    Tangled Roots: Exploring Appalachian Feminisms

    Women have long been overlooked as key figures in the cultural history of Appalachia. The exhibition Tangled Roots: Exploring Appalachian Feminisms seeks to examine the ways in which women artists across the region have kept traditions alive while redefining creative practices that were once seen strictly as “women’s work.” In particular, the exhibition aims to explore how women have reimagined “craft” through skillful attention to materials, manual dexterity, and application of critical and conceptual rigor. The concept of craft is defined in this context to include all hand-made work that requires developed skills, whether they belong to traditional craft-based practices or ones associated with the so-called fine arts.     HPV16 E5 expression induces switching from FGFR2b to FGFR2c and epithelial-mesenchymal transition

    The E5 oncoprotein of the human papillomavirus type 16 (HPV16 E5) deregulates epithelial homeostasis through the modulation of receptor tyrosine kinases and their signaling. Accordingly, the fibroblast growth factor receptor 2b (FGFR2b/KGFR), epithelial splicing transcript variant of the FGFR2, is down-modulated by the viral protein expression, leading to impairment of keratinocyte differentiation. Here, we report that, in cell models of transfected human keratinocytes as well as in cervical epithelial cells containing episomal HPV16, the down-regulation of FGFR2b induced by 16E5 is associated with the aberrant expression of the mesenchymal FGFR2c isoform as a consequence of splicing switch: in fact, quantitative RT-PCR analysis showed that this molecular event is transcriptionally regulated by the epithelial splicing regulatory proteins 1 and 2 (ESRP1 and ESRP2) and is able to produce effects synergistic with those caused by TGFβ treatment. Immunofluorescence analysis revealed that this altered FGFR2 splicing leads to changes in the specificity for the ligands FGFs and in the cellular response, triggering epithelial-mesenchymal transition (EMT). Through 16E5 or FGFR2 silencing as well as inhibition of FGFR2 activity we demonstrated the direct role of the viral protein in the receptor isoform switching and EMT, suggesting that these early molecular events during HPV infection might represent additional mechanisms driving cervical transformation and tumor progression

    This study aims to determine how much influence the product, price, location, and promotion to the buying decision that can build customer satisfaction in stores Essy's Brownies Semarang. This study uses four independent variables are product, price, location, and promotion, customer satisfaction as the dependent variable and the purchase decision as an intervening variable. The population used in this study is that consumers who had visited the shop Essy's Brownies Semarang. This research method using a non-probability sampling technique with accidental sampling method. Used as a sample of 160 respondents. Data obtained from the questionnaires were then processed and analyzed using multiple regression analysis and analysis of SEM (Structural Equation Modeling) through the AMOS program. The results showed that the product has a positive influence on purchasing decisions; price has a positive influence on purchasing decisions; location has a positive influence on purchasing decision; promotion has a positive influence on purchasing decisions; and purchasing decisions have a positive effect on customer satisfaction

    Zika virus is currently attracting a huge international attention. Recently concluded Rio Olympics are a testament to the importance of public health planning and preparedness to counter the threat of emerging infections. Zika is a documented condition from 1940’s and is not so extensively studied until recently. The similarity of the virus transmission and the presence of a common vector pose a significant threat to the community. The strong link of the vector to the environmental conditions is another factor that has to thoroughly explored to prevent the rapid spread that was evident in other regions of the world. Three essential steps are proposed in the control and require further studies. Firstly, raising the awareness efficiency towards the disease. Second, the vector control and the impact of climate change on the spread of the vector needs to be understood. And thirdly, true burden of the disease and the cost of prevention to the health system need to be explored in depth

