    Bone Proteomic Analysis About Chinese Medicine Action on Rat Glucocorticoid-induced Model of Osteonecrsis.

    目的:用蛋白质组学的研究方法进行激素性骨坏死的病理和中药作用机理的研究,有利于临床治激素性骨坏死。方法:按照常用激素性骨坏死的造模方法,建立大鼠坏死模型,并设立中药治疗组及空白对照组,经过6周处理后经骨形态学检查,确定骨坏死造模成功,处死动物取股骨和肱骨,提取骨组织蛋白质样品,双向电泳分离,得到各组骨组织总蛋白质分子解剖图谱,用图像分析软件,找到各组间差异蛋白质点,进一步行胶内酶切,M AD ITOF/M S质谱分析,得到各差异点的蛋白质肽指纹图谱,结合蛋白质生物信息库(M atrix sc ience L td database),对各蛋白质进行初步鉴定。结论:初步鉴定了3个差异蛋白,分别为阻凝蛋白重链ⅡB、磷脂谷胱甘肽过氧分酶及泛素化酶E 2(MW:17 kd),初步认为这三种蛋白质在激素性骨坏死的发病及中药治疗过程中发挥着重要调控作用。Objective:To study the mechanism about glucocorticoid-induced osteonecrosis and Chinese Medicine action on osteonecrosis in proteomics,it is beneficial to prevention and cure this disease.Methods:Rat was randomly divided into three groups:cortrol group,osteonecrosis model group and Chinese Medicine therapy group.In the present proteomic study ,we characterized the potential effects of glucocorticoids and Chinese Medicine on protein expression in rat bone.Using two-dimensional gel electrophoresis,mass spectrometry,and Matrix science Ltd database,we elementarily identified three variational proteins.Conclusion:Three proteins were identified as proteins similar to Myosin heavy chain ⅡB,Phospholipid hydroperoxide glutathione peroxidase and ubiquitin-conjugating enzyme E2-17kD.These proteins have been documented to be glucocorticoid-relted proteins.These results can provide valuable experimental evidences for the research of the molecular mechanism of osteonecrosis response to glucocorticoids and Chinese Medicine in bone.获得国家自然科学基金资助(30273631,3010244);; 广东省自然科学基金(020778);; 广东省教育厅基金(Z02011

    蛋白质组学是目前研究的重要热点.该文拟建立大鼠骨组织蛋白质样品提取的最佳方法,用于双向电泳分离,为进一步进行蛋白组学分析奠定基础.实验对3种不同方法制备的样本进行同一条件的二维电泳(2- DE)图谱,比较2- DE的结果.结果发现,丙酮沉淀法最佳.这些结果提示,只有结合骨组织成分特征与双向电泳的具体要求,选择恰当的样品提取液,采用合理的提取步骤,获得蛋白质样品,才能得到清晰高分辨率的电泳图谱,满足蛋白质组学分析需要.Proteomics is a challenging field that has been growing rapidly in the postgenomic era.Protein preparation is a key procedure for two-dimensional electrophoresis (2-DE) proteomics.In the current study,three procedures of protein sample preparation,namely basic,dialysis and acetone methods,were compared for establishment of an optimal method to extract proteins from rat bone,based on 2-DE maps from protein samples prepared by the three methods.Of the three methods,acetone method is the best one.Using this method to purify proteins from rat bone,high quality of peptide mass fingerprint can be achieved.The procedure of the method is as follows.Prepared bone (1.0 g) was placed in an earthen bowl to grind into powder in liquid nitrogen and then 3 mL ice-cold buffer (1.44 g urea,0.46 g sulfocarbamide,0.12 g CHAPS,0.047 g DTT,0.015 g Tris-base,30 μL 100 mmol/L PMSF) was added to it.After stored at 4℃ overnight,these samples were centrifuged at 12 000 g for 30 min.The supernatant was dialyzed against 0.01 mol/L PBS for 24 h.Then ten times of volumes of cooled acetone were added to the specimen for the concentration of proteins.Followed by the store at -20℃ for 2 h,the proteins were obtained by centrifugation and then dry at 4℃.Before the isoelecric focusing,the sample was redissolved in the buffer (7 mol/L urea,4% CHAPS,100 mmol/L DTT,40 mmol/L Tris,2 mol/L sulfocarbamide,0.5% ampholyte (pH 5~7),1 mmol/L PMSF) at the room temperature for 2 h.The results may provide a valuable method of sample preparation for study of bone proteomics.国家自然.3010244);; 广东省自然科学基金(020778);; 广东省教育厅基金(Z02011)资

    日光诱导叶绿素荧光估算中国典型生态系统总初级生产力的能力

    陆地生态系统总初级生产力(GPP,Gross Primary Productivity)是陆地生态系统碳循环的重要分量,提高其估算精度具有重要的科学意义。由于受多种因子的影响,GPP的时空变异明显,其估算结果存在较大的不确定性。日光诱导叶绿素荧光(SIF,Sun-Induced Chlorophyll Fluorescence)与GPP密切相关,近年来被应用于估算区域和全球GPP,但其在中国生态系统的适用性尚不清楚。以中国8个典型植被生态系统为研究对象,驱动两叶光能利用率模型(TL-LUE,TwoLeaf Light Use Efficiency Model)模拟以站点为中心0.5°×0.5°区域内的月GPP,验证SIF估算GPP的能力。结果表明,SIF具有监测中国典型植被生态系统GPP的能力,月SIF与TL-LUE模拟的月GPP之间显著相关,其中5个生态系统中两者的R2高于0.8,最高达到0.91,GPP与SIF变化的斜率随生态系统类型变化。模拟的GPP与SIF遥感数据的季节变化特征相同,两者之间的一致性在生长季节好于非生长季节;SIF能更好地监测农田GPP的季节变化
