    A New Species of the Genus Nanhaipotamon (Decapoda:Potamid)

    记述了采自福建省武平县的淡水蟹类南海属Nanhaipotamon一新种武平南海溪蟹N.wupingensissp.nov.本新种头胸甲显著隆起,雄性第1腹肢末部宽大,内侧呈半圆形扩张,外末角呈指状突出,末缘中部明显隆突.蟹体并殖吸虫囊蚴感染率27.3%(3/11).Nanhaipotamon wupingensis sp. nov(Fig.1~8)Holotype:♂,carapace length 22 mm,breadth 27.5 mm;allotype: ♀,length 20 mm,breadth 25 mm;paratypes: 3♂2♀,collected from Xiaba village of Wuping county,Fujian (116°2′N.,24°53′E.);October,1998;195~260 metres above sea level;breadth of the stream 0.5~1.5 metres,pH 6.0.All the type specimens are deposited in Fujian Research Institute of Parasite Disease,Fuzhou,Fujian Province.17 crabs were examined,3 of them infected with the metacercaria of Paragonimus westermani and Euparagonimus cenocopiosus.Carapace distinctly convex, surface smooth, cervical groove shallow.The end of the first pleopod of male,exceeding the suture between 4~5th sternum,Subteminal segment 3.0 times as long as the terminal segment. Terminal part widely,anteroborde mountain peaklike.On side distal end half roundshaped and strongly expanded abdominalinner.The subdistal segment of the second pleopod of male about 2.7 times as long as the distal segment.     目的调查云霄县淡水蟹种类及其感染并殖吸虫囊蚴的情况。方法选取不同地理方位为调查点,捕捉各调查点的淡水溪蟹进行分类和并殖吸虫囊蚴的检查。结果全县共调查6个乡(镇)20个村,共捕捉230只蟹,查出漳州华溪蟹(n=136)、漳浦束腰蟹(n=63)和闽溪蟹(待定种)(n=31)3种;检出感染并殖吸虫蟹15只,阳性检出率为6.6%(15/230),蟹的感染指数为0.11,对福建华溪蟹和中华束腰蟹种类及分布作了修正。结论云霄县并殖吸虫宿主淡水溪蟹有3种,蟹感染并殖吸虫呈下降趋势,但仍为中度并殖吸虫疫源地。Objective To study the distribution of freshwater crabs and their infection by Paragonimus in Yunxiao county, Fujian.Methods Freshwater crabs were collected and identified.The crabs were then dissected and examined for the presence of Paragonimus metacercariae.Results Six tons of crabs from 20 villages (Yunxiao county) were collected.Three species of crabs were found , namely Sinopotamon zhangzhouense, Somanniathelphusa zhangpuensis and Minpotamon spp.The infection rate was 6.6% (15/230) for the Paragonimus, and the index of crab infection was 0.11.Species of freshwater crabs and its geographic distribution were revised for Sinopotamon fukienense and Somanniathelphusa sinensis.Conclusion Freshwater crabs are the intermediate hosts of Paragonimus in the Yunxiao county.科技部自然资源平台项目(2005DKA21104

