    Isolation of microsatellite markers from Porphyra haitanensis and their application in genetic diversity of Porphyra

    坛紫菜是一种营养价值很高的海藻,在福建省有悠久的栽培历史。运用分子生物学技术分析坛紫菜种质资源的遗传多样性,寻找相关的分子标记,可以指导培育实践,加快育种进程。本实验首先构建了部分坛紫菜小片段基因组文库,从中筛选到1500左右个阳性克隆,在215个阳性克隆中筛选到4个微卫星位点。选择其中的2个微卫星位点ATN4和GAN5以及从GeneBank里搜索的江蓠的3个微卫星位点AJ512812、AJ512806和AJ512804对9个紫菜样品(LYG、RD、CS、GL1、DXY、BBD、JJS、NH和LSD)进行PCR扩增。5个微卫星位点共检测到31个DNA条带,其中条带最多的位点是AJ512812,...Porphyra haitanensis is seaweeds of high nutiitive value, and it has a long history of culture in Fujian province.     作者简介:陈骁(1979–),男,江西上饶人,硕士研究生; 陈奕欣,通迅作者,教授,从事遗传育种及分子生物学研究, E–mail:[email protected][中文文摘]用随机多态DNA(RAPD)标记方法对11个紫菜(Porphyra)样品进行遗传多样性检测,从46个随机引物中筛选出27个有效引物,共扩增出282条DNA带,其中多态位点224个,占79.4%。用类间平均链锁法对各样品间的遗传距离进行聚类分析。结果显示,2个条斑紫菜之间的亲缘关系很近,遗传相似系数达98.1%,和坛紫菜的亲缘关系较远;坛紫菜不同样品间的遗传相似系数在75.8%~90.6%之间,表明坛紫菜种质资源具有丰富的遗传多样性,是良种选育的遗传基础。[英文文摘]The genetic diversity of 11 Porphyras samples were confirmed by random amplified polimorphic DNA(RAPD) markers. 27 primers were screened from 46 arbitrary primers, and a total of 282 DNA bands were amplified, among them 79.4% were polymorphic. The genetic data were analyzed by means of similarity, in a dendrogram was using between groups linkage method.     Mutagenesis on protoplast of Porphyra haitanensis by ~(60)Co-γ

    利用60Co-γ射线对坛紫菜(Porphyrahaitanensis)原生质体进行辐照诱变,发现原生质体对60Co-γ射线有极强的耐受性。前期的培养结果表明,原生质体是一种理想的诱变材料。诱变结果表明,60Co-γ射线对坛紫菜可诱变剂量范围很大,从200~1300Gy均有突变株生成。诱变后细胞的存活率不宜作为坛紫菜诱变最佳剂量的指标。60Co-γ ray was used to induce mutant on protoplast of Porphyra haitanensis.The results showed that the protoplasts of P.haitanensis could survive under a wide range of γ-rays.Primary culture showed that the protoplast was suitable for mutagenesis.Further results also showed that mutants could be induced in a very wide range of dosages of 60Co-γ ray.The mortality was not a suitable parameter for the evaluation of the mutant-induction efficiency in P.haitanensis.国家863计划项目(2002AA603023);; 福建省重大农业科技项目(2001Z017

    <正> 一、绪言1958年9月—1959年9月在湖北黄石市北郊花马湖(鄂城县境)进行湖泥施肥试验,在室内、室外先后试验了35次,历时343天,在党的正确领导和全体工作人员的积极努力下,利用湖泥施肥的方法获得了成功。湖泊底泥里面,有腐烂的水草碎片、浮游植物及水生动物的尸体残骸等淤积;加之,由于雨水将地面冲下来的表层富有机质肥土也沉积于底部,因此,一般湖泊的底泥都很肥,常厚达0.5—1.0米以上;pH 值介于5.2—7.0间;无机酸度(以甲基橙作指示剂)为零,总酸度(以酚酞作指示剂)介于0.16—0.46毫
