    Evaluation of antidiabetic potentials of Phyllanthus niruri in alloxan diabetic rats

    The antidiabetic potentials of methanol extract (ME) of aerial parts of Phyllanthus niruri L (Euphorbiaceae) was evaluated in normal and alloxan diabetic rats. The results showed that ME significantly (P<0.05) reduced fasting blood sugar in a dose-related manner and suppressed thepostprandial rise in blood glucose after a heavy glucose meal in normoglycaemic rats. Chronic oral administration of ME caused a significant (P<0.05) dose-related reduction in blood glucose levels as well as total cholesterol and triglycerides levels in diabetic and normoglycaemic rats. Sub-chronic toxicity study showed that ME-treated rats had significant (P<0.05) reductions in haemoglobin (Hb) levels, red blood cell (RBC) and white blood cell (WBC) counts followed by a gradual rise which did not,however, attain basal levels; however, there was a progressive rise in the WBC of ME-treated diabetic rats. Also ME-treated and control rats had increases in weight throughout the study. Histological studies showed that ME-treated diabetic rats had the tissue architecture of their pancreas restored as against the control groups where there was evidence of necrosis. The acute toxicity and lethality test of ME in mice gave an oral LD50 of 471.2 mg/kg. Results suggest that extract of aerial parts of P. niruri has great potentials as anti-diabetic remedy

    Anti-inflammatory activity of fatty extract of Vitalleria paradoxa Kernel (Shea butter) and pattern of its clinical use in arthritis in Enugu, South East Nigeria

    Background: The fatty seed extract of Vitellaria paradoxa Kernel (Sapotaceae) commonly called “Shea butter”, is a popular remedy for arthritis in west Africa. This study investigated the knowledge, prevalence and pattern of use of Shea butter in clinically diagnosed arthritic patients as well as its effects on acute (topical and systemic) and chronic inflammation in rodents.Methods: Knowledge, prevalence and pattern of use were determined using pre-tested questionnaire in clinically diagnosed patients whereas the anti-inflammatory activity was studied using xylene-induced mouse ear edema, carrageenan-induced rat paw edema, formaldehyde-induced arthritis in rats and cotton pellet granuloma test in rats.Results: The results showed that of the 164 respondents, 94.1% know about Shea butter and 59.6% have used it mainly as a massage ointment once or twice daily. However, 73.7% of the users combine this remedy with analgesics to achieve relief. The pharmacological tests showed that topical application of Shea butter inhibited acute edema of the mouse ear. Systemic oral administration caused significant (p<0.05) suppression of the development of systemic acute edema of the rat paw in a non-dose related manner. Twice daily topical application of Shea butter significantly (p<0.05) inhibited the edematous response to formaldehyde arthritis whereas once daily administration was not effective. Shea butter also caused a significant (p<0.05) non-dose related inhibition of granuloma tissue formation on implanted cotton pellets.Conclusions: These findings provide a scientific rationale for the use of Shea butter in treatment of disorders of inflammation in traditional medicine

    Potentials of leaves of Aspilia africana (Compositae) in wound care: an experimental evaluation

    <p>Abstract</p> <p>Background</p> <p>The potentials of the leaves of the haemorrhage plant, <it>Aspilia africana </it>C. D Adams (Compositae) in wound care was evaluated using experimental models. <it>A. africana</it>, which is widespread in Africa, is used in traditional medicine to stop bleeding from wounds, clean the surfaces of sores, in the treatment of rheumatic pains, bee and scorpion stings and for removal of opacities and foreign bodies from the eyes. The present study was undertaken to evaluate the potentials for use of leaves of this plant in wound care.</p> <p>Methods</p> <p>The effect of the methanol extract (ME) and the hexane (HF) and methanol (MF) fractions (obtained by cold maceration and graded solvent extraction respectively) on bleeding/clotting time of fresh experimentally-induced wounds in rats, coagulation time of whole rat blood, growth of microbial wound contaminants and rate of healing of experimentally-induced wounds in rats were studied as well as the acute toxicity and lethality (LD<sub>50</sub>) of the methanol extract and phytochemical analysis of the extract and fractions.</p> <p>Results</p> <p>The extract and fractions significantly (<it>P </it>< 0.05) reduced bleeding/clotting time in rats and decreased coagulation time of whole rat blood in order of magnitude of effect: MF>ME>HF. Also, the extract and fractions caused varying degrees of inhibition of the growth of clinical isolates of <it>Pseudomonas fluorescens </it>and <it>Staphylococcus aureus</it>, as well as typed strains of <it>Ps. aeruginosa </it>(ATCC 10145) and <it>Staph. aureus </it>(ATCC 12600), and reduced epithelialisation period of wounds experimentally-induced in rats. Acute toxicity and lethality (LD<sub>50</sub>) test in mice established an i.p LD<sub>50 </sub>of 894 mg/kg for the methanol extract (ME). Phytochemical analysis revealed the presence of alkaloids, saponins, tannins, flavonoids, resins, sterols, terpenoids and carbohydrates.</p> <p>Conclusion</p> <p>The leaves of <it>A. africana </it>possess constituents capable of arresting wound bleeding, inhibiting the growth of microbial wound contaminants and accelerating wound healing which suggest good potentials for use in wound care.</p