    Narapidana ialah terpidana yang telah inkracht dan menjalani hukuman pidana penjara di Lembaga Pemasyararakatan (Lapas). Kebebasan bergerak dari narapidana akan hilang karena dibatasi oleh tembok penjara. Dengan proses pembinaan yang berkelanjutan akan membuat narapidana menjadi manusia yang lebih baik, sadar akan kesalahan dan tidak mengulangi kesalahan yang pernah dibuat. Penelitian ini menggunakan desain studi kuantitatif, yang mana karakteristik dari penelitian ini dilakukan pada lingkungan yang terkontrol dengan maksud tujuan untuk dapat mencapai hasil yang objektif dan tidak dipengaruhi oleh lingkungan eksternal, lebih menitikberatkan prediksi instrument deskripsi, fokus pada perilaku, biasanya melibatkan penggunaan kuesioner terstruktur atau metode eksperimental serta dilakukan pada sejumlah responden yang besar. Penelitian ini menggunakan uji statistik untuk mengetahui tingkat pengaruh tersebut. Adapun uji statistik yangg digunakan yaitu uji normalitas untuk mengetahui apakah data terdistribusi secara normal atau tidak, uji regresi linier sederhana dan uji determinasi untuk mengetahui tingkat pengaruh variabel bebas terhadap variabel terikat, serta uji signifikansi atau uji t untuk melakukan pengujian terhadap hipotesis penelitian. Tujuan utama penelitian ini adalah mengetahui pengaruh dari Kualitas Pelayanan Terhadap Kepuasan Narapidana di Lapas Kelas IIB Slawi. Berdasarkan hasil analisis data yang telah dilakukan oleh peneliti menggunakan SPSS versi 26 maka dapat diambil kesimpulan bahwa kualitas pelayanan memiliki pengaruh positif terhadap kepuasan narapidana di Lapas Kelas IIB Slawi. Besarnya persentase pengaruh kualitas pelayanan terhadap kepuasan narapidana yaitu sebesar sebesar 56,3% sedangkan sisanya sebesar 43,7% dipengaruhi oleh variabel lain yang tidak diteliti dalam penelitian ini

    Algoritmi per la reiezione dei disturbi nei sistemi di acquisizione dei segnali EEG basati sulla tecnica del Compressed Sensing

    Il Compressed Sensing è emerso recentemente come un strumento che simultaneamente acquisisce e comprime segnali analogici su dispositivi a basso consumo. Rispetto alla sua caratterizzazione classica (CS), il sistema di acquisizione può essere adattato alla classe di segnali in ingresso (R-CS) ed è inoltre possibile garantire una buona reiezione dei disturbi (R-CSd). Quest’ultimo modello si basa sulla risoluzione di due problemi di ottimizzazione con un numero di variabili potenzialmente elevato. Il primo dei quali ha una risoluzione analitica, il secondo necessita di un risolutore software che, per un numero elevato di variabili, potrebbe non arrivare ad una soluzione in tempi ragionevoli. Il lavoro che viene presentato supera questo problema con un algoritmo scritto ad hoc che sfrutta l’applicazione del metodo del Gradiente Proiettato Discendente con Proiezioni Alternate, attraverso il quale si riesce a ridurre drasticamente il tempo richiesto dalla CPU per ottenere una soluzione, anche per un numero elevato di variabili. A conclusione del lavoro si è applicato questo metodo alla classe di segnali EEG con l’intento di attuare una reiezione dei disturbi a bassissima frequenza direttamente nello stadio di compressione. Il lavoro mostra la catena di elaborazione per il CS, il R-CS e per R-CSd. I casi analizzati sono: adattamento sulla classe di segnali EEG, adattamento sul singolo canale e divisione dei canali in due distinti cluster. Quello che si dimostra è che l’algoritmo R-CSd mostra le stesse performance di R-CS in tutti e tre i casi, facendo a meno dell’utilizzo di un filtro passa alto. La ricostruzione dei canali con la caratterizzazione dell’intera classe di segnali o con l’uso dei due cluster non si discosta troppo da quanto osservato per l’adattamento sul singolo canale con una conseguente semplificazione del sistema di acquisizione proposto

