    HPLC Determination of Chlorogenic Acid,Baicalin and Puerarin in a TCM Preparation of "Xiao-er Resuqing" Oral Liquor

    用高效液相色谱法测定一种中成药"小儿热速清"口服液中的绿原酸、黄芩苷和葛根素含量。采用Zorbax SB-C18色谱柱分离,以不同比例混合的甲醇和乙酸-水(1+99)溶液为流动相,进行梯度洗脱,在327nm波长处测定绿原酸、黄芩苷的含量,在250nm波长处测定葛根素的含量。绿原酸质量浓度在3.0~30.0mg.L-1范围内呈线性关系;黄芩苷质量浓度在2.7~27.0mg.L-1范围内呈线性关系;葛根素质量浓度在3.1~31.0mg.L-1范围内呈线性关系。方法的平均回收率为99.5%,99.8%,99.3%,相对标准偏差(n=6)分别为0.88%,0.84%,1.0%。The effective components,i.e.chlorogenic acid,baicalin and puerarin,in a TCM preparation,named "xiao-er resuqing" oral liquor,were determined by HPLC.The Zorbax SB-C18 chromatographic column was used as the stationary phase and mixtures of methanol and CH3COOH-H2O(1+99) solution with various proportions were used in the gradient elution.UV detections at 327 nm(for chlorogenic aicd and baicalin) and at 250 nm(for puerarin) were adopted in the determinations.Linear relationships between values of peak area and mass concentration of the 3 compounds were found in the following ranges of ① 3.0-30.0 mg·L-1,② 2.7-27.0 mg·L-1 and ③ 3.1-31.0 mg·L-1 respectively.Values of average recovery obtained were 99.5%(for chlorogenic acid),99.8%(for baicalin) and 99.3%(for puerarin).Values of RSD′s(n=6) found for the 3 compounds were 0.88%,0.84% and 1.0% respectively

    建立了HPLC法同时测定感冒解毒灵颗粒中的橙皮苷和牛蒡苷。采用C18色谱柱,以1%乙酸-甲醇(52︰48)为流动相,检测波长280nm。橙皮苷和牛蒡苷在7.5~150和5~100μg/ml浓度范围内线性关系良好,平均回收率为99.2%和99.6%,RSD为0.64%和0.79%。An HPLC method was established for the simultaneous determination of hesperidin and arctiin in Ganmao Jieduling granules. A C18 column was used with the mobile phase of 1% acetic acid-methanol(52︰48) at the detection wavelength of 280 nm. The calibration curves of hesperidin and arctiin were linear in the concentration ranges of 7.5-150 and 5-100 μg/ml. The average recoveries were 99.2% and 99.6%,with RSDs of 0.64% and 0.79%

    目的:建立高效液相色谱法同时测定止咳宁嗽胶囊中的有效成分绿原酸和橙皮苷的方法。方法:样品用甲醇超声提取,在ZorbaxSB-C18(3.0mm×250mm,5μm)色谱柱上分离,以含1%乙酸的水(A)-乙腈(B)体系为流动相进行阶梯洗脱,程序为0min(82∶18)~5min(82∶18)~5.1min(75∶25)~20min(75∶25);流速为0.35mL·min-1;327nm紫外检测。结果:绿原酸在进样量为0.4~10.0μg·mL-1内呈良好的线性关系;橙皮苷在进样量为6.2~155.0μg·mL-1内呈良好的线性关系。绿原酸的平均回收率为99.2%,RSD为1.0%。橙皮苷的平均回收率为99.6%,RSD为0.9%。结论:该方法简便快速,结果准确,重复性好,可为止咳宁嗽胶囊的质量控制提供参考依据。Objective:To establish a HPLC method for simultaneous determination of chlorogenic acid and hesperidin in Zhike Ningsou Capsule .Method:The sample was extracted with methanol under ultrasonication.The HPLC separation was performed on a Zorbax SB C18(3.0 mm × 250 mm,5 μm) column,using water containing 1% acetic acid(A)–acetonitrile( B) as the mobile phase,in stepwise elution:0 min(82∶18)~5 min(82∶18) ~5.1 min(75∶ 25)~20 min(75∶25),at a flow rate of 0.35 mL·min-1.Result:The linear range was 0.4~10.0 μg·mL-1 and 6.2~155.0 μg·mL-1 for chlorogenic acid and hesperidin; the average recovery was 99.2% and 99.6% ; RSD 1.0% and 0.9% ,respectively.Conclusion:This method can be applied for the quality control of Zhike Ningsou Capsule.广西民族大学学校重点科研项目2009MDZD4

    建立一种同时测定健儿消食口服液中的橙皮苷和黄芩苷的高效液相色谱法(HPLC).样品用甲醇超声提取,采用Zorbax SB-C18色谱柱上分离,以1%乙酸溶液-乙腈为流动相进行梯度洗脱,检测波长280 nm,柱温为25℃时直接测定.橙皮苷在15.5~155.0μg.ml-1内线性关系良好;黄芩苷在58.0~290.0μg.ml-1内线性关系良好.平均回收率为99.5%和99.6%,RSD为1.1%和0.85%.结果表明,所建立的方法简便、准确,为该制剂的质量控制提供研究依据.To establish a high performance liquid chromatographic (HPLC) method for simultaneous determination of hesperidin and baicalin in Jian'er Xiaoshi Oral Liquid. The chromatographic separation was achieved on a Zorbax SB--C18 (3.0mm ×250mm, 5μm) column at the detection wavelength of 280nm, using a mobile phase consisted of 1% acetic acid solution and acetonitrile in gradient elution at a flow rate of 0. 35ml.min--1. The linear ranges were 15.5--155.0μg·ml-1 and 58.0--290.0μg·ml-1 for hesperidin and baicalin, respectively. The average recoveries of hesperidin was 99.5% and the relative standard deviations was 1.1% , those of baicalin were 99.6% and 0.85%

