Impact of Natural Blind Spot Location on Perimetry.
We study the spatial distribution of natural blind spot location (NBSL) and its impact on perimetry. Pattern deviation (PD) values of 11,449 reliable visual fields (VFs) that are defined as clinically unaffected based on summary indices were extracted from 11,449 glaucoma patients. We modeled NBSL distribution using a two-dimensional non-linear regression approach and correlated NBSL with spherical equivalent (SE). Additionally, we compared PD values of groups with longer and shorter distances than median, and larger and smaller angles than median between NBSL and fixation. Mean and standard deviation of horizontal and vertical NBSL were 14.33° ± 1.37° and -2.06° ± 1.27°, respectively. SE decreased with increasing NBSL (correlation: r = -0.14, p \u3c 0.001). For NBSL distances longer than median distance (14.32°), average PD values decreased in the upper central (average difference for significant points (ADSP): -0.18 dB) and increased in the lower nasal VF region (ADSP: 0.14 dB). For angles in the direction of upper hemifield relative to the median angle (-8.13°), PD values decreased in lower nasal (ADSP: -0.11 dB) and increased in upper temporal VF areas (ADSP: 0.19 dB). In conclusion, we demonstrate that NBSL has a systematic effect on the spatial distribution of VF sensitivity
Avaliação do potencial de produção de sementes de espécies, formas e cultivares de trevo-branco
From March 1984 to November 1985, the potential seed yield of the native species of white dover T. riograndense Burkart and T. polymorphum Poir. was evaluated in comparison to the BR-1-Bagé, Regal, Jacuà and GuaÃba cultivars of white dover (T. repens L.). The experiment was performed at the Agricultural Experiments Station of the Federal University of Rio Grande do Sul, GuaÃba, RS, Brazil. The variables regarding to the seed yield and the components of the seed yield, DM yield of forage residue, percentage of crude protein (CP) and in vitro dry mater digestibility (IVOND) of the forage residue after seed harvest were evaluated. The cultivars of the white dover present differences. Rega) showed to have no adaptation for seed yield in this place. The native species T. polytnorphuni and T. riograndense (2x and 4x) showed significantly lower seed yield than the cultivars of the white dover. The autotetraploid form of T. riograndense showed no differences between variables, except for number of inflorescences/m 2 and weight of 1000 seeds, that were superior to those of the diploid form.De março de 1984 a novembro de 1985, foi avaliado o potencial de produção de sementes das espécies nativas Trifolium riograndense Burkart e Trifolium polytnorphum Poir. em relação à s cultivares BR-1-Bagé, Regal, Jacuà e GuaÃba de trevo-branco (Trifolium repens L.). O experimento foi realizado na Estação Experimental Agronômica da Universidade Federal do Rio Grande do Sul (EEA-UFRGS), GuaÃba, RS. Foram avaliadas as variéveis referentes aos componentes do rendimento de sementes, além do rendimento de MS do resÃduo, teor de proteÃna bruta e digestibilidade in vitro da MS e MO desse resÃduo. As cultivares de trevo-branco apresentaram variação entre si e a cultivar Regal mostrou-se sem adaptação para a produção local de sementes. As espécies nativas T. po!yrnorphurn e T. rio-grandense (2x e 4x) mostraram produções de sementes significativamente menores que as das cultivares de trevo-branco. A forma autotetraplóide de T. riograndense não apresentou diferenças signiÃicativas entre as variáveis, com exceção do número de inflorescências/m 2 e peso de 1.000 sementes, que foram superiores à s da forma diplóid
Avaliação da depleção linfóide folicular da Bursa de Fabricius: uma metodologia alternativa utilizando análise digital de imagem e redes neurais artificiais
Fifty Bursa of Fabricius (BF) were examined by conventional optical microscopy and digital images were acquired and processed using Matlab® 6.5 software. The Artificial Neuronal Network (ANN) was generated using Neuroshell® Classifier software and the optical and digital data were compared. The ANN was able to make a comparable classification of digital and optical scores. The use of ANN was able to classify correctly the majority of the follicles, reaching sensibility and specificity of 89% and 96%, respectively. When the follicles were scored and grouped in a binary fashion the sensibility increased to 90% and obtained the maximum value for the specificity of 92%. These results demonstrate that the use of digital image analysis and ANN is a useful tool for the pathological classification of the BF lymphoid depletion. In addition it provides objective results that allow measuring the dimension of the error in the diagnosis and classification therefore making comparison between databases feasible.Cinquenta Bursa de FabrÃcius (BF) foram examinadas através de microscopia óptica convencional e imagens digitais foram obtidas e processadas através do software Matlab® 6.5. Redes Neurais Artificiais (ANN) foram geradas com a utilização do software Neuroshell® Classifier, e os dados das análises óptica e digital foram comparados. A ANN classificou corretamente a maioria dos folÃculos, atingindo sensibilidade e especificidade de 89% e 96%, respectivamente. Quando os folÃculos foram agrupados de forma binária houve um aumento da sensibilidade para 90% e obteve-se um valor máximo para a especificidade de 92%. Estes resultados demonstram que o uso da análise digital de imagem associada à ANNé uma ferramenta bastante útil para a classificação patológica da depleção linfóide da BF. Além disso, fornece resultados objetivos que permitem medir a dimensão do erro classificatório, tornando possÃvel a comparação entre distintos bancos de dados

An Artificial Intelligence Approach to Detect Visual Field Progression in Glaucoma Based on Spatial Pattern Analysis.
