V rámci praktické části je popsána výsledná aplikace, která na základě vytvořeného uživatelského profilu nabízí služby, jako jsou personalizované vyhledávání, doporučování obsahu a ideální průchod konferencí s paralelními sekcemi.This paper describes personalization of web applications and options of usage for scientific web portals. Theoretical part introduces main personalization principles and methods. Practical part of this paper focuses on resulting application. The application provides personalized services based on created user profile such as personalized search, content recommendation and conference planner.
Effects of Ultrasound Power, Temperature and Flow Rate of Solvent on Decontamination of Sensitive Equipment by Extraction
The solvent extraction process is regarded amongst other known methods to be applicable for decontamination of sensitive equipment components, especially in cases the components are contaminated in-depth with chemical warfare agents. Viability of the solvent extraction method was evaluated on coupons of butadiene rubber contaminated by sulphur mustard before decontamination by the solvent extraction. The contaminated coupons were extracted in a flow cell, which the solvent (ethoxynonafluorobutane) passed through. Three following specific operational factors, namely the temperature, the flow rate, and the power of ultrasound bath, were assessed for the extent of influencing upon the respective observed extraction efficiencies. Analytical techniques for determination of mycotoxins in barley, malt and beer: A review
The increasing interest in defining the content of mycotoxins is related to the development of methods for their determination. Several methods of determination have been published. This paper describes analytical procedures such as (a) sample preparation – sampling and homogenization, (b) extraction and purification – solid phase extraction (SPE), a QuEChERS method (acronymic name from quick, easy, cheap, effective, rugged and safe), solid-liquid extraction (SLE) and immuno affinity extraction (IAE). It also provides a review of, (c) instruments, and other analytical methods such as thin layer chromatography (TLC), enzyme linked immune sorbent assay (ELISA) and liquid chromatography (LC), gas chromatography (GC) with different detectors for determination of mycotoxin in barley, malt and beer are discussed
Assessing earlywood-latewood proportion influence on tree-ring stable isotopes
Tree-ring stable isotopes are typically measured in latewood cellulose to mitigate potential carry-over effects from previous year storage pools. The isotopic composition of individual tree-ring segments is thought to include considerable intra-annual variability. This sampling strategy may be complicated by steep intra-annual isotope gradients that can rival the inter-annual variability, however. Consistent sampling of latewood material may not always be possible due to low sample availability or high prevalence of narrow rings or low amounts of latewood because of species-specific changes in ring width. Therefore, years that contain samples with higher portions of non-latewood (earlywood) material may influence the final chronology of isotopic variability. Here, we analyze the potential influence that changing earlywood and latewood components of individual tree rings can have on stable carbon and oxygen records from Quercus spp. and Pinus heldreichii chronologies. Analysis of stable isotopes in oak tree rings with varying amounts of latewood show no statistically significant differences in the range of isotopic composition, nor any major differences when considering the same calendric year. Similar results were found for the pine data, when comparing stable isotope measurements with earlywood-to-latewood ratio and maximum density. We argue that this simple approach should be applied to any long-term tree-ring stable isotope record in order to provide a better understanding of the potential biases that could arise from previously recorded intra-annual variability in the wood
Regulation of Epithelial-Mesenchymal transition by the ERK pathway.
Typical epithelium is uniformly polarized solid structure defined by the presence of cell-cell contacts that are connected to well-organized network of actin cytoskeleton. While epithelium is considered to be rather static, during embryogenesis or cancer development epithelial tissues undergo considerable dynamic changes in their integrity that are characterized by loss of epithelial polarity, disruption of cell-cell adhesions and gaining mesenchymal or mesenchymal-like migratory phenotype. These changes, collectively termed as epithelial-mesenchymal transition (EMT), allow cells to effectively invade surrounding tissues and are considered to be a main factor underlying the formation of metastatic cancer. The MAPK/ERK cascade, comprised of protein kinases Raf, MEK and ERK, induces the breakdown of epithelial integrity and cell autonomous migration in various cell lines. In the ERK pathway, ERK is an effector protein kinase which, depending on the cellular context, phosphorylates a number of different substrates. Spatiotemporal phosphorylation of specific constellation of ERK substrates drives specific biologic outcome. The question arises whether, during conversion of multicellular epithelium to autonomously migrating cells, ERK regulates a "master" controller or whether the ERK regulatory function..
