    Effect of sorafenib and celecoxib combination therapy on proliferation of the human cholangiocarcinoma cell line SK-ChA-1 in vitro

    目的研究索拉非尼联合塞来昔布在体外对胆管癌细胞株Sk-CHA-1增殖的影响。方法体外培养人胆管癌细胞株Sk-CHA-1,通过MTT法检测索拉非尼单用或与塞来昔布联用时对胆管癌细胞株增殖的抑制作用,WESTErn blOT分析索拉非尼单用或与塞来昔布联用时对胆管癌细胞株内多聚AdP核糖聚合酶(PArP)蛋白表达的影响。结果索拉非尼抑制胆管癌细胞株Sk-CHA-1的增殖并诱导细胞凋亡。索拉非尼联合塞来昔布协同抑制胆管癌细胞株Sk-CHA-1的增殖。塞来昔布使索拉非尼诱导的胆管癌细胞株Sk-CHA-1的凋亡增加。结论索拉非尼联合塞来昔布能协同抑制胆管癌细胞株Sk-CHA-1的增殖,这与塞来昔布使索拉非尼诱导的细胞凋亡增加有关。Objective To investigate the effect of sorafenib and celecoxib combination therapy on proliferation of human cholangiocarcinoma(CC) cells,using the cultured SK-ChA-1 cell line.Methods Inhibition of SK-ChA-1 cell proliferation by sorafenib alone and in combination with celecoxib was studied in vitro using the MTT assay.The anti-neoplastic mechanisms of sorafenib alone and in combination with celecoxib were assessed by Western blot detection of changes in the caspase cleavage substrate poly ADP-ribose polymerase(PARP).Results SK-ChA-1 cells treated with sorafenib alone showed a dose-dependent growth inhibition and degradation of PARP.Combination treatment with sorafenib and celecoxib synergistically increased the growth inhibition effects and enhanced the degradation of PARP.Conclusion Combination treatment with sorafenib and celecoxib results in a synergistic anti-proliferative effect in the human CC cell line SK-ChA-1;the addition of celecoxib enhances sorafenib-induced apoptosis.国家自然科学基金资助项目(81072014

    The Impact of School Choice on Subject Well-being of Student: An Empirical Research Based on Evidence from the China Education Panel Survey

    本文利用2013-2014学年中国教育追踪调查数据,实证研究"择校"对学生主观幸福感的影响。结果发现:无论是否控制学生、父母、家庭特征变量和校级效应,"择校"对学生主观幸福感均呈显著负向影响。即使在使用倾向值得分匹配方法缓解可观测变量的自选择问题、使用干预效应模型缓解不可观测变量的自选择问题、使用工具变量Probit校正内生性偏误等情况下,这一结论仍稳健。分样本估计也发现,"以钱择校"、"以权择校"和"以房择校"学生感到幸福的概率基本上显著地低于"非择校生"。本文的实证结果对我国当前的"择校热"有一定的启示意义。We try to estimate the impact of school choice on subject well-being of student by using China Education Panel Survey( CEPS) from 2013 to 2014 academic year. The paper finds that school choice has significantly negative impact on the student's subject well-being whether or not we control some relevant characteristic variables among children,parents,family and fixed effect in the school level. Such conclusion remains the same for various robustness tests,like using propensity score matching to overcome the self-selection of observable variable,using treatment effects model to overcome the self-selection of unobservable variable,using instrumental variable probit to correct endogenous bias. When it divides into several sub-samples included school choice by spending money,school choice by using privilege,school choice by using purchasing house,which has still significantly negative impact on the student's subject well-being basically.     A Study of New Mode of Two-Phase Culture of Dunaliella Salina for Producing β-carotene

    通过模拟现场条件的正交设计实验l_(27)(3--(13)),研究了盐藻两阶段养殖生产β-胡萝卜素的最佳环境因素。结果表明:在相同时间内(14d),两阶段养殖比一次性养殖生产β-胡萝卜素产量(Mg/l)提高1.19倍,其生长速率(d--(-1))提高1.09倍。The optimum environmental factors of producing β-carotene condition of two ?phase culture of Dunaliella Salina is studied by the experiment of the othogonal design [L27 (313)] for simulating the field condition.The results show that theβ-carotene production (mg/L) and the growth rate (d-1) for the new ?mode of two-hase culture have increased by 1.19 times and 1.09 times respectively as compared with the old mode of batch culture in the same period (14 days)

    根据转基因农作物中最常用的花椰菜花叶病毒启动子(CaMV35S)和根癌农杆菌终止子(NOS)的序列,设计并合成了两对不同的引物,建立了PCR 酶切分析 Southern杂交检测并确认转基因成分35S启动子和NOS终止子的方法.并利用该方法对马铃薯、大豆、玉米、甜椒、番茄等实物样品进行了检测,发现13份样品中有6份检出35S启动子和NOS终止子,7份样品结果为阴性.结果表明建立的检测与验证方法能有效检出转基因作物及其食品中的35S启动子和NOS终止子,具有灵敏度高、特异性好和结果准确的特点,可应用于进出境产品的转基因成分检测.The article is to establish PCRRestrictionSouthern method for detecting and identification transgenic component 35S promoter derived from Cauliflower Mosaic Virus and NOS terminator derived from Agrobacterium tumefaciens in foods. According to the specific sequences of 35S and NOS that have been used in transgenic crops frequently, two pairs of primers were designed to amplify parts of 35S promoter and NOS terminator, to allow for general screening of foods. PCRRestrictionSouthern blot method was established for the detection and identification of transgenic component 35S and NOS. With the method we analyzed potato, soybean, corn, pimiento and tomato samples. The results showed that 6 samples were positive, 7 samples were negative. The described method enabled a sensitive, specific and accurate detection of transgenic component and thus provides a useful tool for routine analysis of entryexit raw and processed food products.厦门市科技计划(3502Z2001109)资