    SURVEY OF MIXED INFECTIONS OF FISH-BORNE TREMATODES IN PINYUAN COUNTY AND DNA(RAPD)ANALYSIS

    目的 调查广东省平远县棘隙吸虫及其他鱼源性吸虫混合感染情况。方法 对鱼类宿主取鱼鳃和鱼肉等组织 ,镜检囊蚴 ;用水洗过筛沉淀法粪检虫卵 ,部分虫卵阳性动物解剖检查。采用DNA -RAPD技术 ,比较福建棘隙吸虫广东株与福建株。结果 查出华支睾吸虫 (Clonorchissinensis)、东方次睾吸虫 (Metorchisorientalis)、福建棘隙吸虫 (Echinochasmusfujianensis)、日本棘隙吸虫 (E japonicus)、抱茎棘隙吸虫 (E perfoliatus)和钩棘单睾吸虫 (Haplorchispumilio) 6种。第二中间宿主为麦穗鱼等 11种 ,感染率 48 3% (182 / 377) ,有 92 4%的阳性鱼混合感染 2种以上吸虫囊蚴。保虫宿主狗、猫感染率 79 2 % (19/ 2 4) ,每只阳性动物均有 2种以上吸虫混合感染。 15个引物在福建棘隙吸虫广东株与福建株两者共获DNA片段 185个 ,共享度 (F) =0 984,遗传距离 (D) =0 0 16。结论 当地为人兽共患的多种鱼源性吸虫混合感染区。形态学观察和分子生物学检测结果相印证 ,福建棘隙吸虫广东株与福建棘隙吸虫福建株为同一虫种。Aim To seek the possible existence of mixed infections of Echinochasmus fujianensis and other fish-borne tremtodes in Pinyuan county,Guangdong.Methods Metacercariae of trematodes were detected from gills,muscles and some other tissues of freshwater fish and examintd under microscopy Sedimentation pocedures were used to observe parasite ova in stool Adult worms were obtained from definitive animal hosts upon dissection Experimental animal infection were performed from metacercariae of E fujianensis strains in Fujian and Guangdong,and it's DNA analysis Random amplified Polymorphic DNA(RAPD).Results Six species of trematodes were found,including Clonorchis sinensis,Metorchis orientalis,E fujanensis, E japonicus,E perfoliatus and Haplorchis pumilio Eleven species of freshwater fish comprising Pseudorasbora parve were identified as the second intermediate hosts,with an average positive rate of 48 33%(182/377) The overall mixed infection animal were contracted with 2 or more species Dogs and cats were detected to be reservoir hosts,the total infection rate being 79 2%(19/24).92 4% of infecded fish were contracted with 2 or 6 speies of the metacercariae of trematodes 185 DNA fragments were obtained by 15 primers,F=0 894 and D=0 016 between E fujianensis Guandong and Fujian strain Conclusion There are six species of fish-borne trematodes which may cause infections in human and animal in the regions E fujianensis Guangdong stain and Fujian strain are the same species福建省科技计划基金!资助项目 (No 98-Z - 1 56);; 福建省“百千万人才”人选培养资

    采用RAPD技术和等电聚焦电泳方法 ,对福建棘隙吸虫福建株和广东株虫体进行基因组DNA多态性分析和蛋白质等电点测定 .结果表明 ,15个随机引物在两虫株基因组DNA中共获得扩增带 182条 ,二者共有的 89条 ,广东株特有的 4条 ,其共享度为 0 .978,遗传距离指数为 0 .0 2 2 ;两虫株的蛋白质经电泳分离均可获得 9条区带 ,其中有 7条区带的等电点相同 ,第Ⅳ、Ⅴ条蛋白区带的等电点存在差异 .研究表明在福建和广东两地流行的福建棘隙吸虫是同一种属By using RAPD technique and isoelectric focusing electrophoresis method, genomic DNA and protein from Guangdong strain and Fujian strain of Echinochasmus fujianensis were analysed respectively. One hundred and eighty-two polymorphic DNA fragments were generated by RAPD with fifteen primers. Among them, eighty-nine fragments were shared by two strains. Four fragments in Guangdong strain amplified by three primers were absent from Fujian strain. Their haplotype diversity indices and genetic distance were 0.978 and 0.022 respectively. The result indicated that Guangdong strain and Fujian strain were in the same genotype. There were nine protein bands generated by isoelectric focusing electrophoresis in these E. fujianensis. Their protein characters were acidic. Their isoelectric points were in the range of 4.3~6.4. With the exception of two band, all of the bands had neither significant isoelectric point diversity nor visible relative content diversity between E. fujianensis Guangdong strain and Fujian strain. The result indicated that E. fujianensis Guangdong strain and Fujian strain were the same species
