坛紫菜是一种营养价值很高的海藻，在福建省有悠久的栽培历史。运用分子生物学技术分析坛紫菜种质资源的遗传多样性，寻找相关的分子标记，可以指导培育实践，加快育种进程。本实验首先构建了部分坛紫菜小片段基因组文库，从中筛选到1500左右个阳性克隆，在215个阳性克隆中筛选到4个微卫星位点。选择其中的2个微卫星位点ATN4和GAN5以及从GeneBank里搜索的江蓠的3个微卫星位点AJ512812、AJ512806和AJ512804对9个紫菜样品（LYG、RD、CS、GL1、DXY、BBD、JJS、NH和LSD）进行PCR扩增。5个微卫星位点共检测到31个DNA条带，其中条带最多的位点是AJ512812，...Porphyra haitanensis is seaweeds of high nutiitive value, and it has a long history of culture in Fujian province. 坛紫菜是一种营养价值很高的海藻，在福建省有悠久的栽培历史。运用分子生物学技术分析坛紫菜种质资源的遗传多样性，寻找相关的分子标记，可以指导培育实践，加快育种进程。 本实验首先构建了部分坛紫菜小片段基因组文库，从中筛选到 1500左右个阳性克隆，在 215个阳性克隆中筛选到 4个微卫星位点。选择其中的 2个微卫星位点 ATN4和 GAN5以及从 GeneBank里搜索的江蓠的 3个微卫星位点 AJ512812、AJ512806和 AJ512804对 9个紫菜样品（LYG、RD、CS、GL1、DXY、BBD、JJS、NH和 LSD）进行 PCR扩增。5个微卫星位点共检测到 31个DNA 条带，其中条带最多的位点是 AJ512812，有 12 条；条带最少的位点是AJ512804，只有 1条带，其它位点扩增的条带数在 4～8之间。在所有的 31条带种有 26条具有多态性，占条带总数的 83.9%。根据微卫星检测的结果，采用类间平均链锁法（Between groups linkage）对各样品间的 Nei氏遗传相似系数进行聚类分析。分析结果显示野生坛紫菜样品 NH、JJS和 LSD之间的遗传相似系数最高，达到 96.8％；坛紫菜各样品间的遗传相似系数在 58.1％～96.8%之间，不同海区、栽培样品与野生样品之间存在很大的遗传差异。这表明坛紫菜的种质资源遗传多样性丰富，将为坛紫菜的良种选育奠定基础。从聚类树状图可以看出，9个紫菜样品共聚成两大类，坛紫菜样品 NH、JJS和 LSD首先聚为一类，再和其它 4个种坛紫菜样品聚集，条斑紫菜样品LYG和 RD先聚在一起，最后与坛紫菜样品相聚。不同海区的两种养殖坛紫菜CS和 GL1之间的遗传距离甚至大于坛紫菜与条斑紫菜之间的遗传距离，说明地理环境不同会造成紫菜在遗传上的较大差异，有时甚至大于种间的遗传距离。 5个微卫星位点在 9个紫菜样品中共扩增出 6个特异性条带。条斑紫菜厦门大学博硕士论文摘要库摘要 2   LYG 的特异性条带 ATN4-1140 和 ATN4-175；养殖坛紫菜 CS Abstract Porphyra haitanensis is seaweeds of high nutiitive value, and it has a long history of culture in Fujian province. In this thesis, we first constructed a small-size genomic library of P.haitanensis and got 1500 positive recombinants.Among which we obtained 4 microsatellite sequences from 215 positive clones.Choosing 2 of the microsatellite loci in P.haitanensis and 3 microsatellite loci of Gracilariopsis(AJ512812、AJ512806 and AJ512804) from GenBank as primers.9 DNA samples(LYG、RD、CS、GL1、DXY、BBD、JJS、NH和 LSD)of Porphyra were amplified,and a totle 31 DNA bands were detected. AJ512812 amplified 12 DNA bands were the most while AJ512804 only amplified 1 DNA bands, which was the least of them. Among 31 DNA bands, 83.9% were polymorphic. The data was analyzed by means of Nei’s similarity coefficient, and a phenogram was constructed by using between groups linkage method. The result showed that the similarity coefficient of the 7 P. haitanensis from 58.