Purpose: To detect visual field (VF) progression by analyzing spatial pattern changes.
Methods: We selected 12,217 eyes from 7360 patients with at least five reliable 24-2 VFs and 5 years of follow-up with an interval of at least 6 months. VFs were decomposed into 16 archetype patterns previously derived by artificial intelligence techniques. Linear regressions were applied to the 16 archetype weights of VF series over time. We defined progression as the decrease rate of the normal archetype or any increase rate of the 15 VF defect archetypes to be outside normal limits. The archetype method was compared with mean deviation (MD) slope, Advanced Glaucoma Intervention Study (AGIS) scoring, Collaborative Initial Glaucoma Treatment Study (CIGTS) scoring, and the permutation of pointwise linear regression (PoPLR), and was validated by a subset of VFs assessed by three glaucoma specialists.
Results: In the method development cohort of 11,817 eyes, the archetype method agreed more with MD slope (kappa: 0.37) and PoPLR (0.33) than AGIS (0.12) and CIGTS (0.22). The most frequently progressed patterns included decreased normal pattern (63.7%), and increased nasal steps (16.4%), altitudinal loss (15.9%), superior-peripheral defect (12.1%), paracentral/central defects (10.5%), and near total loss (10.4%). In the clinical validation cohort of 397 eyes with 27.5% of confirmed progression, the agreement (kappa) and accuracy (mean of hit rate and correct rejection rate) of the archetype method (0.51 and 0.77) significantly (P \u3c 0.001 for all) outperformed AGIS (0.06 and 0.52), CIGTS (0.24 and 0.59), MD slope (0.21 and 0.59), and PoPLR (0.26 and 0.60).
Conclusions: The archetype method can inform clinicians of VF progression patterns
MitoTALENs: A general approach to reduce mutant mtDNA loads and restore oxidative phosphorylation function in mitochondrial diseases
We have designed mitochondrially targeted Transcription Activator-Like Effector Nucleases or mitoTALENs to cleave specific sequences in the mitochondrial DNA (mtDNA) with the goal of eliminating mtDNA carrying pathogenic point mutations. To test the generality of the approach we designed mitoTALENs to target two relatively common pathogenic mtDNA point mutations associated with mitochondrial diseases: the m.8344A>G tRNA^(Lys) gene mutation associated with Myoclonic Epilepsy with Ragged-Red Fibers (MERRF) and the m.13513G>A ND5 mutation associated with MELAS/Leigh Syndrome. Transmitochondrial cybrid cells harbouring the respective heteroplasmic mtDNA mutations were transfected with the respective mitoTALEN and analysed after different time periods. MitoTALENs efficiently reduced the levels of the targeted pathogenic mtDNAs in the respective cell lines. Functional assays showed that cells with heteroplasmic mutant mtDNA were able to recover respiratory capacity and oxidative phosphorylation enzymes activity after transfection with the mitoTALEN. To improve the design in the context of the low complexity of mtDNA, we designed shorter versions of the mitoTALEN specific for the MERRF m.8344A>G mutation. These shorter mitoTALENs also eliminated the mutant mtDNA. These reductions in size improve our ability to package these large sequences into viral vectors, bringing the use of these genetic tools closer to clinical trials
Exercise and Caloric Restriction Alter the Immune System of Mice Submitted to a High-Fat Diet
