    Influência do número de camadas na propriedade mecânica de espécimes fabricados com atadura gessada usados para confeccionar splints ortopédicos

    OBJECTIVE: To evaluate the effects of varying numbers of layers of plaster of Paris bandages on the mechanical properties of specimens used on the construction of orthopedic splints. METHODS: Rectangular plate-shaped and cylinder-shaped specimens were constructed and assigned to two groups simulating plaster slabs and cast and further divided into six subgroups according to the number of layers used: 3, 6, 8, 10, 12 and 14 layers. The specimens were subjected to either a three-point bending test (plates/slab) or compressive strength test (cylinders/cast). The following mechanical properties were evaluated: maximum load, elastic limit load and stiffness. Specimen weight was also calculated. Data was analyzed using Kruskal-Wallis and the least significant difference (LSD) tests. RESULTS: Pairwise comparisons of the subgroups 10x12 and 10x14 revealed significant differences for all mechanical properties (p<0.05). The results of this study suggest that when the goal is to construct appliances with high mechanical strength, regardless of weight, such as serial plaster slabs splints for stimulating tissue growth through the application of gradual load, splints made with plaster of Paris bandages with 12 or 14 layers should be preferred. For orthotic devices such as positioning orthotics, the use of 10 layers plaster bandages slab splints is advisable as they were found to have better correlation between mechanical strength and weight in comparison to those made wtih 6 or 8 layers. CONCLUSION: Based on the findings of this study, we suggest the use of 10 layers of plaster of Paris for the construction of orthopedic splints.OBJETIVO: Avaliar as propriedades mecânicas de amostras fabricadas a partir de ataduras de gesso que são utilizadas em órteses ortopédicas e que variam quanto ao número de camadas. MÉTODOS: Foram confeccionados espécimes em forma de placa retangular e em forma cilíndrica, divididos em dois grupos que simulavam splint e gesso circular, os quais foram divididos em seis subgrupos de acordo com o número de camadas utilizadas, ou seja, três, seis, oito, dez, 12 e 14 camadas. Os espécimes foram submetidos a um teste de inclinação de três pontos (placas/splint) ou teste de resistência à compressão (cilindros/gesso circular). As seguintes propriedades mecânicas foram avaliadas: carga máxima e carga no limite de elasticidade e rigidez. O peso da amostra foi calculado. Os dados foram analisados estatisticamente pelos testes de Kruskal-Wallis e diferença mínima significativa (DMS). Comparações pareadas entre os subgrupos 10x12 e 10x14 revelaram diferenças significativas para todas as propriedades mecânicas (p<0,05). RESULTADOS: Os resultados sugerem que, quando o objetivo é construir aparelhos com alta resistência mecânica, independente do peso, tais como órteses seriadas de posicionamento para simular força gradual aplicada no tecido para a melhoria da amplitude de movimento, talas de 12 ou 14 camadas devem ser preferidas. Para os aparelhos ortopédicos que irão ser submetidos a esforços de baixa intensidade, aconselha-se a utilização de dez camadas para as órteses, porque houve uma melhor correlação entre a resistência mecânica e peso para as amostras fabricadas com dez camadas de atadura gessada comparadas com aquelas confeccionadas com seis ou oito camadas. CONCLUSÂO: Baseado nos achados deste estudo, sugere-se a utilização de dez camadas na confecção de órteses ortopédicas

    A Low-Cost Visible Light Positioning System for Indoor Positioning

    Currently, a high percentage of the world’s population lives in urban areas, and this proportion will increase in the coming decades. In this context, indoor positioning systems (IPSs) have been a topic of great interest for researchers. On the other hand, Visible Light Communication (VLC) systems have advantages over RF technologies; for instance, they do not need satellite signals or the absence of electromagnetic interference to achieve positioning. Nowadays, in the context of Indoor Positioning (IPS), Visible Light Positioning (VLP) systems have become a strong alternative to RF-based systems, allowing the reduction in costs and time to market. This paper shows a low cost VLP solution for indoor systems. This includes multiple programmable beacons and a receiver which can be plugged to a smartphone running a specific app. The position information will be quickly and securely available through the interchange between the receiver and any configurable LED-beacon which is strategically disposed in an area.     ABSTRACT Objective: to evaluate scientifi c production, patents and human resources training of nursing researchers with scholarships of research productivity (PQ) in National Counsel for Technological and Scientifi c Development (CNPq) in the 2000-2012 historic series; to verify the association between this production and characteristics of the researchers regarding gender, education and origin. Methods: this analytic cross-sectional study whose inclusion criterion was to be a nursing PQ/CNPq researcher during the period in question. We analyzed curriculum lattes of 208 researchers with scholarships between 2000 and 2012. For statistical analyses we used the SPSS ® software. Results: the study points to female predominance, concentration in the Southeast region and existence of an association between scientifi c production, patents and human resources training and education, gender and origin of the researcher. Conclusion: the study presents a signifi cant participation of nursing PQ/CNPq researchers in scientifi c production and in human resources training, and a modest involvement in the production of patents. Método: estudo transversal analítico cujo critério de inclusão foi ser pesquisador PQ/CNPq da enfermagem no período em questão. Foram analisados curriculum lattes de 208 pesquisadores com bolsas entre 2000-2012. Para as análises estatísticas utilizou-se o programa SPSS ® . Resultados: o estudo aponta predominância do sexo feminino, concentração na região Sudeste e existência de associação entre produção científi ca, patentes e formação de recursos humanos e a formação, sexo e origem do pesquisador. Conclusão: o estudo revela uma expressiva participação dos pesquisadores PQ/CNPq da enfermagem na produção científi ca e na formação de recursos humanos e tímida participação na produção de patentes.     Establishment of a murine model of congenital toxoplasmosis and validation of a qPCR assay to assess the parasite load in maternal and fetal tissues

