坛紫菜是一种营养价值很高的海藻,在福建省有悠久的栽培历史。运用分子生物学技术分析坛紫菜种质资源的遗传多样性,寻找相关的分子标记,可以指导培育实践,加快育种进程。本实验首先构建了部分坛紫菜小片段基因组文库,从中筛选到1500左右个阳性克隆,在215个阳性克隆中筛选到4个微卫星位点。选择其中的2个微卫星位点ATN4和GAN5以及从GeneBank里搜索的江蓠的3个微卫星位点AJ512812、AJ512806和AJ512804对9个紫菜样品(LYG、RD、CS、GL1、DXY、BBD、JJS、NH和LSD)进行PCR扩增。5个微卫星位点共检测到31个DNA条带,其中条带最多的位点是AJ512812,...Porphyra haitanensis is seaweeds of high nutiitive value, and it has a long history of culture in Fujian province.
