    Experimental Evidence for the Population-Dynamic Mechanisms Underlying Extinction Cascades of Carnivores

    SummarySpecies extinction rates due to human activities are high [1–3], and initial extinctions can trigger cascades of secondary extinctions, leading to further erosion of biodiversity [4]. A potential major mechanism for secondary extinction cascades is provided by the long-standing theory that the diversity of consumer species is maintained due to the positive indirect effects that these species have on each other by reducing competition among their respective resource species [5–7]. This means that the loss of one carnivore species could lead to competitive exclusion at the prey trophic level, leading to extinctions of further carnivore species. Evidence for these effects is difficult to obtain due to many confounding factors in natural systems, but extinction cascades that could be due to this mechanism have been demonstrated in simplified laboratory microcosms [8]. We established complex insect food webs in replicated field mesocosms and found that the overharvesting of one parasitoid wasp species caused increased extinction rates of other parasitoid species, compared to controls, but only when we manipulated the spatial distribution of herbivore species such that the potential for interspecific competition at this level was high. This provides clear evidence for horizontal extinction cascades at high trophic levels due to the proposed mechanism. Our results demonstrate that the loss of carnivores can have widespread effects on other species at the same trophic level due to indirect population-dynamic effects that are rarely considered in this context

De nadruk op resultaataansturing heeft veel te maken met de opkomst van New Public Management (NPM); een kernidee van NPM is de gedachte dat organisaties met publieke taken efficiĂ«nter en effectiever gaan opereren wanneer prestaties transparant worden gemaakt en managers in publieke organisaties worden aangesproken op deze prestaties. Hoewel het gedachtegoed van NPM wat terrein lijkt te verliezen, is bij de gedoodverfde ‘opvolger’ van NPM, de Public Value Management (PVM)-stroming, eveneens veel aandacht voor het inzichtelijk maken van de na te streven doelen door prestatiemeting

    Defragmenting the Module Layout of a Partially Reconfigurable Device

    Modern generations of field-programmable gate arrays (FPGAs) allow for partial reconfiguration. In an online context, where the sequence of modules to be loaded on the FPGA is unknown beforehand, repeated insertion and deletion of modules leads to progressive fragmentation of the available space, making defragmentation an important issue. We address this problem by propose an online and an offline component for the defragmentation of the available space. We consider defragmenting the module layout on a reconfigurable device. This corresponds to solving a two-dimensional strip packing problem. Problems of this type are NP-hard in the strong sense, and previous algorithmic results are rather limited. Based on a graph-theoretic characterization of feasible packings, we develop a method that can solve two-dimensional defragmentation instances of practical size to optimality.     De Controller als Academische Professional: Belangrijke Ontwikkelingen voor Controllers en Implicaties voor het Onderwijs

    Uit de beschrijving van de ontwikkelingen die op dit moment van belang zin voor het controllersvakgebied valt op te maken dat voor het opleiden van controllers tot academische professionals niet alleen de nieuwste academische inzichten nodig zin, maar ook een programma dat een kritische, een proactieve, en zelfs een moedige houding stimuleert. Met andere woorden: “Controllership van de toekomst is [...] iets voor kritische geesten die een ondersteunende rol willen spelen die tegelijkertid een tegenrol is”. Het vormgeven van een onderwijsprogramma dat een dergelijke academische houding ontwikkelt en waarin daarnaast zowel specifieke ontwikkelingen als vakgebied overstijgende dwarsverbanden een plek krijgen, is een uitdagende taak. We hopen dat deze bijdrage helpt bij het aangaan van die uitdaging en bij kan dragen aan het verder ontwikkelen van opleidingen voor academische controllers in Nederland. En meer nog dan dat hopen we dat de bijdrage voer mag zin voor kritische geesten

    A high-throughput microfluidic assay to study neurite response to growth factor gradients