% to 96.8%.The highest similarity coefficient is 96.8% which among 3 wide P.haitanensis.The results indicated a high genetic varitions of P.haitanensis,and make a genetic foundation for excellent breed select. According to the dendrogram we can find that the 9 Porphyra were divided into two teams. The 7 P.haitanensis samples had closely relationship in the same team and the 2 P.yezoensis had closely relationship. However Nei’s genetic distance between CS and GL1 was larger than that between P.haitanensis and P.yezoensis.It indicated that different geographical localities where the sample were 厦门大学博硕士论文摘要库摘要 4  collected tend to make the genetic diversity more obviously, sometimes it maybe the distance between two species. In this thesis, we obtain 6 specific bands,ATN4-1140 and ATN4-168 of P.yezoensis LYG； AJ512812-1214、AJ512812-360、AJ512806-404 and AJ512806-313 of excellent strain CS of P.haitanensis. We repeated the experiment for several times and these specific bands appeared stable. Forther study on the linkage between specific bands and excellent trait will be hopefully for molecular gentic breeding of P.haitanensis. Keyword: Porphyra haitanensis；Microsatellite；Genetic marker     厦门大学博硕士论文摘要库 前言  5  一、前言 1遗传标记技术概述 遗传标记[1][2]（Genetic makers）是指用来区分不同的个体或群体，能够稳定遗传的物质或性状。它是基因型的特殊的易于识别的表现形式，是生物分类学、育种学、遗传学和物种起源与进化等研究的主要技术指标之一。理想的遗传标记主要应具备以下几条标准[3][4]：1）具有较强的多态性；2）表现为共显性；3）选择中性（即无基因多效性）；4）对主要的性状无影响；5）经济方便易于观察记载。 随着生物学技术的不断发展，遗传标记技术也随之不断发展，最早的遗传标记是形态标记（Morphological markers）[5][6][7]，仅限于易于识别的形态、生理和生化特性，如颜色、形状、大小、生理功能缺陷等。 20世纪 60年代开始，随着电泳技术的发展，多态蛋白作为遗传标记在研究中得到广泛的应用，但由于可分析的蛋白种类少、变异性低，限制了蛋白质作为遗传标记的发展和应用[3]。以上标记方法都属于基因表达型的标记，易受环境条件和发育阶段的影响，可以利用的多态位点少，存在很大的局限性。后来在显微镜技术的支持下诞生了细胞学标记（Cytological markers）[8]，该标记是指染色体核型（染色体的数目、大小、着丝粒的位置等）和带型（G带、C带、N带等）的标记。 从 20世纪 80年代开始，由于 DNA重组、限制性内切酶消化、分子杂交、DNA序列快速分析、PCR等一系列分子生物学实验技术的建立，人们能够比较方便的分析检测遗传物质 DNA 的多态性，从而出现了多种多样的建立在DNA水平上的分子遗传标记（Molecular genetic markers）[2] [8]。分子遗传标记突破了表达型标记的局限性，其变异只来源于 DNA序列的差异，具有稳定性高、受环境影响较小、信息含量高、不同层次和类群之间广泛可比等优越性，因而成为遗传标记中又一个强有力的工具。厦门大学博硕士论文摘要库 前言  6  在人类基因组计划的推动下，分子遗传标记的研究与应用得到了迅速的发展。已经建立了限制性酶切片段长度多态性（Restriction fragement length pymorphism，RFLP）[9]、单链构型多态性（Single strand conformation pymorphism，SSCP）[10]、小卫星 DNA（Minisatellite DNA）[11]、微卫星 DNA（Microsatellite DNA）[12]、随机扩增多态性 DNA（Random amplified polymorphic DNA，RAPD）[13] [14]、扩增片段长度多态性（Amplified fragment length polymorphism，AFLP）[15]、线粒体 DNA（Mitochondrial DNA，mtDNA）[16]、表达序列标签（Expressed sequence tags，ESTs）[17]等技术。表 1简单概括了目前常用的分子标记技术的原理及其优缺点。 2微卫星的简介 2.1 微卫星的发现 生物学家在 20世纪 70年代就已经知道真核基因组中存在着短串联重复位点，但直至 1982 年 Hamada[12]等发现从酵母到脊椎动物中的多拷贝多聚（TG）n时，这些序列的数目之大和存在之广泛才显现出来。这一发现后来被 Tautz 和 Renz[18]所证实：它们用不同微卫星序列与不同有机体中的基因组 DNA 杂交发现存在着许多类型的简单重复序列。1985 年，Jeffreys[19]等发现了人基因组中有更长重复单元的串联重复序列，即小卫星 DNA（Minisatellites）。与微卫星一样，小卫星的串联重复单位的数目也显示出可变性，因此二者被统称为可变数目串联重复位点（Variable number of tandem rapeats，VNTRs）[20] [21 ]。但在应用方面，小卫星明显逊于微卫星。厦门大学博硕士论文摘要库Degree papers are in the “Xiamen University Electronic Theses and Dissertations Database”.
