    Monitoring of children with risk indicators for hearing loss

    Purpose: to describe the proposal of monitoring children in the first year of life, who were not identified in the newborn hearing screening program but had risk factors for hearing loss. Method: the study included 258 risk children who had obtained the result “pass” in the Universal Newborn Hearing Screening Program of Hospital Santa Isabel – Bauru/SP, from June to November 2008. It was applied by the telephone, a validated questionnaire in a previous study, containing questions about hearing and language. For each question there were two possible answers: “yes” or “no” and we considered “failure” to obtain at least one “no” answer. With such result, the child was scheduled to perform an immediate hearing evaluation. Results: the questionnaire was applied with 169 families; with the others, there was no contact. From the total, 164 (97,04%) obtained “passed” and five (2,96%) “failed”. Between these five children, only three showed up for hearing evaluation and one had no disorders; two presented conductive hearing loss. It was observed distinct prevalence among the risk factors and there was no relation (p>0,05) of the risk factors with the evasion in the monitoring process. Conclusion: the monitoring through a questionnaire applied by telephone proved to be feasible, however, it is necessary to develop strategies to support their execution.Objetivo: descrever uma proposta de monitoramento de crianças, no primeiro ano de vida, que não \ud foram identificadas na triagem auditiva neonatal, mas apresentavam indicadores de risco para deficiência auditiva. Método: participaram do estudo 258 crianças de risco que haviam obtido o resultado \ud “passa” no Programa de Triagem Auditiva Neonatal Universal da Maternidade Santa Isabel – Bauru/\ud SP no período de junho a novembro de 2008. Foi aplicado, via telefone, um questionário de acompanhamento do desenvolvimento da audição e da linguagem, validado em estudo anterior, contendo \ud questões sobre a audição e a linguagem. Para cada questão havia duas possibilidades de resposta \ud “sim” ou “não” e considerou-se como “falha”, a obtenção de pelo menos uma resposta “não”. Tal \ud resultado refletia no agendamento da criança para realização de uma avaliação audiológica imediata. \ud Resultados: o questionário foi aplicado com 169 famílias, com as demais não foi obtido contato. \ud Deste total, 164 (97,04%) apresentaram resultado “passa” e cinco (2,96%) resultado “falha”. Dentre \ud as cinco crianças, apenas três compareceram para avaliação audiológica e destas, uma não apresentava alterações e duas apresentavam perda auditiva condutiva. Observou-se prevalências distintas \ud entre os fatores de risco e não houve relação (p>0,05) dos mesmos com a evasão no processo de \ud monitoramento. Conclusão: o monitoramento por meio de aplicação de questionário via telefone \ud mostrou-se viável, entretanto, é necessário o desenvolvimento de estratégias que favoreçam sua \ud execução

    Recombinant Probiotics and Microbiota Modulation as a Good Therapy for Diseases Related to the GIT

    Many diseases that affect the gastrointestinal tract (GIT) have great influence on the quality of life of the majority of patients. Many probiotic strains are being highly studied as a promising candidate due to their beneficial effect reported in the GIT. With the purpose of increasing the beneficial characteristics of some probiotics strains and, consequently, to improve further the reported results, many probiotic strains expressing or encoding different proteins, with anti-inflammatory activities, have been developed. These recombinant strains have been reported as good candidates for the treatment of different pathological conditions, especially colitis and mucositis disease since they have been shown to have positive results and good perspectives for GIT inflammation. Thus, this chapter will first address the aspects of the gastrointestinal tract in humans as well as its microbiota. In a second moment, it will discuss about chronic diseases, mainly the intestinal ones. Finally, it will discuss about probiotics, especially concerning on lactic acid bacteria (LAB), and its action in the prevention and treatment of these diseases. At the final part, we will point out aspects on the development of recombinant strains and the results found in the literature on disease models

    Prospective Uses of Genetically Engineered Lactic Acid Bacteria for the Prevention of Inflammatory Bowel Diseases

