    擬球藻養殖、類胡蘿蔔素萃取、及高含量玉米黃 素奈米粉粒體製備

    本研究旨在開發微藻玉米黃素的保眼營養補充原料,並製備含高純度玉米黃素的奈米粉粒體。實驗以海水養殖的擬球藻,經過超音波攪拌丙酮萃取,獲得每克擬球藻藻粉含3.613毫克的玉米黃素。隨後以小型逆相樹脂劃分超音波丙酮萃出物,可得固形物含41.0 %玉米黃素且回收率為82.3 %。大型管柱層析純化,可得42.6%玉米黃素固形物且回收率達82.7%。再以超臨界二氧化碳抗溶結晶1.5 mg/ml近飽和的乙酸乙酯溶液,得659.3 mg/g玉米黃素沉澱物,但是抗溶2.0 mg/ml近飽和的丙酮溶液,僅得479.0 mg/ml。當以應答曲面實驗設計超臨界二氧化碳抗溶乙酸乙酯溶液的造粒程序時,探討進料濃度、二氧化碳流速、抗溶壓力,分別對玉米黃素純度、產率、回收率及沉澱顆粒粒徑的影響,並發現以進料濃度1.5 mg/ml、二氧化碳流速48.6 g/min及壓力135 bar的超臨界抗溶結晶乙酸乙酯溶液,可得841.7 mg/g玉米黃素沉澱物且回收率達85.3 %,此沉澱顆粒的平均粒徑介於103至327 奈米之間。屬於不規則的薄片顆粒堆疊,降低壓力有助於鬆散堆積程度。 英文摘要 This work investigated supercritical carbon dioxide (CO2) anti-solvent recrystallization of zeaxanthin-containing particulates from microalgal Nannochloropsis Oculata species. The extract of the algae containing 3.613 mg/g zeaxanthin using ultrasonic stirred acetone extraction subjected to column elution fractionation for increasing the amount of zeaxanhin in the fraction to 425.6 mg/g by weight with the recovery of 82.7 %. Supercritical anti-solvent (SAS) precipitation of the column fraction at 215 bar, 50 oC, 36 g/min of CO2 flow rate, 0.5 ml/min of feed solution containing, 1.5 mg/ml of feed concentration results in 659.3 mg/g of zeaxanthin in the precipitates. However, another SAS precipitation of acetone solution at the 2.0 mg/ml of feed concentration yields 479.0 mg/g of zeaxanthin. A three-factor center composited schemed response surface method by changing feed concentration, CO2 flow rate, and pressure designed for the SAS precipitation was carried out in finding total yield, purity, recovery and particle size of the precipitates. The highest purity is 841.7 mg/g of zeaxanthin with 85.3 % recovery, obtained at feeding concentration of 1.5 mg/ml, CO2 flow rate of 48.6 g/min and pressure of 135 bar.     应用 2 0条Sangon的随机引物 ,对6 0 Coγ射线辐照诱变所获新株系漳蕉 8号(原漳农 8号 )及其对照品种台湾北蕉 (MusAAAGrandCavendish)的总DNA进行RAPD ,结果发现有 1 2条引物扩增出 68条谱带 ,其中同源带 53条 ,占总带数的77 9% ,差异带 1 5条 ,占总带数的 2 2 1 % ;漳蕉 8号与对照多态性差异高达 2 4 6% ,且显示出稳定性遗传。其中S10 和S19的 3条差异带 ,可作为漳蕉 8号株系鉴定和防退化复壮的筛选标记。Zhangjiao No.8,a new variety induced from irradiation by 60 Co γ rays,was analyzed by RAPD using 20 random primers against Mus AAA Grand Cavendish. The result showed that 68 bands could be amplified by 12 primers,53 were homologous,accounted for 77 9% of total bands; only 22 1% were differential bands,the polymorphism difference up to 24 6% between Zhangjiao No.8 and CK,indicating its stable heredity.Three differential bands from S 10 and S 19 could be used as markers for identification and anti degeneration of Zhangjiao No.8
