    A multiproxy analysis of extreme wave deposits in a tropical coastal lagoon in Jamaica, West Indies

    The Small Island Developing States (SIDS) of the Caribbean Region are vulnerable to natural hazards including earthquakes, tsunamis and tropical cyclones that can cause widespread devastation. Sedimentary archives of these hazards are often well-preserved in coastal lagoons; however, few studies in the Caribbean have adopted a multiproxy approach to their reconstruction. Here, we present a 1200-year multiproxy record of extreme washover events deposited within a coastal mangrove lagoon on the south coast of Jamaica. Manatee Bay lagoon is a permanent fresh-brackish-water mangrove lagoon separated from the Caribbean Sea by a low-elevation carbonate beach. Fifteen sediment cores recovered along five shore-normal transects contain ostracod-rich authigenic carbonate lake muds interspersed with beds of organic lake mud and mangrove peat. The cores contain evidence of multiple palaeo-washover deposits that are readily distinguished by their sedimentology, geochemistry and microfossil assemblages. Hypersaline conditions dominated the early part of the record (~ 800 to 900 CE), and we infer a freshening of lagoonal waters and the subsequent expansion of the mangrove community following an extreme wave event that occurred some time before ~ 1290 to 1400 CE. We constrain the primary historical-washover deposit to 1810–1924 CE (2σ; 71% probability), a period characterised by extreme tectonic and meteorological events, which include the Great Kingston Earthquake of 1907 and a local episode of enhanced hurricane activity. Whilst the balance of circumstantial evidence indicates that the deposit was probably emplaced during the tsunami generated by the 1907 earthquake, we are currently unable to differentiate between tectonically and meteorologically driven washover events based on their sedimentological characteristics

    A sediment-based reconstruction of Caribbean effective precipitation during the Little Ice Age from Freshwater Pond, Barbuda

    Contemporary climate dynamics of the circum-Caribbean region are characterised by significant precipitation variability on interannual and interdecadal timescales controlled primarily by El Niño Southern Oscillation (ENSO) and the Atlantic Multidecadal Oscillation (AMO). However, our understanding of pre-industrial climate variability in the region is hampered by the sparse geographic distribution of palaeoclimate archives. Here, we present a high-resolution reconstruction of effective precipitation for Barbuda since the mid-16th century, based on biostratigraphic and stable isotope analyses of fossil ostracods and gastropods recovered from lake sediment cores from Freshwater Pond, the only freshwater lake on the island. We interpret episodic fluctuations in shell accumulation in the sediment record to represent changes in the balance between precipitation and evaporation during the ‘Little Ice Age’ (LIA; ~1400–1850 CE) and Industrial (1850–present) periods. Comparisons between indices of reconstructed ENSO and AMO variability, the abundance of the freshwater gastropod Pyrgophorus parvulus and the δ18O records from ostracod calcite suggest that the relative influence of ENSO and AMO on long-term rainfall patterns in Barbuda has changed over the last 400 years. Our findings are in agreement with other high-resolution palaeoclimate studies that suggest that long-term changes in effective precipitation during the LIA were much more variable, temporally and spatially, than previously suggested

    Nanocomposite titanium dioxide/polymer photovoltaic cells: effects of TiO2 microstructure, time and illumination power.

    Nanocomposite titanium dioxide/polymer photovoltaic cells have been fabricated using poly[2-(2-ethylhexyloxy)-5-methoxy-1,4-phenylenevinylene] (MEHPPV). Two different types of titanium dioxide were used, one synthesized using a sol-gel method, the other was a commercial paste. The crystal structure, porosity and absorption spectra of the titanium dioxide layers were measured, and the titanium dioxide synthesized using the sol-gel method had a much lower level of anatase. The photovoltaic properties of the ITO/TiO2/MEHPPV/Au cells, which were similar for both types of TiO2, were measured as a function of illumination power and compared with equivalent circuit models. A simple equivalent circuit model incorporating a diode, two resistances and a light induced current was inconsistent with the illumination - dependent data and was improved by adding an illumination dependent shunt resistance. A very long lived, photo-induced increase in dark current was observed, which could not be explained by a polymer degradation mechanism or an increase in temperature under illumination, but was more likely to be due to trapped charge

    Non-steady state operation of polymer/TiO2 photovoltaic devices

    We present data on the initial period of operation of Gilch-route NMH-PPV/TiO2 composite solar cells (CSCs) which show that during this period the CSCs operate in a non-steady state regime. The behavior is complex and may include a gradual rise of the open circuit voltage (V-oc) and of the short-circuit current density (J(sc)) with time, a passage through a maximum of either or both parameters, and even a sign reversal. The mechanisms most probably contributing to the transient processes are: i) diffusion driven redistribution of charges resulting in the build up of a quasi steady state charge density profile across the device; ii) photo-doping resulting in a relatively slow increase of the average charge carrier concentration and consequently of the conductivity of the device. The latter is responsible for a strong decrease in V-oc, and is evidenced by the significant increase in dark current after device illumination

    Characterisation of the bacterial and fungal communities associated with different lesion sizes of Dark Spot Syndrome occurring in the Coral Stephanocoenia intersepta