    Inflammatory bowel disease (IBD) is a term used to describe a group of intestinal disorders in which inflammation is a major feature. Although rare forms of IBD exist, these diseases normally pertain to ulcerative colitis (UC) (Head & Jurenka, 2003) and Crohn’s disease (CD) (Baumgart & Sandborn, 2007). There is evidence that these do not represent distinct conditions but rather are the same disease with shared etiological factors (Price, 1992); however, clinical manifestations (such as the exact location of the pathology or the affected individual’s immunological and constitutional endowment) are distinctive between both. Despite many years of study, the exact etiology and pathogenesis of these disorders remain unclear but great advances have been made using experimental animal models and have provided insights into the complex, multi-factorial processes and mechanisms that can result in chronic intestinal inflammation (Elson & Weaver, 2003). The aim of this chapter is to present an overview of the current expanding knowledge of the mechanisms by which lactic acid bacteria and other probiotic microorganisms participate in the prevention and treatment of IBD and how genetic engineering techniques can be used to improve their effectiveness or create novel therapeutic strains. In the following sections, the mechanisms by which these beneficial microorganisms exert their therapeutic effects, which include changes in the gut microbiota, stimulation of the host immune responses, enhancement of intestinal barrier function and reduction of the oxidative stress due to their antioxidant properties will be discussed.Fil: Leblanc, Jean Guy Joseph.     Mechanisms involved in the anti-inflammatory properties of native and genetically engineered lactic acid bacteria

    Lactic acid bacteria (LAB) represent a heterogeneous group of microorganisms that have been shown to possess therapeutic properties since they are able to prevent the development of some diseases, as shown mostly on animal models for cancer, infections and gastrointestinal disorders such as intestinal inflammation. LAB have been shown to regulate mucosal immune responses by modulating the production and liberation of regulatory agents such as cytokines by the host. Some of these cytokines, such as the anti-inflammatory interleukin-10 (IL-10), modulate the inflammatory immune response, thus immunomodulation is a mechanism by which LAB can prevent certain inflammatory bowel diseases (IBD). Since oxidative stress participates to the inflammatory processes and to the appearance of damages in pathologies of the gastrointestinal tract of humans such as IBD, LAB could also prevent inflammation by eliminating reactive oxygen species (ROS) through the activity of antioxidant enzymes.In order to obtain novel strains or enhance beneficial effects of LAB, genetic engineering has been used to produce either antioxidant enzymes (such as catalases and superoxide dismutases) or anti-inflammatory cytokines (such as IL-10) producing LAB. These novel strains have successfully been used to prevent inflammatory bowel diseases in animal models and could be evaluated in human clinical trials. Here, we present an overview of the current knowledge of the mechanisms by which LAB can be used to prevent undesired intestinal inflammatory responses and could be used as a therapeutic tool for IBD.Fil: Leblanc, Jean Guy Joseph.     In vitro differentiation of mesenchimal stem cells of dogs into osteogenic precursors

