    Torasemide Improves the Propionic Acid-Induced Autism in Rats: A Histopathological and Imaging Study

    Objective: Autism spectrum disorder is a neurodevelopmental disease in which impaired social behaviors, impaired sociality, and restricted and repetitive behaviors are seen. Bumetanide is a loop diuretic that inhibits Na+-K+-2Cl(-) cotransporter 1 and it is currently used in clinical phase studies in patients with autism spectrum disorder. In present research, it is purposed to demonstrate the beneficial effects of torasemide which is another Na+-K+-2Cl(-) cotransporter 1 inhibitor on an experimental autism model induced with propionic acid by providing imaging and brain tissue investigations. Methods: Male Wistar rats were used in the present study (n = 30). Propionic acid of 250 mg/kg/day was administrated intraperitoneally in rats to induce autism for 5 days. Three groups were created for present study as follows: group 1, normal control (n = 10); group 2, propionic acid and saline given group (n = 10); group 3, propionic acid + torasemide -administrated group (n = 10). Results: Torasemide group scored higher on behavioral tests compared to saline group. The brain levels of malondialdehyde, tumor necrosis factor-alpha, interleukin-2, interleukin-17, and Nuclear Factor kappa B (NF-kappa B), Glial fibrillary acidic protein (GFAP) were remarkably higher in propionic acid + saline group. In histopathology assessments, torasemide group had higher neuronal count of Cornu Ammonis 1, neuronal count of Cornu Ammonis 2 in hippocampus, and Purkinje cells in cerebellum. GFAP immunostaining index (Cornu Ammonis 1) and cerebellum were lower in torasemide group. Magnetic resonance spectroscopy revealed that mean lactate value was higher in propionic acid + saline group compared to torasemide group. Conclusion: Our experimental results showed that torasemide might enhance gamma-aminobutyric acid activity. Torasemide can be considered another promising Na+-K+-2Cl(-) cotransporter 1 inhibitor in the treatment of autism with a longer half-life and less side effects after further studies

Akatizi ciddi bir yan etkidir ve genellikle tipik antipsikotiklerin yan etkisi olarak kabul edilir. Akatiziyi fark etmek olası intihar girişimlerini önlemek açısından oldukça önemlidir. Bu yazıda akatizi hakkında genel bilgi vermeyi ve antidepresan tedaviyle olan ilişkisini gözden geçirmeyi amaçladık.Common adverse effects associated with antidepressants such as headache, nause or vomitting, sedation, insomnia and sexual dysfunction are well known; however our knowledge about the relationship between antidepressants and akathisia is limited. Akathisia is a severe side effect and it is commonly considered as a side effect of conventional antipsychotics. Recognizing akathisia is very important in terms of preventing possible suicide attempts.     Long-lasting cognitive effects of COVID-19: is there a role of BDNF?

    Coronavirus disease 2019 (COVID-19) affects numerous systems of the body during the illness, and there have been long-lasting effects. BDNF plays an important role in synaptic plasticity and synaptic communication. According to the inclusion and exclusion criteria, 54 patients who had COVID-19 infection participated in this study. Thirty-six age-, sex-, body mass index (BMI)-, education level- and smoking status-matched healthy controls were included in the present study. All participants were individually administered the Stroop test and Visual Aural Digit Span Test Form B (VADS-B). Serum BDNF levels were measured by ELISA. Stroop test word reading spontaneous correction number and reading time, word color saying wrong number, spontaneous correction number and reading time, box color speaking spontaneous correction number and reading time, Stroop interference and speed factor duration were significantly higher in the COVID-19 group than in the control group. All scores of the VADS-B test were found to be significantly lower in the COVID-19 group. The mean serum BDNF levels were found to be 10.9 +/- 6.9 ng/ml in the COVID-19 group and 12.8 +/- 6.4 ng/ml in the healthy control group. Two-way ANOVA showed that the serum mean BDNF level was significantly lower in the COVID-19 group than in the control group. Gender had a significant effect on BDNF levels (F = 12.21; p = 0.008). The present study is the first to demonstrate the association between the role of serum BDNF and cognitive decline in patients with COVID-19 infection. Additionally, there is a significant role of male gender in terms of lower BDNF level and cognitive decline