    Toxoplasma gondii is the causative agent of toxoplasmosis, a disease that affects warm-blooded animals and one third of the human population worldwide. Pregnant women who have never been exposed to the parasite constitute an important risk group, as infection during pregnancy often leads to congenital toxoplasmosis, the most severe form of the disease. Current therapy for toxoplasmosis is the same as it was 50 years ago and has little or no effect when vertical transmission occurs. Therefore, it is urgent to develop new strategies to prevent mother-to-fetus transmission. The implementation of experimental animal models of congenital toxoplasmosis that reproduces the transmission rates and clinical signs in humans opens an avenue of possibilities to interfere in the progression of the disease. In addition, knowing the parasite load in maternal and fetal tissues after infection, which may be related to organ abnormalities and disease outcome, is another important step in designing a promising intervention strategy. Therefore, we implemented here a murine model of congenital toxoplasmosis with outbred Swiss Webster mice infected intravenously with tachyzoites of the ME49 strain of T. gondii that mimics the frequency of transmission of the parasite, as well as important clinical signs of human congenital toxoplasmosis, such as macrocephaly, in addition to providing a highly sensitive quantitative real-time PCR assay to assess parasite load in mouse tissues. As the disease is not restricted to humans, also affecting several domestic animals, including companion animals and livestock, they can also benefit from the model presented in this study

    A cyclopalladated complex interacts with mitochondrial membrane thiol-groups and induces the apoptotic intrinsic pathway in murine and cisplatin-resistant human tumor cells

    <p>Abstract</p> <p>Background</p> <p>Systemic therapy for cancer metastatic lesions is difficult and generally renders a poor clinical response. Structural analogs of cisplatin, the most widely used synthetic metal complexes, show toxic side-effects and tumor cell resistance. Recently, palladium complexes with increased stability are being investigated to circumvent these limitations, and a biphosphinic cyclopalladated complex {Pd<sub>2 </sub>[<it>S<sub>(-)</sub></it>C<sup>2</sup>, N-dmpa]<sub>2 </sub>(μ-dppe)Cl<sub>2</sub>} named C7a efficiently controls the subcutaneous development of B16F10-Nex2 murine melanoma in syngeneic mice. Presently, we investigated the melanoma cell killing mechanism induced by C7a, and extended preclinical studies.</p> <p>Methods</p> <p>B16F10-Nex2 cells were treated <it>in vitro </it>with C7a in the presence/absence of DTT, and several parameters related to apoptosis induction were evaluated. Preclinical studies were performed, and mice were endovenously inoculated with B16F10-Nex2 cells, intraperitoneally treated with C7a, and lung metastatic nodules were counted. The cytotoxic effects and the respiratory metabolism were also determined in human tumor cell lines treated <it>in vitro </it>with C7a.</p> <p>Results</p> <p>Cyclopalladated complex interacts with thiol groups on the mitochondrial membrane proteins, causes dissipation of the mitochondrial membrane potential, and induces Bax translocation from the cytosol to mitochondria, colocalizing with a mitochondrial tracker. C7a also induced an increase in cytosolic calcium concentration, mainly from intracellular compartments, and a significant decrease in the ATP levels. Activation of effector caspases, chromatin condensation and DNA degradation, suggested that C7a activates the apoptotic intrinsic pathway in murine melanoma cells. In the preclinical studies, the C7a complex protected against murine metastatic melanoma and induced death in several human tumor cell lineages <it>in vitro</it>, including cisplatin-resistant ones. The mitochondria-dependent cell death was also induced by C7a in human tumor cells.</p> <p>Conclusions</p> <p>The cyclopalladated C7a complex is an effective chemotherapeutic anticancer compound against primary and metastatic murine and human tumors, including cisplatin-resistant cells, inducing apoptotic cell death via the intrinsic pathway.</p