    O objetivo principal da nossa pesquisa foi avaliar o potencial de diferenciação osteogênica de células-tronco mesenquimais (MSC) obtidas da medula óssea do cão. As MSC foram separadas pelo método Ficoll e cultivadas sob duas condições distintas: DMEM baixa glicose ou DMEM/F12, ambos contendo L-glutamina, 20% de SFB e antibióticos. Marcadores de MSC foram testados, confirmando células CD44+ e CD34- através da citometria de fluxo. Para a diferenciação osteogênica, as células foram submetidas a quatro diferentes condições: Grupo 1, as mesmas condições utilizadas para a cultura de células primárias com os meios DMEM baixa glicose suplementado; Grupo 2, as mesmas condições do Grupo 1, mais os indutores de diferenciação dexametasona, ácido ascórbico e b-glicerolfosfato; Grupo 3, células cultivadas com meios DMEM/F12 suplementado; e Grupo 4, nas mesmas condições que no Grupo 3, mais indutores de diferenciação de dexametasona, ácido ascórbico e b-glicerolfosfato. A diferenciação celular foi confirmada através da coloração com alizarin red e da imunomarcação com o anticorpo SP7/Osterix. Nós observamos através da coloração com alizarin red que o depósito de cálcio foi mais evidente nas células cultivadas em DMEM/F12. Além disso, usando a imunomarcação com o anticorpo SP/7Osterix obtivemos positividade em 1:6 células para o Meio DMEM/F12 comparada com 1:12 para o meio DMEM-baixa glicose. Com base nos nossos resultados concluímos que o meio DMEM/F12 é mais eficiente para a indução da diferenciação de células-tronco mesenquimais caninas em promotores osteogênicos. Este efeito provavelmente ocorre em decorrência da maior quantidade de glicose neste meio, bem como da presença de diversos aminoácidos.The aim of our research was to evaluate the potential for osteogenic differentiation of mesenchimal stem cells (MSC) obtained from dog bone marrow. The MSC were separated using the Ficoll method and cultured under two different conditions: DMEM low glucose or DMEM/F12, both containing L-glutamine, 20% of FBS and antibiotics. MSC markers were tested, confirming CD44+ and CD34- cells with flow cytometry. For osteogenic differentiation, cells were submitted to four different conditions: Group 1, same conditions used for primary cell culture with DMEM supplemented media; Group 2, same conditions of Group 1 plus differentiation inductors Dexametazone, ascorbic acid and β-glicerolphosphate. Group 3, Cells cultured with supplemented DMEM/F12 media, and Group 4, same conditions as in Group 3 plus differentiation inductors Dexametazone, ascorbic acid and β-glicerolphosphate. The cellular differentiation was confirmed using alizarin red and imunostaining with SP7/Osterix antibody. We observed by alizarin staining that calcium deposit was more evident in cells cultivated in DMEM/F12.Furthermore, by SP/7Osterix antibody immunostaining we obtained 1:6 positive cells when using DMEM/F12 compared with 1:12 for low-glucose DMEM. Based on our results, we conclude that the medium DMEM/F12 is more efficient for induction of differentiation of mesenchymal stem cells in canine osteogenic progenitors. This effect is probably due to the greater amount of glucose in the medium and the presence of various amino acids

    In vivo and in vitro evaluation of an Acetobacter xylinum synthesized microbial cellulose membrane intended for guided tissue repair

    <p>Abstract</p> <p>Background</p> <p>Barrier materials as cellulose membranes are used for guided tissue repair. However, it is essential that the surrounding tissues accept the device. The present study histologically evaluated tissue reaction to a microbial cellulose membrane after subcutaneous implantation in mice. Furthermore, the interaction between mesenchymal stem cells and the biomaterial was studied <it>in vitro </it>to evaluate its ability to act as cellular scaffold for tissue engineering.</p> <p>Methods</p> <p>Twenty-five Swiss Albino mice were used. A 10 × 10 mm cellulose membrane obtained through biosynthesis using <it>Acetobacter xylinum </it>bacteria was implanted into the lumbar subcutaneous tissue of each mouse. The mice were euthanatized at seven, 15, 30, 60, and 90 days, and the membrane and surrounding tissues were collected and examined by histology.</p> <p>Results</p> <p>A mild inflammatory response without foreign body reaction was observed until 30 days post-surgery around the implanted membrane. Polarized microscopy revealed that the membrane remained intact at all evaluation points. Scanning electron microscopy of the cellulose membrane surface showed absence of pores. The <it>in vitro </it>evaluation of the interaction between cells and biomaterial was performed through viability staining analysis of the cells over the biomaterial, which showed that 95% of the mesenchymal stem cells aggregating to the cellulose membrane were alive and that 5% were necrotic. Scanning electron microscopy showed mesenchymal stem cells with normal morphology and attached to the cellulose membrane surface.</p> <p>Conclusion</p> <p>The microbial cellulose membrane evaluated was found to be nonresorbable, induced a mild inflammatory response and may prove useful as a scaffold for mesenchymal stem cells.</p