    Increased serum dehydroepiandrosterone sulfate in the first episode but not in subsequent episodes in male patients with schizophrenia

    BACKGROUND: Many studies have investigated the relationship between blood levels of dehydroepiandrosterone (DHEA) and its sulfate ester (DHEA-S), cortisol, progesterone, and testosterone and the onset, prognosis, symptom severity, and treatment response of schizophrenia. In the present study, we assessed potential differences in blood levels of neurosteroids between drug-naïve first-episode patients with schizophrenia (FES), and drug-free patients with schizophrenia who were not in the first episode but were in a phase of acute exacerbation (DFP). MATERIALS AND METHODS: The present study included 32 male FES, 28 male DFP, and 24 male healthy controls (HC). Groups were compared in terms of blood levels of adrenocorticotropic hormone (ACTH), cortisol, testosterone, progesterone, and DHEA-S. RESULTS: Blood levels of ACTH, cortisol, testosterone, and progesterone were similar among the groups. The mean value of serum DHEA-S was significantly different among the groups (P<0.001). The value of serum DHEA-S was higher in the FES group than in the DFP and HC groups (both P<0.001). The mean values of serum DHEA-S in the HC and DFP groups were found to be similar (P=0.33). CONCLUSION: We suggest that higher values of DHEA-S in the FES group compared with both the DFP and HC groups indicate that this neurosteroid response is unique to first-episode schizophrenia patients. Further studies are needed to investigate the difference in blood levels of neurosteroids in different groups in terms of age of diagnosis

    Molekularna dijagnostika i serološka istraživanja pestivirusa u ovaca.

    In this research, the virological and serological presence of pestiviruses, such as border disease virus (BDV) was investigated in a sheep in the Central and Eastern Black Sea Regions of Turkey. The study material consisted of 40 organ materials collected from 13 aborted lambs, which were suspected to have pestiviruses, BDV and Bovine Viral Diarrhoea Virus (BVDV). Viral nucleic acids were investigated by using the reverse transcription polymerase chain reaction (RT-PCR). Nine of 13 aborted lambs (69.2%) and 24 of 40 organ samples (60%) obtained from those sheep were BDV RNA positive, whilst all the animals and organ samples were BVDV-RNA negative. Serum samples collected from 401 randomly selected sheep were investigated for pestivirus antibodies using competitive ELISA (cELISA) and the serum microneutralisation test (SNT) using BVDV-NADL strain. Seropositivity was found in between 7.22 and 74.38% with cELISA and 4.81 and 67.76% with SNT. Seropositivity rates in Amasya and Tokat provinces were higher than in Samsun and Giresun. The obtained data indicated that pestivirus infection in sheep is widespread in the Central and Eastern Black Sea Region.U radu su prikazana virološka i serološka istraživanja pestivirusa odnosno borderske bolesti u ovaca na središnjem i istočnom području Crnoga mora u Turskoj. Ukupno je bilo pretraženo 40 uzoraka tkiva različitih organa uzetih od 13 pobačenih plodova pod sumnjom da je uzročnik bio virus borderske bolesti ili virus virusnog proljeva goveda. Uzorci su bili pretraženi na prisutnost virusne nukleinske kiseline lančanom reakcijom polimerazom uz prethodnu reverznu transkripciju (RT-PCR). Devet od 13 pobačenih plodova (69,2%) i 24 od 40 uzoraka tkiva organa (60%) sadržavalo je RNA virusa borderske bolesti. Svi pretraženi uzorci bili su negativni na virus virusnoga proljeva. Uzorci seruma nasumce prikupljeni od 401 ovce bili su pretraženi na prisutnost specifičnih protutijela uporabom kompetitivnoga imunoenzimnoga testa (cELISA) i serum neutralizacijskoga testa (SNT) sa sojem NADL virusa virusnoga proljeva goveda. Ustanovljeno je da sepostotak serološki pozitivnih cELISA-om kretao od 7,22 do 74,38%, a SNT-om od 4,81 do 67,76%. Postotak serološki pozitivnih životinja u provincijama Amasya i Tokat bio je veći nego u provincijama Samsun i Giresun. Rezultati upućuju na zaključak da su pestivirusne zaraze rasprostranjene u ovaca na središnjem i istočnom području Crnoga mora
