    Study on solid-state fermentation and purification of cellulase from Penicillium sp.T24-2

    青霉T24-2是一株具有较高固态发酵产酶能力的菌株。本文通过对青霉T24-2的固态发酵条件进行研究,确定了该菌株的最佳产酶条件为:固体培养基蔗麸比为4:6,添加0.2%尿素为氮源,按10%接种量接种种龄为18h的菌悬液,每250mL三角瓶中固体培养基装瓶量30g(干重10g),pH自然,30℃下培养3d。最佳糖化条件为:曲料比1:3,加水比1:6,在50℃,170r/min条件下糖化18h,最高糖化率可达32.4%,每100g甘蔗渣糖化液经酿酒酵母发酵后,酒精产量可达11.3g。 甘蔗渣经4%二甲基亚砜预处理后,制曲并糖化,CMCase和FPA酶活分别可达219.38U/g和16.07U/g...Penicillium sp.T24-2 is a high cellulase-producing strain for solid-state fermentation.     以头肾细胞为材料,采用PHA体内注射,空气干燥法制片,分析了双棘黄姑(Nibeadiacanthus)的染色体组型.结果表明:其染色体组型为:2n=48=24t,NF=48.在已进行过染色体组型分析的石首鱼类中,除大黄鱼外,染色体组型均为2n=48t.The karyotypes of Nibea diacanthus were examined in the paper.The samples were separately injected by PHA and colchicines.The chromosomes were received from the renal tissue by using air drying method and Giemsa staining.The result showed that the chromosome number of Nibea diacanthus was 48;and its karyotype formula was 2n=48=48 t,NF=48;it was the same as the other croakers determined,except Pseudoceana crocea in Sciaenidae.福建省自然科学基金(B0110004)资助细胞数目938770

    一种新型微孔聚合物电解质的制备与表征

    以介孔分子筛SBA-15为填料,探索出一种无需使用增塑剂制备微孔型聚合物电解质的新方法.当SBA-15︰PVdF-HFP(偏氟乙烯-六氟丙烯共聚物)=3︰8时,所制备的复合固体聚合物电解质(CSPE)室温电导率达0.30mS·cm~(-1).红外透射光谱显示,成膜液中溶剂分子和SBA-15分子筛之间的相互作用所引发的相分离导致微孔的形成.X射线衍射(XRD)结果表明,SBA-15分子筛的加入,降低了聚合物电解质的结晶度,而SBA-15分子筛仍保持原有的骨架结构.所组装的Li/CSPE/MCF电池具有较为平稳的充放电平台,首次循环的库仑效率约为85.0%,经过20次循环后,容量保持率为94.0%

    采用尾静脉抽血法对斜带髭鲷厦门网箱养殖群体的染色体核型进行了研究,结果显示其核型公式为2n=46=4m+8sm+6st+28t,染色体总臂数NF=58,与喻子牛等对青岛近海斜带髭鲷的研究结果存在差异.通过对比发现厦门斜带髭鲷染色体2倍体数2n=46比青岛的少1对,但大型中部着丝点染色体数目又比青岛海域群体多出一对.从而推断斜带髭鲷厦门网箱养殖群体的染色体核型2n=46=4m+8sm+6st+28t可能是从核型2n=48=2m+8sm+2st+36t通过一对非同源的端部着丝点染色体经罗伯逊易位,着丝点融合形成了一中部着丝点染色体而形成的,进而造成斜带髭鲷不同地理种群间染色体的多态现象.The karyotypes of cultured Hapalogenys nitens from Xiamen stock were examined from the renal tissue.The number of chromosomes is 2n=46,which include two pair of mid-centromere chromosome,four pair of sub-mid dle-centromere chromosome,three pair of sub-top-centromere chromosome and twenty-eight top-centromere chromosome.It differs from the usual number of chromosomes of saltwater fish,which is 2n=48. The karyotypic formulae is:2n=46=4m+8sm+6st+28t,which was different from the result of Qingdao coastal water population:2n=48=2m+8sm+2st+36t.The number of diploid chromosomes of Xiamen stock cultured population was one pair less than Qingdao coastal water population,but the number of large mid-centromere chromosome of Xiamen population was one pair more than the other one.One of the reasons for this different might be the polymorphism.It might be that the centromeres of one couple of dis-homologous top-centromere chromosome of the Qingdao coastal water population amalgamated into one midcentromere chromosome,and the karyotypic formulae which is:2n=48=2m+8sm+2st+36t turned into 2n=46=4m+8sm+6st+28t,and polymorphism came into being between the two different groups of Hapalogenys nitens.The number of sub-top-centromere chromosomes and top-centromere chromosomes was more than that of the sub-middle-centromere chromosomes and mid-centromere chromosomes,indicating that Hapalogenys nitens was more advanced than most another populations in Perciformes.国家自然科学基金(30070595);; 福建省自然科学基金(B0010003)资

