Optimization of RAPD analysis for identification of olive varieties
Entre las técnicas moleculares, RAPD-PCR es una de las más rápidas y operativamente simple para la caracterización de cultivares. Sin embargo, la confiabilidad de
sus resultados radica, en gran parte, en la optimización previa de variables que pueden afectar los patrones de amplificación. Se investigó la repetibilidad de patrones RAPD de olivo bajo diferentes condiciones experimentales. Entre ellas, la pureza del ADN, el tipo de Taq ADN-polimerasa, las concentraciones de Mg+2 y dNTPs, el uso de tejidos atacados por patógenos y el uso de diferentes termocicladores, modificaron los patrones de bandas. Por el contrario, pequeñas modificaciones de la temperatura de apareamiento de cebadores, la concentración del ADN molde y la edad de los tejidos vegetales de los cuales se aisló ADN, no afectaron los patrones amplificados.RAPD-PCR, although a relatively fast and simple technique for cultivar characterization, is influenced by several parameters that need to be optimized prior using it as a routine identification methodology. The repeatability of olive RAPD patterns was investigated under different PCR conditions. Among the conditions tested the quality of the DNA, the Taq DNA polymerase source, changes in Mg2+ and dNTPs concentration, pathogen infestation of olive tissues and different thermocyclers could alter the RAPD patterns. In contrast, small changes in the temperature of annealing, the DNA concentration and the age of tissues from which DNA is isolated did not affect the amplification patterns.Fil: Cavagnaro, Pablo.

Evaluation of genotypic homogeneity in olive nurseries using molecular markers
La difusión de variedades tradicionales
de olivo (Olea europea L.) y la obtención
de nuevas variantes fenotípicas han generado
confusión en la correcta identificación
y denominación de algunas de las aprox.
2 000 variedades conocidas a nivel mundial.
En la Argentina, en los últimos años se ha
triplicado la superficie implantada con olivo
principalmente a partir de plantas de viveros
locales e importadas. Con el fin de evaluar
la homogeneidad genotípica del material
comercializado en Mendoza, se probaron
7 marcadores RAPD altamente reproducibles
en muestras de 5 viveros, correspondientes
a 5 variedades de olivo. Los marcadores
RAPD fueron previamente desarrollados
para caracterizar las variedades del INTA
Junín. Arbequina y Arauco fueron los materiales
más homogéneos. En ambas variedades,
todos los individuos compartieron el
100 % de los marcadores utilizados. Los lotes
de muestras de Empeltre, Farga y Aloreña
no fueron genotípicamente homogéneos, observándose
2-3 patrones diferentes por lote.
Todas las variedades ensayadas -en alguna
de sus muestras- tuvieron diferencias con su
respectiva variedad del INTA Junín. Arbequina
y Arauco también compartieron el 100 % de
marcadores entre sí, no pudiéndose separar
ambos grupos.Approx. 2 000 olive (Olea europea L.)
varieties are known in the world. Due to the
commerce, promotion of old varieties and the
development of new phenotypic variants, a
situation of uncertainty exists regarding the
correct identification and denomination of
some cultivars. In the last years the Argentine
olive-cultivated area has been triple folded.
Most of the plant materials came from local
nurseries and some from importation. In order
to test the genotypic homogeneity of the
materials commercialized by the local
nurseries, plant samples from 5 of them were
tested using 7 reproducible RAPD markers.
These markers were previously developed
for characterizing cultivars from the
germplasm bank of INTA Junín. Only
samples from cultivars Arbequina and
Arauco showed complete homogeneity
regarding the markers assayed. Samples
from cultivars Empeltre, Farga and Aloreña
were not genotypically homogeneous,
showing from 2 to 3 different RAPD patterns
per lot of samples. All the varieties tested
from the nurseries, showed differences with
their respective varieties from INTA Junín,
for at least 1 of the samples. Arbequina and
Arauco also shared 100 % of the markers
between them. Therefore it was not possible
to distinguish one from the other by the use

Desarrollo y caracterización de marcadores moleculares SSR para Trichloris crinita usando secuencias de gramíneas filogenéticamente cercanas
Trichloris crinita is among the most important native forage grasses in arid regions
of America. Despite its importance, molecular resources and sequence data are
extremely scarce in this species. In the present study, SSR markers were developed
using available DNA sequences from grass taxa phylogenetically-related to Trichloris
(Eleusine coracana, Cynodon dactylon and ‘Cynodon dactylon x Cynodon transvaalensis’).