    The number and prevalence of coral diseases/syndromes are increasing worldwide. Dark Spot Syndrome (DSS) afflicts numerous coral species and is widespread throughout the Caribbean, yet there are no known causal agents. In this study we aimed to characterise the microbial communities (bacteria and fungi) associated with DSS lesions affecting the coral Stephanocoenia intersepta using nonculture molecular techniques. Bacterial diversity of healthy tissues (H), those in advance of the lesion interface (apparently healthy AH), and three sizes of disease lesions (small, medium, and large) varied significantly (ANOSIM R = 0.052 p,0.001), apart from the medium and large lesions, which were similar in their community profile. Four bacteria fitted into the pattern expected from potential pathogens; namely absent from H, increasing in abundance within AH, and dominant in the lesions themselves. These included ribotypes related to Corynebacterium (KC190237), Acinetobacter (KC190251), Parvularculaceae (KC19027), and Oscillatoria (KC190271). Furthermore, two Vibrio species, a genus including many proposed coral pathogens, dominated the disease lesion and were absent from H and AH tissues, making them candidates as potential pathogens for DSS. In contrast, other members of bacteria from the same genus, such as V. harveyii were present throughout all sample types, supporting previous studies where potential coral pathogens exist in healthy tissues. Fungal diversity varied significantly as well, however the main difference between diseased and healthy tissues was the dominance of one ribotype, closely related to the plant pathogen, Rhytisma acerinum, a known causal agent of tar spot on tree leaves. As the corals’ symbiotic algae have been shown to turn to a darker pigmented state in DSS (giving rise to the syndromes name), the two most likely pathogens are R. acerinum and the bacterium Oscillatoria, which has been identified as the causal agent of the colouration in Black Band Disease, another widespread coral disease

    Exome sequencing in dementia with Lewy bodies.

    Dementia with Lewy bodies (DLB) is the second most common form of degenerative dementia. Siblings of affected individuals are at greater risk of developing DLB, but little is known about the underlying genetic basis of the disease. We set out to determine whether mutations in known highly penetrant neurodegenerative disease genes are found in patients with DLB. Whole-exome sequencing was performed on 91 neuropathologically confirmed cases of DLB, supplemented by independent APOE genotyping. Genetic variants were classified using established criteria, and additional neuropathological examination was performed for putative mutation carriers. Likely pathogenic variants previously described as causing monogenic forms of neurodegenerative disease were found in 4.4% of patients with DLB. The APOE ɛ4 allele increased the risk of disease (P=0.0001), conferred a shorter disease duration (P=0.043) and earlier age of death (P=0.0015). In conclusion, although known pathogenic mutations in neurodegenerative disease genes are uncommon in DLB, known genetic risk factors are present in >60% of cases. APOE ɛ4 not only modifies disease risk, but also modulates the rate of disease progression. The reduced penetrance of reported pathogenic alleles explains the lack of a family history in most patients, and the presence of variants previously described as causing frontotemporal dementia suggests a mechanistic overlap between DLB and other neurodegenerative diseases.This study was funded by the NHS National Institute of Health Research Biomedical Research Unit for Lewy body dementia at Newcastle upon Tyne Hospitals NHS Foundation Trust and Newcastle University.     Interaction between polymorphisms in aspirin metabolic pathways, regular aspirin use and colorectal cancer risk: A case-control study in unselected white European populations

    Regular aspirin use is associated with reduced risk of colorectal cancer (CRC). Variation in aspirin’s chemoprevention efficacy has been attributed to the presence of single nucleotide polymorphisms (SNPs). We conducted a meta-analysis using two large population-based case-control datasets, the UK-Leeds Colorectal Cancer Study Group and the NIH-Colon Cancer Family Registry, having a combined total of 3325 cases and 2262 controls. The aim was to assess 42 candidate SNPs in 15 genes whose association with colorectal cancer risk was putatively modified by aspirin use, in the literature. Log odds ratios (ORs) and standard errors were estimated for each dataset separately using logistic regression adjusting for age, sex and study site, and dataset-specific results were combined using random effects meta-analysis. Meta-analysis showed association between SNPs rs6983267, rs11694911 and rs2302615 with CRC risk reduction (All P<0.05). Association for SNP rs6983267 in the CCAT2 gene only was noteworthy after multiple test correction (P = 0.001). Site-specific analysis showed association between SNPs rs1799853 and rs2302615 with reduced colon cancer risk only (P = 0.01 and P = 0.004, respectively), however neither reached significance threshold following multiple test correction. Meta-analysis of SNPs rs2070959 and rs1105879 in UGT1A6 gene showed interaction between aspirin use and CRC risk (Pinteraction = 0.01 and 0.02, respectively); stratification by aspirin use showed an association for decreased CRC risk for aspirin users having a wild-type genotype (rs2070959 OR = 0.77, 95% CI = 0.68–0.86; rs1105879 OR = 0.77 95% CI = 0.69–0.86) compared to variant allele cariers. The direction of the interaction however is in contrast to that published in studies on colorectal adenomas. Both SNPs showed potential site-specific interaction with aspirin use and colon cancer risk only (Pinteraction = 0.006 and 0.008, respectively), with the direction of association similar to that observed for CRC. Additionally, they showed interaction between any non-steroidal anti-inflammatory drugs (including aspirin) use and CRC risk (Pinteraction = 0.01 for both). All gene x environment (GxE) interactions however were not significant after multiple test correction. Candidate gene investigation indicated no evidence of GxE interaction between genetic variants in genes involved in aspirin pathways, regular aspirin use and colorectal cancer risk

    A regional Bayesian POT model for flood frequency analysis

    Flood frequency analysis is usually based on the fitting of an extreme value distribution to the local streamflow series. However, when the local data series is short, frequency analysis results become unreliable. Regional frequency analysis is a convenient way to reduce the estimation uncertainty. In this work, we propose a regional Bayesian model for short record length sites. This model is less restrictive than the index flood model while preserving the formalism of "homogeneous regions". The performance of the proposed model is assessed on a set of gauging stations in France. The accuracy of quantile estimates as a function of the degree of homogeneity of the pooling group is also analysed. The results indicate that the regional Bayesian model outperforms the index flood model and local estimators. Furthermore, it seems that working with relatively large and homogeneous regions may lead to more accurate results than working with smaller and highly homogeneous regions