As the size of adipocytes increases during obesity, the establishment of resident immune cells in adipose tissue becomes an important source of proinflammatory mediators. Exercise and caloric restriction are two important, nonpharmacological tools against body mass increase. To date, their effects on the immune cells of adipose tissue in obese organisms, specifically when a high-fat diet is consumed, have been poorly investigated. Thus, after consuming a high-fat diet, mice were submitted to chronic swimming training or a 30% caloric restriction in order to investigate the effects of both interventions on resident immune cells in adipose tissue. These strategies were able to reduce body mass and resulted in changes in the number of resident immune cells in the adipose tissue and levels of cytokines/chemokines in serum. While exercise increased the number of NK cells in adipose tissue and serum levels of IL-6 and RANTES, caloric restriction increased the CD4+/CD8+ cell ratio and MCP-1 levels. Together, these data demonstrated that exercise and caloric restriction modulate resident immune cells in adipose tissues differently in spite of an equivalent body weight reduction. CarnÃvoros (Mammalia, Carnivora) do Quaternário da Serra da Bodoquena, Mato Grosso do Sul, Brasil
A report on fossil remains of carnivores from the Quaternary of Serra da Bodoquena (Mato Grosso do Sul, Brazil), recovered from two limestone flooded caves, is presented. A total of six species could be identified belonging to three carnivore families, namely Felidae (Smilodon populator, Panthera onca, Leopardus braccatus), Canidae (Protocyon troglodytes, Chrysocyon brachyurus) and Mustelidae (Pteronura brasiliensis), a mosaic of extinct and extant species presumably related to the Holo-Pleistocene fauna of the region. These findings support in part previous suggestions of a paleoecological scenario of open savannas rich in wetlands for the Quaternary environment of Serra da Bodoquena.Uma primeira apreciação relativa aos fragmentos fósseis de carnÃvoros do Quaternário da Serra da Bodoquena (Mato Grosso do Sul, Brasil) é apresentada. A amostra analisada foi recuperada em duas cavernas submersas e, presumivelmente, representam depósitos de origem associada ao intervalo entre Pleistoceno inferior e o Holoceno. Esta hipótese é corroborada pela megafauna de mamÃferos associada. Seis espécies puderam ser identificadas, compreendendo três famÃlias de CarnÃvora: Felidae (Smilodon populator, Panthera onca, Leopardus braccatus), Canidae (Protocyon troglodytes, Chrysocyon brachyurus) e Mustelidae (Pteronura brasiliensis), retratando, assim, um mosaico de espécies recentes e extintas da fauna de carnÃvoros que compõem a região. Estes achados suportam parcialmente sugestões anteriores de um cenário paleoecológico de savanas ricas em áreas úmidas para o Quaternário da Serra da Bodoquena
Effect of antibodies to intercellular adhesion molecule type 1 on the protection of distant organs during reperfusion syndrome in rats
We investigated kidney and lung alterations caused by intercellular adhesion molecule type 1 (ICAM-1) blockade after ischemia and reperfusion of hind limb skeletal muscles. Rats were submitted to ligature of the infrarenal aorta for 6 h. The animals were randomized into three groups of 6 rats each: group I, sacrificed after ischemia; group II, reperfusion for 24 h, and group III, reperfusion for 24 h after receiving monoclonal anti-ICAM-1 antibodies. At the end of the experiment, blood samples were collected for creatinine, lactate dehydrogenase, creatine phosphokinase, potassium, pH and leukocyte counts. Samples were taken from the muscles of the hind limbs and from the kidneys and lungs for histological analysis and measurement of the neutrophil infiltrate by myeloperoxidase staining. The groups did not differ significantly with regard to the laboratory tests. There were no major histological alterations in the kidneys. An intense neutrophil infiltrate in the lungs, similar in all groups, was detected. Myeloperoxidase determination showed that after reperfusion there was significantly less retention of polymorphonuclear neutrophils in the muscles (352 ± 70 vs 1451 ± 235 × 10² neutrophils/mg; P<0.01) and in the kidneys (526 ± 89 vs 852 ± 73 × 10² neutrophils/mg; P<0.01) of the animals that received anti-ICAM-1 before perfusion compared to the group that did not.
