However, it is still unclear how the auditory cortical changes accompany the subcortical neural changes. The present study aimed to find the neural plastic changes in the cortical and subcortical auditory system following adult-onset single-sided deafness (SSD) using Mn-enhanced magnetic resonance imaging (MEMRI). Material and methods: B57BL/6 mice (postnatal 8 weeks old) were divided into three groups: the SSD-4-week group (postnatal 12 weeks old, n = 11), the SSD-8-week group (postnatal 16 weeks old, n = 11), and a normal hearing control group (postnatal 8 weeks old, n = 9). The left cochlea was ablated in the SSD groups. White Gaussian noise was delivered for 24 h before MEMRI acquisition. T1-weighted MRI data were analyzed from the cochlear nucleus (CN), superior olivary complex (SOC), lateral lemniscus (LL), inferior colliculus (IC), medial geniculate body (MG), and auditory cortex (AC). The differences in relative Mn2+-enhanced signal intensities (Mn2+SI) and laterality were analyzed between the groups. Results: Four weeks after the SSD procedure, the ipsilateral side of the SSD showed significantly lower Mn2+SI in the CN than the control group. On the other hand, the contralateral side of the SSD demonstrated significantly lower Mn2+SI in the SOC, LL, and IC. These decreased Mn2+SI values were partially recovered at 8 weeks after the SSD procedure. The interaural Mn2+SI differences representing the interaural dominance were highest in CN, then became less prominent higher in the auditory neural system. The SSD-8-week group still showed interaural differences in the CN, LL and IC. In contrast, the MG and AC did not show any significant intergroup or interaural differences in Mn2+SI. Conclusion: Subcortical auditory neural activities were decreased after SSD, and the interaural differences were diluted in the higher auditory nervous system. These findings were attenuated with time. Soc., 2013, 135, 18957).Calix[4]arene-triacid-monoquinone (CTAQ), a quinone-containing water-soluble ionophore, was utilized to investigate how the proton-coupled electron transfer (PCET) reaction of quinone in water would be altered by a redox-inactive metal ion. This ionophoric quinone derivative captured a Ca2+ ion, which drastically changed the voltammetric behavior of the quinone. Its characteristic response to pH and unique redox potential separation allowed us to decipher the underlying mechanism. Spectroelectrochemistry verified significant stabilization of the semiquinone, and electrocatalytic currents were observed in the presence of Ca2+-free CTAQ. Using digital simulation of cyclic voltammograms to clarify how the thermodynamic properties of quinones were altered, a simple scheme was proposed that successfully accounted for the observations. The change induced by Ca2+ complexation was explained on the basis of the combined effects of electrostatic influence of the captured metal ion and hydrogen bonding of water molecules with the support of DFT calculation. The uncertainties and risks from a member of the community can be further divided into those stemming from the hidden type problem and those stemming from the hidden action problem in the context of information asymmetry. These uncertainties and risks posed by community members can be alleviated by a prudently designed selection mechanism that uses repeated communication and learning. Nevertheless, there exists an incentive to commit a violation for a community member who is selected by the selection mechanism. A complementary mechanism such as reputation or third party intervention is therefore required to resolve this problem. On the other hand, the alleviation of the uncertainty or risk posed by an outsider of the community requires the effort of the entire community and individual investment by each community member to protect their information and systems. Enhancing trust is a critical factor in the development of virtual and offline societies. Just as various policy tools have been used continuously to build trust in the real society, various policy guidelines also need to be suggested to build trust in the virtual society. Although previous studies have focused on suggesting policy guidelines based on observed phenomena, this study provides the theoretical foundation for analyzing the process of trust building in various environments of virtual society using the game theory approach. The theoretical analysis in this research suggests that the most critical task is to make a pool of trustworthy providers to establish an efficient market. Prudent policies also need to be designed to differentiate the signaling costs for different types of providers. The trusted third party method can be one of the possible alternatives. As this study suggests, even in a trustworthy market, minimum monitoring and penalty contracts are necessary and individual users need to invest in optimal security. This research also contributes to the development of a new trust-management mechanism that is not only more objective and robust but also has a simple structure that can be easily understood by users in the virtual society. Existing studies have merely focused on one of the two conflicting values or indicated the limitations of pervasive reputation mechanisms. Moreover, flexible-monitoring levels cannot be chosen when the service participants are highly concerned about their privacy or when the expected loss from the invasion of privacy is high. In such cases, the level of punishment is inevitably highlegal enforcement is therefore required to complement the voluntary punishment scheme for virtual society models such as the utility-computing service market. Finally, this research contributes to the decision-making process of the defender. The proposed model gives a defender more practical instruments to decide the optimal level of security investment