FROM SUB-ELITE

    In this talk the inter-limb coordination of elite and sub-elite swimmers are discussed with a view to informing coaches of the similarities and differences between these groups. In the past it has been reported that the inter-limb coordination should show an opposition mode, i.e. a propulsive continuity between the propulsion of one limb and those of the other limb, in order to minimize the intra-cyclic velocity variations. However, the research of our centre of research highlighted the fact that the inter-limb coordination mode adopted by the swimmers corresponds to three types of constraint defined by Newell (1986): organismic, task and environmental constraint. The skill level of the swimmers, the specialty, the gender, the handedness and the breathing laterality act as organismic constraints; the imposed race pace, the stroke frequency, the number of strokes, the breathing frequency and pattern could be consider as task constraints while the active drag and his correspondent velocity relate to the environmental constraints. Inter-limb coordination was found to vary from catch or glide coordination mode to superposition mode, showing that the opposition mode is only the best “theoretical” mode and the glide mode is not a technical mistake. Therefore it is advised for coaches to don’t consider an ideal coordination mode in the absolute but to teach the swimmers in different ways when developing coordination. This talk presents new information based on recent scientific research conducted at the CETAPS. The variables of interest were: average swim speed, stroke length, stroke frequency, intra-cyclic velocity variations, breathing laterality, relative duration of arm and leg stroke phases, time gap between propulsive actions assessed by total time gap (TTG) in the simultaneous strokes and by index of coordination (IdC) in the alternate strokes. Interesting findings emerged that have implications for the both elite and sub-elite swimmers should be coached

    Solar interacting protons versus interplanetary protons in the core plus halo model of diffusive shock acceleration and stochastic re-acceleration

    With the first observations of solar γ-rays from the decay of pions, the relationship of protons producing ground level enhancements (GLEs) on the Earth to those of similar energies producing the γ-rays on the Sun has been debated. These two populations may be either independent and simply coincident in large flares, or they may be, in fact, the same population stemming from a single accelerating agent and jointly distributed at the Sun and also in space. Assuming the latter, we model a scenario in which particles are accelerated near the Sun in a shock wave with a fraction transported back to the solar surface to radiate, while the remainder is detected at Earth in the form of a GLE. Interplanetary ions versus ions interacting at the Sun are studied for a spherical shock wave propagating in a radial magnetic field through a highly turbulent radial ray (the acceleration core) and surrounding weakly turbulent sector in which the accelerated particles can propagate toward or away from the Sun. The model presented here accounts for both the first-order Fermi acceleration at the shock front and the second-order, stochastic re-acceleration by the turbulence enhanced behind the shock. We find that the re-acceleration is important in generating the γ-radiation and we also find that up to 10% of the particle population can find its way to the Sun as compared to particles escaping to the interplanetary space

    Digital histologic analysis reveals morphometric patterns of age-related involution in breast epithelium and stroma

    Complete age-related regression of mammary epithelium, often termed post-menopausal involution, is associated with decreased breast cancer risk. However, most studies have qualitatively assessed involution. We quantitatively analyzed epithelium, stroma, and adipose tissue from histologically normal breast tissue of 454 patients in the Normal Breast Study (NBS). High-resolution digital images of normal breast Hematoxylin & Eosin stained slides were partitioned into epithelium, adipose tissue, and non-fatty stroma. Percentage area and nuclei per unit area (nuclear density) were calculated for each component. Quantitative data were evaluated in association with age using linear regression and cubic spline models Stromal area decreased (p=0.0002) and adipose tissue area increased (p<0.0001), with an approximate 0.7% change in area for each component, until age 55 when these area measures reached a steady state. While epithelial area did not show linear changes with age, epithelial nuclear density decreased linearly beginning in the third decade of life. No significant age-related trends were observed for stromal or adipose nuclear density. Digital image analysis offers a high-throughput method for quantitatively measuring tissue morphometry and for objectively assessing age-related changes in adipose tissue, stroma, and epithelium. Epithelial nuclear density is a quantitative measure of age-related breast involution that begins to decline in the early premenopausal period

