    The crystal structure of Canavalia brasiliensis lectin suggests a correlation between its quaternary conformation and its distinct biological properties from Concanavalin A

    AbstractCanavalia brasiliensis lectin was isolated from the seeds of a Brazilian autochthonous Leguminosae plant. Despite extensive amino acid sequence similarity with Concanavalin A, C. brasiliensis lectin exerts in vitro and in vivo cellular effects that are markedly different from those displayed by Concanavalin A. We have solved the crystal structure of the C. brasiliensis lectin at 3.0 Å resolution. The three-dimensional structure of the lectin monomer can be superimposed onto that of Concanavalin A with a root-mean-square deviation for all Cα atoms of 0.65 Å. However, this parameter is 0.84 and 1.62 Å when the C. brasiliensis lectin dimer and tetramer, respectively, are compared with the same structures of Concanavalin A. We suggest that these differences in quaternary structure may account for the different biological properties of these two highly related Leguminosae lectins.© 1997 Federation of European Biochemical Societies

    Satisfiability Modulo Transcendental Functions via Incremental Linearization

    In this paper we present an abstraction-refinement approach to Satisfiability Modulo the theory of transcendental functions, such as exponentiation and trigonometric functions. The transcendental functions are represented as uninterpreted in the abstract space, which is described in terms of the combined theory of linear arithmetic on the rationals with uninterpreted functions, and are incrementally axiomatized by means of upper- and lower-bounding piecewise-linear functions. Suitable numerical techniques are used to ensure that the abstractions of the transcendental functions are sound even in presence of irrationals. Our experimental evaluation on benchmarks from verification and mathematics demonstrates the potential of our approach, showing that it compares favorably with delta-satisfiability /interval propagation and methods based on theorem proving

    Leguminous lectins as tools for studying the role of sugar residues in leukocyte recruitment.

    The natural physiological ligands for selectins are oligosaccharides found in glycoprotein or glycolipid molecules in cell membranes. In order to study the role of sugar residues in the in vivo lectin anti-inflammatory effect, we tested three leguminous lectins with different carbohydrate binding affinities in the peritonitis and paw oedema models induced by carrageenin in rats. L. sericeus lectin was more anti-inflammatory than D. virgata lectin, the effects being reversed by their specific binding sugars (N-acetylglucosamine and alpha-methylmannoside, respectively). However, V. macrocarpa, a galactose-specific lectin, was not anti-inflammatory. The proposed anti-inflammatory activity of lectins could be due to a blockage of neutrophil-selectin carbohydrate ligands. Thus, according to the present data, we suggest an important role for N-acetylglucosamine residue as the major ligand for selectins on rat neutrophil membranes

    Efeito alelopático do extrato bruto de Brachiaria decumbens na germinação e no vigor de sementes e de plântulas de Brachiaria brizantha.

    Objetivou-se identificar os possíveis efeitos alelopáticos produzidos pelo extrato bruto de um acesso da coleção de Brachiaria spp da Embrapa Gado de Corte, denominado Brachiaria decumbens BRA001996 na germinação, no vigor de sementes e no vigor de plântulas de três cultivares de B. brizantha (cvs. Marandu, BRS Piatã e Xaraés). As sementes das cultivares foram semeadas em papel germitest, colocadas em gerbox e incubadas em câmara climática com fotoperíodo controlado. Utilizaram-se quatro repetições de 100 sementes para os testes de germinação e vigor e 25 sementes para o teste de vigor de plântulas. O extrato bruto foi obtido das folhas da gramínea, por meio de técnica laboratorial utilizada para detecção e quantificação de avaliação de saponinas esteroidais. Foram avaliadas duas testemunhas: uma somente com água destilada e outra com água destilada e acrescida de 1% de DMSO e quatro concentrações: 1,85g/mL (10%); 3,7g/mL (20%); 5,55g/mL (30%) e 9,25 mg/mL (50%), totalizando seis tratamentos. Observou-se efeito ?dose-resposta? inibitório significativo (P<0,05) em todos as variáveis avaliadas, nas sementes das três cultivares estudadas. Sabendo-se que os compostos hormonais agem estimulando ou inibindo o metabolismo celular, sugere-se que os efeitos alelopáticos observados no presente trabalho possam estar relacionados ao efeito direto das saponinas esteroidais nas células das sementes e das plântulas das B. brizantha avaliadas. Conclui-se que o extrato bruto de Brachiaria decumbens BRA001996 reduz o vigor e a germinação das sementes, bem como o vigor das plântulas de Brachiaria brizantha, cultivares Xaraés, Piatã e Marandu.bitstream/item/68749/1/DOC188.pd

    Anti-inflammatory effect of glucose—mannose binding lectins isolated from Brazilian beans