    采用PHA结合秋水仙素活体注射,取条纹斑竹鲨(Chiloscyllium plagiosum)脾脏经低渗处理,空气干燥制片法制作染色体标本,对染色体进行Giemsa染色,完成条纹斑竹鲨染色体核型分析.研究结果表明,条纹斑竹鲨的染色体组由20对中部着丝粒染色体、12对亚中部着丝粒染色体、5对亚端部着丝粒染色体和14对端部着丝粒染色体构成,染色体核型公式为:2n=102=28t+10st+24sm+40m,染色体臂数NF=166,其中1对亚中部着丝粒染色体带有明显的次缢痕.与其他软骨鱼类相似,条纹斑竹鲨同样具有数目众多的染色体二倍体数,但以中部着丝粒和亚中部着丝粒染色体为主.The chromosome specimens of the lip shark Chiloscyllium plagiosum were obtained from the spleen cell by vivo injection of PHA and cultivation of colchicines,hypatoic-air drying technique,and then,by studying their Giemsa stain,the analysis of the karyotype was completed.According to the chromosome centromeric index values found,the karyotype of C.plagiosum is composed of 20 metacentric,12 metacentric-submetacentric,5 telocentric-subtelocentric and 14 telocentric pairs,and the karyotype formula is:2n=102=28t+10st+24sm+40m.NF=166,and there was one pair of metacentric-submetacentric chromosomes with secondary constriction.Compared to the other cartilaginous fishes,C.plagiosum had high diploid chromosome number,most of which were metacentric and metacentric-submetacentric chromosomes.国家自然科学基金(40776083,40876080);; 厦门市科技项目(3502Z20063022)资

    Antigenicity Analysis of V.harveyi TS-628 Strain

    哈维氏弧菌(Vibrio harveyi)是海水鱼虾养殖中常见的致病菌,由该菌引发的病害给世界各地的养殖业带来重大经济损失,但对其有效抗原的筛选或相关疫苗的研究报道相当少,对其病害的防治也尚无有效措施.本文以患病青石斑鱼分离到的病原菌哈维氏弧菌TS-628菌株为研究对象,分别提取它的鞭毛蛋白、脂多糖(LPS)和外膜蛋白(OMP),并采用Western blot技术分析检测这3种成分的抗原性.结果显示,鞭毛蛋白主要的免疫印迹带约有4条,其中435、2 ku为主要免疫反应显色带;OMP主要的免疫印迹带约有5条,其中43 ku为最主要免疫反应显色带,35、38、47和52 ku也具有较强的免疫显色反应.LPS没有检测到免疫印迹反应.这一研究结果将为灵敏检测哈维氏弧菌以及研制高效疫苗奠定基础.Vibrio harveyi,the major causal agent of vibriosis,affects a diverse range of marine cultured organisms over a wide geographical area,while the reports about screening the effective antigen and the researches on vaccines of V.harveyi are scarce.Flagellin,lipopolysaccharide(LPS) and outer membrane proteins(OMP) are major immunogenic antigens in many Gram-negative bacteria.In this study,the flagellin,OMP and LPS of V.harveyi TS-628 strain isolated from the groupers infected by it were extracted and Western blot analysis was used to detect the antigenicity of these extractions.The results of the Western blot assay revealed that there were 4 positive flagellin bands about 35,38,43 and 52 ku,of which the 43 and 52 ku bands displayed the strongest positive reaction.There were 5 positive OMP bands about 35,38,43,47 and 52 ku,of which the 43 ku appeared the strongest positive reaction and the other four proteins also displayed strong reaction.However the LPS was Western blot-negative.These results indicated that the 43 and 52 ku flagellin and the OMP such as 43,52 ku proteins could be the candidates to develope vaccines against V.harveyi.国家“863”计划(2003AA603011);; 福建省科技项目(2002N009)资