    12^{12}C/13^{13}C ratio in planetary nebulae from the IUE archives

    We investigated the abundance ratio of 12^{12}C/13^{13}C in planetary nebulae by examining emission lines arising from \ion{C}{3} 2s2p ^3P_{2,1,0} \to 2s^2 ^1S_0. Spectra were retrieved from the International Ultraviolet Explorer archives, and multiple spectra of the same object were coadded to achieve improved signal-to-noise. The 13^{13}C hyperfine structure line at 1909.6 \AA was detected in NGC 2440. The 12^{12}C/13^{13}C ratio was found to be 4.4±\sim4.4\pm1.2. In all other objects, we provide an upper limit for the flux of the 1910 \AA line. For 23 of these sources, a lower limit for the 12^{12}C/13^{13}C ratio was established. The impact on our current understanding of stellar evolution is discussed. The resulting high signal-to-noise \ion{C}{3} spectrum helps constrain the atomic physics of the line formation process. Some objects have the measured 1907/1909 flux ratio outside the low-electron density theoretical limit for 12^{12}C. A mixture of 13^{13}C with 12^{12}C helps to close the gap somewhat. Nevertheless, some observed 1907/1909 flux ratios still appear too high to conform to the presently predicted limits. It is shown that this limit, as well as the 1910/1909 flux ratio, are predominantly influenced by using the standard partitioning among the collision strengths for the multiplet 1S0^1S_0--3PJ^3P_J according to the statistical weights. A detailed calculation for the fine structure collision strengths between these individual levels would be valuable.Comment: ApJ accepted: 19 pages, 3 Figures, 2 Table

    Synthesis and Antimalarial Activity of 11 Dispiro-1,2,4,5-tetraoxane Analogues of WR 148999. 7,8,15,16-Tetraoxadispiro[5.2.5.2]hexadecanes Substituted at the 1 and 10 Positions with Unsaturated and Polar Functional Groups

    Eleven novel dispiro-1,2,4,5-tetraoxanes 3 bearing unsaturated and polar functional groups were designed to enhance the oral antimalarial activity of the prototype tetraoxane 2 (WR 148999). With the exception of 3g and 3h, tetraoxanes 3 were available via the peroxidation of corresponding cyclohexanone derivatives in H2SO4/CH3CN. Tetraoxanes 3g and 3h were prepared by hydrolysis of ester tetraoxanes 3e and 3i, respectively. Five of the 11 tetraoxanes were inactive, but six tetraoxanes had IC50 values of 6-26 nM against the K1 and NF54 strains of Plasmodium falciparum compared to corresponding IC50 values of 28 and 39 nM for 2, and 10 and 12 nM for artemisinin (1). Ester tetraoxane 3e was the most active in vitro, some 2-fold more potent than 1. However, none of the six tetraoxanes active in vitro were as effective as either 1 or 2 in vivo; at single doses of 100 mg/kg most possessed little to no vivo activity in mice infected with Plasmodium berghei. Unsaturated tetraoxane 3a was uniquely more active when administered per os (po) than subcutan (sc). For this series of tetraoxanes, the discrepancy between vitro and vivo activities underscores the limitations of conclusions drawn solely from in vitro antimalarial data and illustrates a practical benefit of complementary single-dose in vivo antimalarial screens

    Heliospheric Transport of Neutron-Decay Protons

    We report on new simulations of the transport of energetic protons originating from the decay of energetic neutrons produced in solar flares. Because the neutrons are fast-moving but insensitive to the solar wind magnetic field, the decay protons are produced over a wide region of space, and they should be detectable by current instruments over a broad range of longitudes for many hours after a sufficiently large gamma-ray flare. Spacecraft closer to the Sun are expected to see orders-of magnitude higher intensities than those at the Earth-Sun distance. The current solar cycle should present an excellent opportunity to observe neutron-decay protons with multiple spacecraft over different heliographic longitudes and distances from the Sun.Comment: 12 pages, 4 figures, to be published in special issue of Solar Physic

The selection of appropriate services at the right moment in order to compose meaningful applications is however a real issue. In this paper, we propose a FCA-based solution to this problem.We have integrated FCA algorithms in our pervasive gateways and adapted them in order to allow efficient runtime selection of heterogeneous and dynamic services.