    目的考察青天葵Nervilia fordii(Hance)Schltr.氯仿、乙酸乙酯、正丁醇、乙醇提取物抗氧化活性与其总黄酮和总酚含有量的相关性。方法 Al Cl3-HAc-Na Ac显色法和福林-酚比色法分别测定青天葵4种提取物中总黄酮和总酚含有量,通过测定DPPH自由基清除能力、羟自由基清除能力、ABTS自由基清除能力、还原能力评价提取物抗氧化活性,SPSS17.0软件分析抗氧化活性与2种成分含有量的相关性。结果青天葵4种提取物的抗氧化活性均呈量效关系,乙酸乙酯提取物最强,而乙醇提取物最弱。其与总黄酮含有量有一定相关性(0.6040.977)。结论青天葵乙酸乙酯提取物可用于天然抗氧化剂开发,总酚含有量是影响其抗氧化活性的主要因素.IM To investigate the correlations between antioxidant activities and contents of total flavonoids and total phenols of chloroform,ethyl acetate,n-butanol and ethanol extracts from Nervilia fordii( Hance) Schltr.. METHODS The contents of total flavonoids and total phenols in four extracts from N. fordii were determined by Al Cl3-HAc-Na Ac coloration method and Folin-Ciocalteu colorimetric method,respectively. The antioxidant activities of extracts were evaluated by determining scavenging activities of DPPH radical,hydroxyl radical and ABTS radical,together with reducing power. Then the correlations between antioxidant activities and contents of two constituents were analyzed by SPSS17. 0 software. RESULTS The antioxidant activities of four extracts from N. fordii all showed dose-effect relationships,that of ethyl acetate extract was the strongest,while that of ethanol extract was the weakest. They had some correlations with the content of total flavonoids( 0. 604 r 0. 638) and significant correlations with the content of total phenols( r〉0. 977).     目的:对宽叶金粟兰中总黄酮的提取工艺及抗氧化活性进行了研究,旨在为其开发利用提供理论依据。方法:采用微波辅助提取法对宽叶金粟兰中总黄酮的提取工艺进行优化,采用分光光度法测定提取物的还原力,并研究其对羟基自由基、DPPH自由基的清除作用。结果:最佳提取条件为:乙醇体积分数60%、料液比1∶30、微波功率300W,微波处理时间15min;在此条件下宽叶金粟兰的总黄酮提取率为6.11%。在实验浓度(0.2~0.8mg/mL)范围内,宽叶金粟兰黄酮对羟基自由基的清除率可达到100%,对DPPH自由基的清除率达到76.2%,同时还具有较强的还原能力。结论:研究结果表明宽叶金粟兰中的黄酮类物质具有较强的抗氧化活性,该植物具有一定的开发和应用前景。Abstract: Objective: To study the extraction technology of total flavonoids from Chloranthus henryi and its antioxidant activity.Methods: The optimum condition for extracting total flavonoids from Chloranthus henryi by microwave - assisted was obtained through orthogonal experiments.And the antioxidant activity of the extract was evaluated by spectrophotometry on its ability of scavenging hydroxyl free radical(·OH) and 1- diphenyl-2- picrylhydrazy free radical( DPPH·) ,total reducing power.Results: The optimum procedure for microwave- assisted extraction of total flavonoids were ethanol concentration of 60%,ratio of material to solvent of 1 ∶ 30,microwave power of 300W, microwave time of 15min and that the flavonoids yield was up to 6.11%.The scavenging rates of total flavonoids from Chloranthus henryi against hydroxyl free radical and DPPH free radical were found to be 100% and 76.2%,and the extract also had a strong total reducing power at a certain concentration range( 0.2 ~ 0.8mg /mL) .     Chemical Components of Essential Oils from Chloranthus henryi

    目的:分析宽叶金粟兰挥发油的主要化学成分及其抗氧化活性。方法:采用水蒸气蒸馏法获得宽叶金粟兰挥发油,运用气相色谱-质谱联用进行成分分析,同时运用紫外分光光度计测定其对1,1-二苯基-2-三硝基苯肼(DPPH)自由基的清除能力以及总还原能力。结果:从宽叶金粟兰的挥发油中鉴定了21个化合物,占总量的82.30%;挥发油具有较好的还原能力,但对DPPH自由基的清除能力较弱。结论:宽叶金粟兰挥发油主要成分为乙酸冰片酯(45.43%)、3-亚甲基-2-降冰片酮(12.36%)、莰烯(8.74%)等,挥发油具有一定的抗氧化活性。[Abstract] Objective: This article aimed to study the chemical components of essential oils from Chloranthus henryi and their antioxidant activity. Method: The essential oil was extracted by steam distillation, and its constituents were identified by GC-MS. The antioxidant activity of the essential oil was evaluated on its ability of total reducing and scavenging of 1-diphenyl-2-picrylhydrazy ( DPPH) free radical by spectrophotometry. Result: Twenty-one compounds,accounting for 82. 30% of the essential oil,were identified. The ability of scavenging DPPH free radical of the essential oils was weak,while the total reducing ability was strong. Conclusion: The main chemical constituents of the essential oil from C. henryi were bornyl acetate ( 45. 43%) ,3- methylene-2-norbornanone ( 12. 36%) and camphene ( 8. 74%) .