    Selectins are essential for leukocyte recruitment in inflammation. Because of a lectin domain present in the selectin structure, we investigated the anti-inflammtory activity of six mannose–glucose binding lectins from brazilian beans: Dioclea guianensis-DguiL; D. grandiflora-DgL; Cratylia floribunda-CfL; D. violacea-D.vL; D. virgata-DvirL and Canavalia brasiliensis-ConBr. The lectins were injected intravenously (i.v.) into rats (0.1 and 1.0 mg/kg; 30 min before irritants) and its activities compared to E. coli endotoxin (LPS,30 μg/kg i.v.). Three lectins (DvL, CfL and DguiL), although less intense than LPS, inhibited the neutrophil migration induced by carrageenan (Cg, 300 μg) in a dose-dependent manner (0.1 and 1.0 mg/kg). DvL activity was reversed by 0.1 M α-D-methyl-mannoside (α-CH3), but not by 0.1 M α-D-galactose. The fMLP (44 ng)-induced neutrophil migration was also reduced by these lectins. Endotoxin contamination of lectin samples could be excluded since α-CH3 treatment reversed the DvL effect, but did not modify LPS inhibitory activity. Carrageenan (300 μg)-induced paw oedema was also reduced by LPS or lectin treatments. Conversely, none of the tested lectins inhibited dextran (Dex, 300 μg)-induced paw oedema, a classical leukocyte independent model, or zymosan (Zy, 1.0 mg)-induced peritonitis and paw oedema. LPS showed no effect upon Dex-induced paw oedema and barely reduced (25%) the oedematogenic effects of zymosan. As proposed for LPS, the lectin inhibitory activity was better observed on neutrophil-mediated inflammatory reactions. We speculate that the plant lectin antiinflammatory activity is probably due to a competitive blockage of a common leukocyte and/or endothelial selectin carbohydrate ligand

    The genus Burkholderia represents a challenge to the fields of taxonomy and phylogeny and, especially, to the understanding of the contrasting roles as either opportunistic pathogens or bacteria with biotechnological potential. Few genomes of nonpathogenic strains, especially of diazotrophic symbiotic bacteria, have been sequenced to improve understanding of the genus. Here, we contribute with the complete genome sequence of Burkholderia phenoliruptrix strain BR3459a (CLA1), an effective diazotrophic symbiont of the leguminous tree Mimosa flocculosa Burkart, which is endemic to South America

    Symbolic Computation via Program Transformation

    Symbolic computation is an important approach in automated program analysis. Most state-of-the-art tools perform symbolic computation as interpreters and directly maintain symbolic data. In this paper, we show that it is feasible, and in fact practical, to use a compiler-based strategy instead. Using compiler tooling, we propose and implement a transformation which takes a standard program and outputs a program that performs semantically equivalent, but partially symbolic, computation. The transformed program maintains symbolic values internally and operates directly on them hence the program can be processed by a tool without support for symbolic manipulation. The main motivation for the transformation is in symbolic verification, but there are many other possible use-cases, including test generation and concolic testing. Moreover using the transformation simplifies tools, since the symbolic computation is handled by the program directly. We have implemented the transformation at the level of LLVM bitcode.     ConBr lectin modulates MAPKs and Akt pathways and triggers autophagic glioma cell death by a mechanism dependent upon caspase-8 activation

    Glioblastoma multiforme is the most aggressive type of glioma, with limited treatment and poor prognosis. Despite some advances over the last decade, validation of novel and selective antiglioma agents remains a challenge in clinical pharmacology. Prior studies have shown that leguminous lectins may exert various biological effects, including antitumor properties. Accordingly, this study aimed to evaluate the mechanisms underlying the antiglioma activity of ConBr, a lectin extracted from the Canavalia brasiliensis seeds. ConBr at lower concentrations inhibited C6 glioma cell migration while higher levels promoted cell death dependent upon carbohydrate recognition domain (CRD) structure. ConBr increased p38MAPK and JNK and decreased ERK1/2 and Akt phosphorylation. Moreover, ConBr inhibited mTORC1 phosphorylation associated with accumulation of autophagic markers, such as acidic vacuoles and LC3 cleavage. Inhibition of early steps of autophagy with 3-methyl-adenine (3-MA) partially protected whereas the later autophagy inhibitor Chloroquine (CQ) had no protective effect upon ConBr cytotoxicity. ConBr also augmented caspase-3 activation without affecting mitochondrial function. Noteworthy, the caspase-8 inhibitor IETF-fmk attenuated ConBr induced autophagy and C6 glioma cell death. Finally, ConBr did not show cytotoxicity against primary astrocytes, suggesting a selective antiglioma activity. In summary, our results indicate that ConBr requires functional CRD lectin domain to exert antiglioma activity, and its cytotoxicity is associated with MAPKs and Akt pathways modulation and autophagy- and caspase-8- dependent cell death.Fil: Wolin, Ingrid A.