    Bonek is the name of a Java language acronym that means “Bondo Nekat”, Bonek itself has a long history for the development of our supporters in Indonesia, Surabaya supporter of community pride is instrumental in supporting Persebaya wherever located. Along with moderniasasi supporters, Bonek was walking toward the positive stigma but people still think Bonek is a collection of anarchist supporters. The general objective of the visual design of the book titled "Nyali Wani, History of Bonek Surabaya Subculture" This is an asset that mix past historical events of the early emergence of Bonek, so as to be evidence of the amount of history held Bonek for movement supporters in Indonesia. The method used is an ethnographic research designs to study the behavior of the target audience and the picture of the surrounding environment as a visual reference, depth interviews and observations to obtain data on book content, as well as the literature review as a good theoretical basis in the visualization and preparation of a book's content. The results of the research analysis is then used as a reference in making design concept in this design. Book distinctive visual history bold concept Arek-Arek Suroboyo visualized with collage technique which appeared in the style of design in the late modern era in accordance with the nature of its appearance. Made visual book about the history of this Persebaya supporters, is expected to clarify who actually Bonek to the public, as well as historical archives in the future. ================================================================================================ Bonek adalah nama dari sebuah akronim bahasa jawa yang berarti Bondo Nekat, Bonek sendiri memiliki sejarah yang panjang bagi perkembangan supporter di Indonesia, supporter kebanggaan masyarakat Surabaya ini berjasa dalam mendukung Persebaya dimanapun berada. Seiring dengan moderniasasi suporter, Bonek sudah berjalan menuju stigma positifnya namun masyarakat masih menganggap Bonek adalah kumpulan suporter anarkis. Tujuan umum perancangan buku visual berjudul “Nyali Wani, Sejarah Bonek Subktultu Asli Surabaya” ini adalah menjadi aset sejarah yang meruntutkan peristiwa-peristiwa sejarah di masa awal kemunculan Bonek, sehingga mampu menjadi bukti besarnya sejarah yang dimiliki Bonek untuk pergerakan suporter di Indonesia. Metode riset yang digunakan adalah etnografi desain untuk mengetahui perilaku target audiens dan gambaran lingkungan sekitarnya sebagai referensi visual, depth interview dan observasi untuk memperoleh data konten buku, serta literature review sebagai landasan teori baik dalam visualisasi maupun penyusunan konten buku. Hasil analisa riset tersebut kemudian dijadikan sebagai acuan dalam pembuatan konsep desain pada perancangan ini. Buku visual sejarah berkonsep berani khas arek-arek Suroboyo ini divisualisasikan dengan teknik kolase yang muncul pada gaya desain di era modern akhir sesuai dengan sifat kemunculannya. Dibuatnya buku visual sejarah tentang suporter Persebaya Surabaya ini, diharapkan mampu menjelaskan siapa Bonek sebenarnya kepada masyarakat, serta sebagai arsip sejarah di masa mendatang

    Rosiglitazone inhibits metastasis development of a murine mammary tumor cell line LMM3

