    Key requirements for the validity of a neuropathological study are the inclusion of large numbers of biopsy or autopsy cases and proper controls, the rigorous classification of the basic neuropathology and the selection of the most suitable technologies for investigation. Whether the studies are performed with the fanciest, new, and state of the art technology or with rather conventional methodology is of minor importance. Fol-lowing these criteria, a spectrum of neuropathological studies has been published in 2020, which provides new insights on important questions related to neurological disease. They include the pathological substrate of brain disease in COVID-19 infected patients, the nature of the adaptive and innate inflammatory response, or the type and mechanisms of tissue injury and repair in multiple sclerosis, and diagnostically relevant or mechanistic new insights into antibody-mediated diseases of the central nervous system. Other studies de-scribe in detail the dynamic changes of brain inflammation in patients with trisomy 21 as a disease model for Alzheimer’s disease, or the presence and consequences of vascular comorbidities in a chronic inflammatory disease, such as multiple sclerosis. All these contributions have provided important, highly relevant clues for basic and translational neuroscience

    Ten neuropathological studies, published in 2019, are discussed, which address important aspects of neuroimmunology and inflammatory brain disease. They include topics related to new mechanisms of inflammation and immune mediated neurodegeneration, which are relevant for multiple sclerosis (publications 1 to 4) and discuss the role of specific autoantibodies against myelin oligodendrocyte glycoprotein or aquaporin 4 in neuromyelitis optica spectrum disorders (publications 5 and 6). Other studies highlight the discovery of new virus induced diseases of the nervous system and their relevance for clinical neurology and diagnostic neuropathology (publications 7 and 8). Finally, very interesting studies are discussed dealing with microglia and immune mechanisms in neurodegeneration (publication 9) and the neuropathological long-term outcome of Aß vaccination in Alzheimer’s disease (publication 10). All these studies highlight the central role of neuropathology in neurological disease research

    The role of regional surveillance networks in enhancing global outbreak reporting

    BACKGROUND: The Program for Monitoring Emerging Diseases (ProMED) is a moderated electronic reporting system dedicated to the rapid, global dissemination of outbreak reports. Its moderators are globally diverse, carefully selected, highly trained specialists. To improve cross-border communication and rapidly identify regional health threats, ProMED created regional networks where locally-based moderators use their access to local and regional medical and public health networks and media sources to obtain information not readily available outside of their region. In this analysis, we assess the impact of the establishment of ProMED's Middle East/North Africa (MENA) and South Asia (SoAs) regional networks in April 2014 on ProMED's outbreak reports for these regions. METHODS & MATERIALS: Outbreak reports in countries within the two regions were extracted from ProMED's database, and included country, disease name, species type, spatial coordinates, and report issue date. Data analysis included visualizing spatial information, identifying unique reports, and reporting trends per country and region. Data processing and analysis were conducted using R 3.4.0 statistical software. Rates of outbreak events per total number of ProMED reports per year were calculated to adjust for temporal trends in the total number of reports posted on ProMED. Rate comparison used a two-sided t-test; P < 0.05 was considered statistically significant. RESULTS: The mean monthly incidence of ProMED reports concerning outbreaks in the MENA region increased from 28 reports (May 2012 - April 2014) to 83 reports after the establishment of the networks (May 2014 - April 2016), and from 29 reports to 101 reports concerning outbreaks in the SoAs region over the same time period. The number of reports per total number of ProMED reports increased by 259% for MENA, and 289% for SoAs (P < 0.01). MENA reports most often addressed MERS (32.3%), foot-and-mouth disease (7.0%), avian influenza (6.7%), and measles (3.8%); whereas SoAs most often addressed dengue (14.9%), anthrax (7.3%), Japanese encephalitis (7.0%), CCHF (4.9%), and rabies (4.8%). CONCLUSION: The establishment of MENA and SoAs regional networks with locally-based, expert moderators resulted in a significant increase in ProMED's outbreak reports from these regions and an increased flow of disease information across regional borders and to the global public health community

    Decomposing the margins of transfer pricing

    This paper examines the determinants and margins of profit shifting through transferpricing. We develop a theory model, where transfer pricing patterns are governed by a generalized concealment cost function (CCF). Our empirical analysis draws on micro-level data about transaction-level imports, firm-level characteristics, as well as tax differentials between regions in Switzerland and countries abroad. We find, both theoretically and empirically, that more productive multinational firms deviate less from the arms’ length price and trade lower quantities, compared to MNEs with lower productivity. Moreover, the decision of firms to engage in transfer pricing depends negatively on a fixed cost component in the CCF, as well as trade costs. The model allows us to estimate a theory-consistent concealment cost function, which can be used for counterfactual analysis

    Kalign – an accurate and fast multiple sequence alignment algorithm

    BACKGROUND: The alignment of multiple protein sequences is a fundamental step in the analysis of biological data. It has traditionally been applied to analyzing protein families for conserved motifs, phylogeny, structural properties, and to improve sensitivity in homology searching. The availability of complete genome sequences has increased the demands on multiple sequence alignment (MSA) programs. Current MSA methods suffer from being either too inaccurate or too computationally expensive to be applied effectively in large-scale comparative genomics. RESULTS: We developed Kalign, a method employing the Wu-Manber string-matching algorithm, to improve both the accuracy and speed of multiple sequence alignment. We compared the speed and accuracy of Kalign to other popular methods using Balibase, Prefab, and a new large test set. Kalign was as accurate as the best other methods on small alignments, but significantly more accurate when aligning large and distantly related sets of sequences. In our comparisons, Kalign was about 10 times faster than ClustalW and, depending on the alignment size, up to 50 times faster than popular iterative methods. CONCLUSION: Kalign is a fast and robust alignment method. It is especially well suited for the increasingly important task of aligning large numbers of sequences

    Partitioning the triangles of the cross polytope into surfaces

    We present a constructive proof that there exists a decomposition of the 2-skeleton of the k-dimensional cross polytope βk\beta^k into closed surfaces of genus g≤1g \leq 1, each with a transitive automorphism group given by the vertex transitive Z2k\mathbb{Z}_{2k}-action on βk\beta^k. Furthermore we show that for each k≡1,5(6)k \equiv 1,5(6) the 2-skeleton of the (k-1)-simplex is a union of highly symmetric tori and M\"obius strips.Comment: 13 pages, 1 figure. Minor update.