Marker transferability was evaluated in a panel of eight T. crinita accessions and five
closely-related species. Of the 105 SSR primer pairs evaluated, 16 amplified products
of expected size in T. crinita, whereas transferability to other grass species ranged from
12 (in Chloris castilloniana) to 28 SSRs (in Eleusine coracana). Six of the 16 SSR markers
successfully transferred to T. crinita (37.5%) were polymorphic, and were further used
to assess genetic diversity in eight T. crinita accessions. The analysis revealed a total of
23 SSR alleles (3.83 alleles/locus), allowing the discrimination of all T. crinita accessions,
with pair-wise genetic similarities ranging from 0.35 to 0.81 (Jaccard coefficient). Mean
(and range) values for observed (Ho) and expected heterozygosity (He) were 0.53
(0.0-1.0) and 0.63 (0.48-0.79), respectively.Trichloris crinita es una importante gramínea forrajera, nativa de regiones áridas del

Effect of processing and cooking conditions on onion (Allium cepa L.) induced antiplatelet activity and thiosulfinate content
Allium vegetables serve as sources of antiplatelet agents that may contribute to the prevention of cardiovascular disease. However, onion and garlic, the major Allium species, are usually cooked before consumption. Here, we examined the effect of cooking on onion in vitro antiplatelet activity (IVAA). Two different cooking systems (convection oven and microwaves) and several time−temperature variables were tested on whole bulbs, quarters of bulbs, and completely crushed bulbs, monitoring the degradation of sulfur antiplatelet compounds (e.g., thiosulfinates) by analysis of pyruvate levels. Although heating was, in general, detrimental for onion IVAA, the extent of this effect varied greatly, from unaffected antiplatelet activity (AA) (i.e., similar to raw onion) to a complete lost of activity, depending upon the manner in which onions were prepared prior to heating, the cooking method, and the intensity of the heat treatment. “Whole”, “quarters”, and “crushed” onions lost their IVAA after 30, 20, and 10 min of oven heating, respectively. The longer retainment of AA in intact bulbs was attributed to a later alliinase inactivation. Proaggregatory effects observed in samples subjected to the most intense oven and microwave heat treatments suggest that extensively cooked onions may stimulate rather than inhibit platelet aggregation. High Temperature Alters Anthocyanin Concentration and Composition in Grape Berries of Malbec, Merlot, and Pinot Noir in a Cultivar-Dependent Manner
Climate is determinant for grapevine geographical distribution, berry attributes, and wine quality. Due to climate change, a 2–4 °C increase in mean diurnal temperature is predicted by the end of the century for the most important Argentine viticulture region. We hypothesize that such temperature increase will affect color intensity and other quality attributes of red grapes and wines. The present study investigated the effect of high temperature (HT) on anthocyanin concentration and composition, pH, and resveratrol and solids content in berries of three major wine-producing varieties during fruit ripening in two seasons. To this end, a structure that increased mean diurnal temperature by 1.5–2.0 °C at berry sites, compared to Control (C) plants grown without such structure, was implemented in field grown vineyards of Malbec, Merlot, and Pinot Noir. Results revealed a cultivar-dependent response to HT conditions, with Malbec and Pinot Noir berries exhibiting significant decreases in total anthocyanin concentration (TAC) at veraison and harvest, respectively, while Merlot maintained an unaffected pigment content under HT. The decrease in TAC was associated with reduced levels of delphinidin, cyanidin, petunidin, peonidin, and malvidin glycosides, and increased ratios of acylated (AA)/non-acylated anthocyanins (NAA), suggesting pigment acylation as a possible stress-response mechanism for attenuating HT negative effects. Under HT, Pinot Noir, which does not produce AA, was the only cultivar with lower TAC at harvest (p < 0.05). pH, resveratrol, and solids content were not affected by HT. Our results predict high, medium, and low plasticity with regard to color quality attributes for Malbec, Merlot, and Pinot Noir, respectively, in the context of climate change.EEA La ConsultaFil: de Rosas, María Inés. MOLECULAR CHARACTERIZATION OF GRAPE VARIETIES ( Vitis vinifera L.) OF ENOLOGICAL VALUE USING MICROSATELLITES MARKERS
Siete variedades de vid empleadas en Argentina para la elaboración de vinos de alta gama fueron caracterizadas molecularmente a través del empleo de microsatélites. Seis de los 8 pares de cebadores usados amplificaron