through consideration of the attackers strategic decisions. The majority of existing studies have considered only the defenders perspective and have regarded the actions of attackers as a given. The last analysis suggested a model of interdependent decision-making processes of two players behaving strategically. The strategic attacker bases its strategies such as attack frequency on the actions of the defender, whereas the strategic defender bases its strategies such as the level of security investment on the actions of the attacker. This study was designed to develop a novel anti-cancer immunogene therapy effective against malignant melanoma in the C57BL/6 mouse model. A recombinant plasmid containing MART1, a human melanoma-specific tumor antigen, was used to induce an immune reaction against the mouse melan-A epitope in order to overcome the peripheral tolerance of the mouse to murine melan-A. In addition, mouse granulocyte macrophage-colony stimulating factor (mGM-CSF) and short hairpin RNA against mouse transforming growth factor-ฮฒ2 (shmTGF-ฮฒ2) genes were delivered together with MART1, because GM-CSF is known to be the most potent inducer of antitumor immunity, and TGF-ฮฒ is known to be involved in tumor survival and host immune suppression. These genes were delivered to cancer cells by using an oncolytic adenovirus. A recombinant DNA expressing human MART1 (MART1 plasmid) and a recombinant adenovirus expressing human MART1 were investigated for their potential effects on priming and boosting immune responses.First, the expression of MART1 was increased in MART1 plasmid-transfected B16BL6 mouse melanoma cells in a dose-dependent manner in vitro. Notably, the cytotoxic activity of splenocytes isolated from MART1 plasmid-injected non-tumor-bearing mice was enhanced compared to those isolated from control plasmid-injected mice.Thus, recombinant oncolytic adenovirus expressing mGM-CSF and shmTGF-ฮฒ2 were also investigated for their potential to stimulate the non-specific immune response and decrease the expression of signaling molecules involved in tumor cell survival and growth, respectively. To this end, the effect of recombinant oncolytic adenovirus expressing both mGM-CSF and shmTGF-ฮฒ2 (GT virus) was compared to that of the recombinant adenovirus expressing mGM-CSF only (G virus), following intratumoral injection of the virus into melanoma-bearing C57BL/6 mice. This investigation shows that administration of the G virus leads to delayed tumor growth compared to the empty viral control, while tumor growth in mice that received the GT virus was significantly decreased (P < 0.001) compared to both the control- and G virus-treated mice. Finally, an oncolytic adenovirus expressing MART1, mGM-CSF, and shmTGF-ฮฒ2 (MGT virus) was constructed and administered to boost the immune response and cancer cell death. Administration of this virus induced a stronger and longer-lived immune response than that observed in the controls. Interestingly, none of the mice that received MART1 plasmid pre-treatment in addition to MGT viral injection showed any signs of tumor growth and 100% were viable 43 days after tumor cell injection. This study investigates the anti-tumor effects of repeated MART1 plasmid vaccination and immune stimulation/tumor cell lysis with an oncolytic adenovirus expressing MART1/mGM-CSF/shmTGF-ฮฒ2.     We previously observed that TRAIL induces acquired TRAIL resistance coinciding with increased Akt phosphorylation brought about by the Src-PI3K-Akt signaling pathways and mediated by c-Cbl. c-Cbl, a ubiquitously expressed cytoplasmic adaptor protein, is simultaneously involved in the rapid degradation of TRAIL receptors and Akt phosphorylation during TRAIL treatment. Here, we show that Akt phosphorylation is not exclusively responsible for acquired TRAIL resistance. Akt catalytic activation is known to increase during metabolic oxidative stress, but we show that TRAIL also dramatically induces the catalytic activation of Akt in TRAIL-sensitive cells, but not in TRAIL-resistant cells. This suggests that Akt catalytic activation during TRAIL-induced apoptosis is likely to play a compensatory role in the maintenance of cell homeostasis.     Serum inflammatory markers are used in the prognostication of colorectal cancer (CRC); however, the corresponding role of positron emission tomography (PET)-derived inflammatory markers remains unclear. This study aimed to investigate the prognostic value of 18F-fluorodeoxyglucose (FDG) uptake in the bone marrow and spleen of patients with CRC and evaluate the relationship between FDG uptake estimates in these organs and serum inflammatory markers. In total, 411 patients who underwent preoperative FDG PET/computed tomography (CT) within 1 month of surgery were enrolled. The mean standardized uptake values of the bone marrow and spleen were normalized to the value of the liver, thereby generating bone marrow-to-liver uptake ratio (BLR) and spleen-to-liver uptake ratio (SLR) estimates. The value of BLR and SLR in predicting overall survival (OS) was assessed using the Cox proportional hazards model. The correlation between BLR or SLR and neutrophil-to-lymphocyte ratio (NLR) was evaluated. The predictive accuracy of BLR alone and in combination with SLR was compared using the integrated area under the receiver operating characteristic curves (iAUC). In the univariate analysis, BLR (> 1.06) and SLR (> 0.93) were significant predictors of OS. In the multivariate analysis, BLR was an independent predictor of OS (hazard ratio = 5.279; p < 0.001). Both BLR and SLR were correlated with NLR (p < 0.001). A combination of BLR and SLR was better than BLR alone at CRC prognostication (iAUC, 0.561 vs. 0.542). FDG uptake estimates in the bone marrow and spleen may be useful imaging-derived biomarkers of systemic inflammation, supporting CRC prognostication.ope