    Acanthus montanus: An experimental evaluation of the antimicrobial, anti-inflammatory and immunological properties of a traditional remedy for furuncles

    <p>Abstract</p> <p>Background</p> <p><it>Acanthus montanus </it>(Nees) T. Anderson (Acanthaceae) is a shrub widespread in Africa, the Balkans, Romania, Greece and Eastern Mediterranean. It is used in African traditional medicine for the treatment of urogenital infections, urethral pain, endometritis, urinary disease, cystitis, leucorrhoea, aches and pains. In southeastern Nigeria, the root is popular and acclaimed highly effective in the treatment of furuncles. This study was undertaken to experimentally evaluate the antimicrobial and anti-inflammatory properties of the root extract as well as its effect on phagocytosis and specific cell-mediated immune response which may underlie the usefulness of the roots in treatment of furuncles.</p> <p>Methods</p> <p>The aqueous root extract (obtained by hot water maceration of the root powder) was studied for effects on the growth of clinically isolated strains of <it>Pseudomonas aeruginosa </it>and <it>Staphylococcus aureus</it>. The anti-inflammatory activity was investigated using acute topical edema of the mouse ear induced by xylene, acute paw edema induced by agar in rats, formaldehyde arthritis in rats, vascular permeability induced by acetic acid in mice and heat- and hypotonicity-induced haemolysis of ox red blood cells (RBCs). Also evaluated were the effects on <it>in vivo </it>leukocyte migration induced by agar, phagocytic activity of macrophages on <it>Candida albicans </it>and specific cell-mediated immune responses (delayed type hypersensitivity reaction (DTHR) induced by sheep red blood cell (SRBC)). The acute toxicity and lethality (LD<sub>50</sub>) in mice and phytochemical constituents of the extract were also determined.</p> <p>Results</p> <p>The extract moderately inhibited the growth of the test organisms and significantly (<it>P </it>< 0.05) inhibited (57%) topical acute edema in the mouse ear. It significantly (<it>P </it>< 0.05) suppressed the development of acute edema of the rat paw in a non-dose-related manner and was not effective in inhibiting the global edematous response to formaldehyde arthritis. It also inhibited vascular permeability induced by acetic acid in mice and the haemolysis of ox RBCs induced by heat- and hypotonicity. The extract increased total leukocyte and neutrophil counts and caused a significant (<it>P </it>< 0.05) dose-related increase in the total number of macrophages at the 800 mg/kg dose. On phagocytic activity, the extract evoked a significant (<it>P </it>< 0.05) increase in the number of macrophages with ingested <it>C. albicans </it>at 800 mg/kg dose, and significantly (<it>P </it>< 0.05) inhibited DTHR in a dose-related manner. Phytochemical tests on the extract revealed an abundant presence of alkaloids and carbohydrates while saponins, glycosides, and terpenoids occurred in trace amounts. Acute toxicity test established an oral and intraperitoneal LD<sub>50 </sub>greater than 5,000 mg/kg.</p> <p>Conclusion</p> <p>The effectiveness of the root of <it>A. montanus </it>in the treatment of furuncles may largely derive from mobilization of leukocytes to the site of the infection and activation of phagocytic activity as well as suppression of exacerbated immune responses by its constituents. Antimicrobial and anti-inflammatory activities are likely contributory mechanisms. Phytochemical constituents such as alkaloids and carbohydrates may be responsible for these pharmacological activities.</p

    Tannin extracts from immature fruits of Terminalia chebula Fructus Retz. promote cutaneous wound healing in rats