    <p>Abstract</p> <p>Background</p> <p>Activation of peroxisome proliferator-activated receptors γ (PPARγ) induces diverse effects on cancer cells. The thiazolidinediones (TZDs), such as troglitazone and ciglitazone, are PPARγ agonists exhibiting antitumor activities; however, the underlying mechanism remains inconclusive. Rosiglitazone (RGZ), a synthetic ligand of PPARγ used in the treatment of Type 2 diabetes, inhibits growth of some tumor cells and is involved in other processes related to cancer progression. Opposing results have also been reported with different ligands on tumor cells. The purpose of this study was to determine if RGZ and 15d-PGJ<sub>2 </sub>induce antitumor effects <it>in vivo </it>and <it>in vitro </it>on the murine mammary tumor cell line LMM3.</p> <p>Methods</p> <p>The effect on LMM3 cell viability and nitric oxide (NO) production of different doses of RGZ, 15-dPGJ<sub>2</sub>, BADGE and GW9662 were determined using the MTS colorimetric assay and the Griess reaction respectively. <it>In vivo </it>effect of orally administration of RGZ on tumor progression was evaluated either on s.c. primary tumors as well as on experimental metastasis. Cell adhesion, migration (wound assay) and invasion in Transwells were performed. Metalloproteinase activity (MMP) was determined by zymography in conditioned media from RGZ treated tumor cells. PPARγ expression was detected by inmunohistochemistry in formalin fixed tumors and by western blot in tumor cell lysates.</p> <p>Results</p> <p>RGZ orally administered to tumor-bearing mice decreased the number of experimental lung metastases without affecting primary s.c. tumor growth. Tumor cell adhesion and migration, as well as metalloproteinase MMP-9 activity, decreased in the presence of 1 μM RGZ (non-cytotoxic dose). RGZ induced PPARγ protein expression in LMM3 tumors. Although metabolic activity -measured by MTS assay- diminished with 1–100 μM RGZ, 1 μM-treated cells recovered their proliferating capacity while 100 μM treated cells died. The PPARγ antagonist Biphenol A diglicydyl ether (BADGE) did not affect RGZ activity. On the contrary, the specific antagonist GW9662 completely abrogated RGZ-induced decrease in cell viability. A decrease in NO levels was detected in the presence of either 1 or 100 μM RGZ. The natural ligand 15d-PGJ<sub>2 </sub>did not affect metabolic activity although it induced a significant decrease in NO production.</p> <p>Conclusion</p> <p>A significant decrease in the number of experimental LMM3 lung metastasis, but not on primary tumor growth, after oral RGZ administration was observed. <it>In vitro</it>, 100 μMRGZ also reduced cell viability and NO production, while no changes were observed in the presence of 15d-PGJ<sub>2</sub>. BADGE did not reverse RGZ effect while the antagonist GW9662 completely abrogated it, suggesting a PPARγ- dependent mechanism. Inhibition of lung metastatic nodules by RGZ administered in vivo, might be associated with the observed decrease in MMP-9 expression, in cell adhesion, migration and invasion. RGZ augmented its expression. PPARγ was detected in cell lysates by western blot and by immunohistochemistry in tumors from RGZ-treated mice. In summary we can suggest that RGZ or any other TZDs might be possible future approaches in the treatment of metastasis of PPARγ-expressing cells.</p

    Vinyl Cations As Cyclopentenone Precursors Via C‒h Insertion Or Alkene Addition Reactions

    We report that Lewis acid mediated reactions of β-hydroxy-α-diazo carbonyls provide facile access to vinyl cation intermediates, which can form mono- and bicyclic cyclopentenone ring systems. Vinyl cations are sp hybridized cations that can be exploited as carbene surrogates or as electrophiles. The methodology reported in this dissertation is important because cyclopentenone rings are common motifs in natural products or can be used as synthetic intermediates. The first portion of this dissertation will discuss vinyl cations we’ve used as carbene surrogates in metal free C‒H insertion reactions. Treating β-hydroxy-α-diazo carbonyls with a Lewis acid provided vinyl cation intermediates after loss of hydroxide and nitrogen gas. A 1,2-methylene shift then provided a vinyl cation which can insert into an inert C‒H bond to give mono- and bicyclic cyclopentenone rings. Modification at the point of insertion to a more substituted center provides a competitive elimination type reaction. This result sparked a deeper investigation into the concertedness of the C‒H insertion. Further expansion of this work included migratory aptitude studies in the 1,2-shift step for non-symmetric aliphatic and tetralone systems. The second portion of this dissertation forms vinyl cations in a similar fashion, but uses them as electrophiles that are susceptible to a nucleophilic attack by a pendant alkene. This reaction, which we hypothesize proceeds through a unique acylium intermediate, forms α-alkylidene cyclopentenones in good yields. The efficiency of this reaction was influenced by the group adjacent to the vinyl cation. Aryl groups which were electron rich or were sterically hindered formed α-alkylidene cyclopentenones in higher yields than electron poor aryl rings or alkyl groups.