    A novel interleukin-10 DNA mucosal delivery system attenuates intestinal inflammation in a mouse model

    Inflammatory bowel diseases (IBD) describe a group of complex intestinal disorders characterized by inflammation in the gastrointestinal tract. Current treatments for IBD include the use of anti-inflammatory drugs; furthermore, recombinant lactic acid bacteria have been used as a therapeutic vehicle for anti-inflammatory agents in IBD models. Interleukin-10 (IL-10) is one of the most important anti-inflammatory cytokines; however, its oral administration is limited because it is quickly degraded in the gastrointestinal tract and systemic treatments have led to undesirable side effects. In this study, an engineered invasive strain of Lactococcus (L.) lactis producing Fibronectin Binding Protein A (FnBPA+), from Staphylococcus aureus capable of delivering, directly inside eukaryotic cells, an eukaryotic DNA expression vector containing the ORF coding for IL-10 of Mus musculus (pValac:il-10) was developed and its functionality was evaluated using in vitro and in vivo assays. Functionality of the plasmid and the invasive strain was demonstrated by transfection and invasiveness assays using cell cultures and in vivo in mice by fluorescence microscopy. TNBS inoculated mice that received this novel strain showed lower damage scores in their large intestines (at both macroscopic and microscopic levels), lower microbial translocation to liver, and increased anti-inflammatory/pro-inflammatory cytokine ratios compared to mice that received L. lactis FnBPA+ without the pValac:il-10 plasmid. The effectiveness was demonstrated of this novel DNA delivery therapeutic strategy in the prevention of inflammation using a murine model of colitis.Fil: del Carmen, Silvina Andrea.     Consumo Alimentar e Atividade Física de Universitários de Enfermagem em Tempos de Pandemia por COVID-19 / Food Consumption and Physical Activity of Nursing University Students in Times of Pandemic by COVID-19

    Após a chegada do coronavírus ao Brasil, medidas de segurança foram tomadas para contenção da doença, como o isolamento social, causando grandes impactos na vida das pessoas, principalmente estudantes, que tiveram que se adaptar a uma nova rotina de aulas remotas. O objetivo deste estudo foi identificar o consumo alimentar e a mudança na rotina de atividade física de estudantes de enfermagem durante o período de pandemia por Covid-19. Tratou-se de um estudo transversal, descritivo, no qual foram utilizados dados quantitativos. Dessa forma, evidenciou-se que, durante o período de isolamento social e de quarentena doméstica, ocorreram mudanças no comportamento alimentar e nos padrões de atividade física, influenciando no peso corporal, na saúde e no bem-estar dos estudantes. Conclui-se que o esforço na escolha de um estilo de vida saudável tem sido uma grande responsabilidade dos indivíduos durante a pandemia de Covid-19 e deve ser incentivado

    A influência da utilização de medicamentos no risco de quedas em idosos de instituições de longa permanência do distrito federal / The influence of drug use on the risk of falls in elderly in long stay institutions of the Federal District

    As quedas se constituem como um problema de saúde pública com altas taxas de morbimortalidade na população idosa. Vale salientar que no Brasil, os idosos fazem uso de grande quantidade de medicamentos, alguns com possíveis efeitos colaterais que podem elevar o risco de quedas. Diante disso o presente estudo trata-se de uma pesquisa descritiva, de corte transversal, com abordagem quantitativa, que teve como objetivo identificar os fatores existentes entre a utilização de medicamentos e o risco de quedas em duas instituições de longa permanência para idosos (ILPIs) do DF. Os resultados mostraram predomínio de polifarmácia em 100% participantes, incluindo o uso de medicamentos com potencial para elevar o risco de quedas. Conclui-se que devido ao crescente envelhecimento populacional, é essencial que mais pesquisas sejam realizadas e que medidas sejam implementadas para a redução dos riscos relacionados às quedas por meio da prevenção de condições de vulnerabilidade