patrones de bandas reproducibles. El número de alelos detectados por locus varió entre 5 y 10, con un total de 42 alelos, registrándose desde 1 a 7 alelos únicos. El número de genotipos microsatélites encontrados para cada locus osciló entre 3 y 7, con un total de 28. Las variedades Tempranillo y Chenin
mostraron 6 y 5 genotipos «únicos», respectivamente, con los 6 loci analizados. La heterocigosidad observada varió entre 42,9 y 100 %, mientras que la heterocigosidad esperada osciló entre 64,3 y 87,8 %. El locus
más informativo fue VrZAG79 (con un contenido de información polimórfica de 84,9 % y un número de alelos efectivos de 8,17) mientras que el menos informativo fue VrZAG62. La probabilidad acumulada de obtener genotipos idénticos fue de 7,68 x 10-05 indicando que, mediante el uso combinado de
estos 6 cebadores microsatélites se pueden discriminar todas las variedades ensayadas, con alto grado de certeza. El análisis de agrupamiento por el método UPGMA separó claramente las variedades francesas (Merlot, Pinot Noir y Cabernet Sauvignon) y Syrah de los cepajes Tempranillo y Bonarda. Esta técnica podría ofrecerse como servicio a
viveristas y productores vitícolas interesados en garantizar la identidad genética de sus materiales comercializados.SSR markers characterized seven grapevine varieties used for high quality wines in Argentina. Six of the 8 primers used gave reproducible band profiles. The number of alleles per locus varied from 5 to 10, with a total of 42
and 1 to 7 unique alleles. Three to 7 SSR genotypes per locus were found, with a total of 28. Tempranillo and Chenin showed 6 and 5 unique alleles, respectively, taking in account all the primers assessed. The observed
heterozygosity varied between 42.9 and 100 %, while the expected heterozygosity ranged from 64.3 to 87.8 %. VrZAG79 was the most
informative locus (PIC 84.9 % and ne 8.17), while VrZAG62 was the least informative. Combining the use of 6 primers, the accumulative probability of missidentifying two random different genotypes as the same, was very low:
7.68 x 10-05. The UPGMA cluster analysis separated the French varieties (Merlot, Pinot Noir, Cabernet Sauvignon), and Syrah from Tempranillo and Bonarda. This technique can provide a service to grapevine nurseries and

Characterization of the purple carrot germplasm for their root anthocyanin composition and selection for carrot fresh consumption and food dies
Entre los principales objetivos del programa nacional de mejoramiento de zanahoria, con sede en el INTA La Consulta, se encuentran la diversificación varietal y el aumento del valor funcional (i.e., su valor como alimento saludable). En este contexto, las zanahorias moradas, originarias de Turquía y medio oriente, revisten gran interés para estos fines, ya que algunos genotipos suelen acumular gran cantidad antocianos en sus raíces y en otros órganos y tejidos de la planta. Estos pigmentos poseen reconocidas propiedades benéficas para la salud y son utilizados en la industria alimenticia como colorantes naturales. Químicamente, pueden presentar distintos grados de glicosilación y acilación, afectando estas estructuras sus propiedades físico-químicas y biológicas. Antocianos acilados (AA) poseen mayor estabilidad química pero son menos biodisponibles que los antocianos no acilados (ANA). En este contexto, es de interés la obtención de variedades con alto contenido de pigmento y con diferente proporción relativa de AA y ANA, según sea su destino el consumo en fresco o la extracción de colorante. Para ello, es fundamental previamente, caracterizar el germoplasma de zanahorias moradas que se usara como Este proyecto propone caracterizar el germoplasma de zanahorias moradas según sus perfiles de antocianos y seleccionar genotipos con alto contenido de antocianos acilados (AA) y genotipos con alto contenido de antocianos no-acilados (ANA). Además, propone investigar como varia el contenido y perfil de estos compuesto en diferentes tejidos de la raíz (floema y xilema) y en hojas. Por último, se investigará también como varia la composición de antocianos en raíz en función de las condiciones agroclimáticas de cultivo. Los resultados esperados de este proyecto permitirán identificar genotipos valiosos y condiciones óptimas de cultivo para la producción de zanahorias moradas para consumo en fresco y para la producción de colorantes alimenticios naturales.Cultivar diversification and increasing carrot functional value are among the main objectives of the Argentine carrot breeding program at INTA La Consulta (Mendoza). In this context, purple carrots, which originated in the Middle East, are of interest, since some genotypes can accumulate large amounts of anthocyanins in their roots and other plant organs. These pigments are well-known for their health-promoting properties and they