    Aβ1-42诱导的小胶质细胞炎性反应对神经元细胞Mecp2蛋白表达的影响

    目的:体外研究Aβ1-42诱导的小胶质细胞炎症反应对海马神经元细胞甲基化Cp G结合蛋白(Mecp2)表达的影响。方法:(1)用不同浓度的Aβ1-42(终浓度分别为0,5,10,20,30μmol/L)处理小胶质细胞系(N9),24h后取上清液,ELISA检测每组N9细胞培养上清中炎症因子TNF-α、IL-1β的含量,选择合适的Aβ1-42的终浓度。(2)用Aβ1-42(终浓度为20μmol/L)及TLR4拮抗剂TAK-242(终浓度为1μg/ml)处理小胶质细胞系(N9),分为3组:正常对照组(con组)、单纯Aβ1-42处理组(Aβ1-42组)、Aβ1-42处理后TAK-242干预组(TAK-242组)。24 h后取上清液,ELISA检测每组N9细胞培养上清中炎症因子TNF-α、IL-1β的含量。(3)实验分组同(2),24 h后收集培养液,分别干预生长良好的海马神经元细胞系(HT-22)。24 h后,通过Western Blot、免疫荧光染色方法检测每组HT-22细胞中Mecp2蛋白的表达。结果:(1)ELISA结果显示:与con组相比,Aβ1-42处理后,N9细胞分泌的TNF-α、IL-1β的表达量随Aβ1-42浓度的增加而增加(P<0.05)。(2)ELISA结果显示:与con组相比,Aβ1-42组的TNF-α、IL-1β的表达水平明显提高(P<0.05);TAK-242组的细胞分泌TNF-α、IL-1β水平较Aβ1-42组显著降低(P<0.05)。(3)Western Blot结果显示:与con组相比,Aβ1-42组HT-22细胞中Mecp2蛋白表达明显增加(P<0.05);与Aβ1-42组相比,TAK-242组HT-22细胞中Mecp2蛋白表达明显减少(P<0.05)。(4)免疫荧光染色结果显示:与con组相比,Aβ1-42组HT-22细胞中Mecp2蛋白表达明显增强;而与Aβ1-42组相比,TAK-242组HT-22细胞中Mecp2蛋白表达减弱。结论:Aβ诱导的小胶质细胞发生炎性反应,导致神经元的损伤,可能是由于这种炎性反应使得神经元细胞中Mecp2蛋白的表达量增加,从而损伤神经元。国家自然科学基金(31570845);国家自然科学基金青年项目(81501207);;陕西省自然科学基础重大研究项目(2016ZDJC-16

    Studies on chemical constituents in ethanolic extract from Acanthus ilicifolius as a pharmaceutic mangrove

    目的探讨老鼠簕的化学成分。方法将老鼠簕全株粉碎后,用95%乙醇浸提3次,合并浓缩,浸膏的石油醚部位经反复硅胶柱层析分离,结合波谱学数据鉴定化合物结构。结果从石油醚部位分离得到8个化合物,分别为β-谷甾醇(Ⅰ)、豆甾醇(Ⅱ)、正十六烷酸(Ⅲ)、正二十八烷酸(Ⅳ)、4,22-二烯-3-酮豆甾烷(stig-masta-4,22-dien-3-one)(Ⅴ)、4-烯-3-酮豆甾烷(stigmast-4-en-3-one)(Ⅵ)、5,22-二烯-7-酮-3β-羟基豆甾烷(3β-hydroxystigmasta-5,22-dien-7-one)(Ⅶ)、2-唑啉酮(2-benzoxazolinone)(Ⅷ)。结论其中Ⅴ,Ⅵ,Ⅶ为首次从该植物中分离得到。Objective To study the chemical constituents of Acanthus ilicifolius.Methods The air-dried and powdered plant material was extracted with 95% ethanol.Silica gel and Sephadex LH-20 column chromotography were used in the isolation from the ethanol extracts of the whole plant,the compounds were determined on the basis of various modern spectroscopic analyses and physical constants.Results Eight compounds were isolated from the petroleum ether soluble portion,identified as β-sitosterol(Ⅰ),stigmasterol(Ⅱ),n-hexadecane acid(Ⅲ),n-octacosanic acid(Ⅳ),stigmasta-4,22-dien-3-one(Ⅴ),stigmasta-4-en-3-one(Ⅵ),3β-hydroxystigmasta-5,22-dien-7-one(Ⅶ) and 2-benzoxazolinone(Ⅷ).Conclusion CompoundⅤ,Ⅵ,Ⅶ were obtained from the genus for the first time.国家自然科学基金(30671646);; 福建省科技计划资助项目(2003Y036

    Thermodynamics of Basic Compounds on Novel Stationary Phase Embed Polar Groups

    以苯、吡啶、尼古丁和普鲁卡因为探测因子,考察了酰胺型、胺型、醚型和酯型4种“包埋”极性官能团的新型键合固定相对碱性化合物在流动相pH值分别为3·0和7·0时的热力学焓变。实验结果表明,新型固定相由于烷基分子链中“嵌入”极性官能团,因此在流动相pH为3·0时温度对碱性溶质的保留影响相互差异较大。在流动相pH为7·0时,根据Van′t Hoff方程计算出4种待测物质从流动相转移到固定相上的ΔH0,从ΔH0的绝对值大小、相关系数以及实际的分离效果可以发现,在分离强碱性物质时,若实验条件一样,则分离效果是酰胺型和胺型>酯型固定相>醚型固定相,但它们均优于传统的固定相。The thermodynamis character of three basic probes and a neutral reference compound were investigated as a function of temperature using four novel high performance liquid chromatography columns that are amide-type、 amine-type and ester-type stationary phases at both pH 3.0 and pH 7.0. The results showed that considerable differences of retention for bases at pH 3.0 might result from the existence of polar group in the long chain alkyl chain on the packing. The ΔH~0 values of four probes on four novel columns and a conventional C_8 column were calculated according the van Deemter curves. The separation results of basic samples on the novel columns and C_8 column shold amide-type and amine-type exceed ester-type and ether-type, but their all exceed conventional C_8 column according to absolute values of ΔH~0 , correlation coefficient and practical separation.厦门大学引进人才启动基金资