    <p>Abstract</p> <p>Background</p> <p>Tannins extracted from immature fruits of <it>Terminalia chebula Fructus Retz</it>. are considered as effective components promoting the process of wound healing. The objective of this study is to explore the optimal extraction and purification technology (OEPT) of tannins, while studying the use of this drug in the treatment of a cutaneous wound of rat as well as its antibacterial effects.</p> <p>Methods</p> <p>The content of tannin extracts was measured by the casein method, and antibacterial ability was studied by the micro-dilution method in vitro. In wound healing experiment, animals in group Ⅰ, Ⅱ and Ⅲ were treated with vaseline ointment, tannin extracts (tannin content: 81%) and erythromycin ointment, respectively (5 mg of ointment were applied on each wound). To evaluate the process of wound healing, selected pharmacological and biochemical parameters were applied.</p> <p>Results</p> <p>After optimal extraction and purification, content of tannin extracts was increased to 81%. Tannin extracts showed the inhibition of <it>Staphylococcus aureus </it>and <it>Klebsiella Pneumonia </it>in vitro. After excision of wounds, on days 7 and 10, the percent of wound contraction of group Ⅱ was higher than that of group Ⅰ. After being hurt with wounds, on days 3, 7, and 10, the wound healing quality of group Ⅱ was found to be better than that of group Ⅰ in terms of granulation formation and collagen organization. After wound creation, on day 3, the vascular endothelial growth factor expression of group Ⅱ was higher than that of group Ⅰ.</p> <p>Conclusion</p> <p>The results suggest that tannin extracts from dried immature fruits of <it>Terminalia chebula Fructus Retz</it>. can promote cutaneous wound healing in rats, probably resulting from a powerful anti-bacterial and angiogenic activity of the extracts.</p

    Syzygium jambolanum treatment improves survival in lethal sepsis induced in mice

    <p>Abstract</p> <p>Background</p> <p>The leaves and the fruits from <it>Syzygium jambolanum </it>DC.(Myrtaceae), a plant known in Brazil as sweet olive or 'jambolão', have been used by native people to treat infectious diseases, diabetes, and stomachache. Since the bactericidal activity of <it>S. jambolanum </it>has been confirmed <it>in vitro</it>, the aim of this work was to evaluate the effect of the prophylactic treatment with <it>S. jambolanum </it>on the <it>in vivo </it>polymicrobial infection induced by cecal ligation and puncture (CLP) in mice.</p> <p>Methods</p> <p>C57Bl/6 mice were treated by the subcutaneous route with a hydroalcoholic extract from fresh leaves of <it>S. jambolanum </it>(HCE). After 6 h, a bacterial infection was induced in the peritoneum using the lethal CLP model. The mice were killed 12 h after the CLP induction to evaluate the cellular influx and local and systemic inflammatory mediators' production. Some animals were maintained alive to evaluate the survival rate.</p> <p>Results</p> <p>The prophylactic HCE treatment increased the mice survival, the neutrophil migration to infectious site, the spreading ability and the hydrogen peroxide release, but decreased the serum TNF and nitrite. Despite the increased migration and activation of peritoneal cells the HCE treatment did not decrease the number of CFU. The HCE treatment induced a significant decrease on the bone marrow cells number but did not alter the cell number of the spleen and lymph node.</p> <p>Conclusion</p> <p>We conclude that the treatment with <it>S. jambolanum </it>has a potent prophylactic anti-septic effect that is not associated to a direct microbicidal effect but it is associated to a recruitment of activated neutrophils to the infectious site and to a diminished systemic inflammatory response.</p

    Evaluation of biogas production rate and biochemical changes in pig dung used in a simple mobile bio-digester

    The rate of biogas generation and biochemical changes in pig dung used in a simple mobile biogas digester designed and constructed at the Department of Environmental Technology, Federal University of Technology Owerri, Nigeria were evaluated. Measurable gas production started 4 days after feeding the digester with pig dung with daily gas volume at this point standing at 0.005m3 and stabilizing at 0.035m 3 for the 5 th and 6 th day. Thereafter, daily gas production dropped and was fluctuating between 0.001m3 and 0.002m3 from the 7th to the 22nd day, and then rose to 0.003m3 on the 23rd day and maintained a daily production volume of 0.009m3 from the 24th to 27th day. A constant volume of 0.008m3 produced for the next 5 days. From the 33rd day the daily gas volume began to increase exponentially to an optimum volume of 0.070m3 on the 42nd day after feeding the digester. All biochemical parameters of the dung analyzed such as Moisture (%), Ether Extract (%), Ash (%), Crude Protein (%), Crude Fiber (%), Crude Fiber (%), Nitrogen (%), Phosphorous mg/100gm and Potassium mg/100mg, showed significant decreases in values after 37 days of digestion except crude fiber value which increased significantly (

    Profile of Canine Cases in Government Veterinary Clinics in Imo State, Nigeria (1986 - 2000)

    A retrospective study of canine cases presented to Veterinary clinics in Imo State (1986-2000) was conducted using case files. Information sought included date of case presentation, breed, sex, age and type of case. During the study period 2,766 dogs were presented for treatment. Of these, 98.4% (2,721) were Nigerian local breeds and 1.8%, exotic breeds. Also, forty different cases were handled in these dogs. The result of the study indicated low mean annual case load, with parasites (endo, 47.29%, ecto, 9.68% and blood protozoan 8.88%) dominating (66% of all cases). These were closely followed by surgical and other related cases (14.75%). Other medical cases such as allergic dermatitis, poisoning and unknown etiologies constituted 2% and 10% respectively.