are also used as dyes in the food industry. Anthocyanin chemical structure affects their chemical stability and biological properties. Acylated anthocyanins (AA) present higher chemical stability and therefore are more suitable as food dyes than non-acylated anthocyanins (NAA). However, the latter (NAA) have much higher bioavailability, and therefore are of greater for fresh-carrot consumer, from a health-promoting perspective. In this context, it is of interest for the carrot breeding program to develop purple carrot cultivars with high AA, for the production of natural food dyes, and cultivars with high NAA, for fresh consumption. To this end, this project will characterize the purple carrot germplasm for their root anthocyanin content and composition, with the goal of identifying and selecting valuable genotypes suitable for production purposes, fresh consumption and natural food dyes. Variation in anthocyanin composition between the root phloem and xylem tissues and leaves will also be examined. In addition, environmental influence due to agroclimatic conditions on this trait will be investigated. The expected results from this project will allow the identification of valuable materials and optimal growing conditions for the production of purple carrots for fresh consumption and food dyes
Regional climate variability impacts on the annual grape yield in Mendoza, Argentina
The Mendoza Province is the major Argentinian vitivinicultural region and its grape production is fundamental for the national vintage. 1979-2009 climate-annual grape yield relationships are analyzed. Total grape yield is shown to depend significantly on regional ´summer´ (October through March) precipitation. Precipitation negatively impacts yields through plant disease and damage/destruction by hail. At interannual scales, ´summer´ regional precipitation variability can explains 25% of the yield variance. Summer precipitation modulates yield with a 6-8 years period: wet (dry) summers can be associated with larger (smaller) grape damage/loss probability during the summer preceding the vintage, as well as lower (higher) grape yields in the subsequent annual campaign due to bud damage. When monthly mean precipitation in Mendoza Observatory is considered wetter Novembers/Decembers can lead to lower yields. Hail during the summer of the previous harvest and during December could lower yields. Winter, late spring and early summer mean maximum temperatures can impact current and subsequent annual yields: warmer (colder) months are linked to enhanced (decreased) yields. These relationships can be associated with circulation and SST conditions in the equatorial and extratropical Pacific basin and southern South America: SSTs within the southeastern South Pacific are related to western equatorial Pacific SSTs and convection, which modify circulation and water vapor transport over southern South America. Statistical multilinear modeling shows that the observed relationships between yield, precipitation and temperature can explain at least 60% of the observed interannual yield variability. A gene-derived SNP-based high resolution linkage map of carrot including the location of QTL conditioning root and leaf anthocyanin pigmentation
Purple carrots accumulate large quantities of anthocyanins in their roots and leaves. These flavonoid pigments possess antioxidant activity and are implicated in providing health benefits. Informative, saturated linkage maps associated with well characterized populations segregating for anthocyanin pigmentation have not been developed. To investigate the genetic architecture conditioning anthocyanin pigmentation we scored root color visually, quantified root anthocyanin pigments by high performance liquid chromatography in segregating F2, F3 and F4 generations of a mapping population, mapped quantitative trait loci (QTL) onto a dense gene-derived single nucleotide polymorphism (SNP)-based linkage map, and performed comparative trait mapping with two unrelated populations. Results: Root pigmentation, scored visually as presence or absence of purple coloration, segregated in a pattern consistent with a two gene model in an F2, and progeny testing of F3-F4 families confirmed the proposed genetic model. Purple petiole pigmentation was conditioned by a single dominant gene that co-segregates with one of the genes conditioning root pigmentation. Root total pigment estimate (RTPE) was scored as the percentage of the root with purple color. Conclusions: This study generated the first high resolution gene-derived SNP-based linkage map in the Apiaceae. Two regions of chromosome 3 with co-localized QTL for all anthocyanin pigments and for RTPE, largely condition anthocyanin accumulation in carrot roots and leaves.