    Studying neurite guidance by diffusible or substrate bound gradients is challenging with current techniques. In this study, we present the design, fabrication and utility of a microfluidic device to study neurite guidance under chemogradients. Experimental and computational studies demonstrated the establishment of a steep gradient of guidance cue within 30 min and stable for up to 48 h. The gradient was found to be insensitive to external perturbations such as media change and movement of device. The effects of netrin-1 (0.1–10 ”g mL−1) and brain pulp (0.1 ”L mL−1) were evaluated for their chemoattractive potential on neurite turning, while slit-2 (62.5 or 250 ng mL−1) was studied for its chemorepellant properties. Hippocampal or dorsal root ganglion (DRG) neurons were seeded into a micro-channel and packed onto the surface of a 3D collagen gel. Neurites grew into the matrix in three dimensions, and a gradient of guidance cue was created orthogonal to the direction of neurite growth to impact guidance. The average turning angle of each neurite was measured and averaged across multiple devices cultured under similar conditions to quantify the effect of guidance cue gradient. Significant positive turning towards gradient was measured in the presence of brain pulp and netrin-1 (1 ”g mL−1), relative to control cultures which received no external guidance cue (p < 0.001). Netrin-1 released from transfected fibroblasts had the most positive turning effect of all the chemoattractive cues tested (p < 0.001). Slit-2 exhibited strong chemorepellant characteristics on both hippocampal and DRG neurite guidance at 250 ng mL−1 concentration. Slit-2 also showed similar behavior on DRG neuron invasion into 3D collagen gel (p < 0.01 relative to control cultures). Taken together, the results suggest the utility of this microfluidic device to generate stable chemogradients for studying neurobiology, cell migration and proliferation, matrix remodeling and co-cultures with other cell lines, with potential applications in cancer biology, tissue engineering and regenerative medicine.Seoul R&BD Support Center (program PA090930

    Real-time detection of viable microorganisms by intracellular phototautomerism

    BACKGROUND: To date, the detection of live microorganisms present in the environment or involved in infections is carried out by enumeration of colony forming units on agar plates, which is time consuming, laborious and limited to readily cultivable microorganisms. Although cultivation-independent methods are available, they involve multiple incubation steps and do mostly not discriminate between dead or live microorganisms. We present a novel generic method that is able to specifically monitor living microorganisms in a real-time manner. RESULTS: The developed method includes exposure of cells to a weak acid probe at low pH. The neutral probe rapidly permeates the membrane and enters the cytosol. In dead cells no signal is obtained, as the cytosolic pH reflects that of the acidic extracellular environment. In live cells with a neutral internal pH, the probe dissociates into a fluorescent phototautomeric anion. After reaching peak fluorescence, the population of live cells decays. This decay can be followed real-time as cell death coincides with intracellular acidification and return of the probe to its uncharged non-fluorescent state. The rise and decay of the fluorescence signal depends on the probe structure and appears discriminative for bacteria, fungi, and spores. We identified 13 unique probes, which can be applied in the real-time viability method described here. Under the experimental conditions used in a microplate reader, the reported method shows a detection limit of 10(6) bacteria ml(-1), while the frequently used LIVE/DEAD BacLight Syto9 and propidium iodide stains show detection down to 10(6) and 10(7) bacteria ml(-1), respectively. CONCLUSIONS: We present a novel fluorescence-based method for viability assessment, which is applicable to all bacteria and eukaryotic cell types tested so far. The RTV method will have a significant impact in many areas of applied microbiology including research on biocidal activity, improvement of preservation strategies and membrane permeation and stability. The assay allows for high-throughput applications and has great potential for rapid monitoring of microbial content in air, liquids or on surfaces

    Artificial nighttime light changes aphid-parasitoid population dynamics

Available from Nature Publishing Group via the DOI in this record.Artificial light at night (ALAN) is recognized as a widespread and increasingly important anthropogenic environmental pressure on wild species and their interactions. Understanding of how these impacts translate into changes in population dynamics of communities with multiple trophic levels is, however, severely lacking. In an outdoor mesocosm experiment we tested the effect of ALAN on the population dynamics of a plant-aphid-parasitoid community with one plant species, three aphid species and their specialist parasitoids. The light treatment reduced the abundance of two aphid species by 20% over five generations, most likely as a consequence of bottom-up effects, with reductions in bean plant biomass being observed. For the aphid Megoura viciae this effect was reversed under autumn conditions with the light treatment promoting continuous reproduction through asexuals. All three parasitoid species were negatively affected by the light treatment, through reduced host numbers and we discuss induced possible behavioural changes.     Defensive insect symbiont leads to cascading extinctions and community collapse

    Animals often engage in mutualistic associations with microorganisms that protect them from predation, parasitism or pathogen infection. Studies of these interactions in insects have mostly focussed on the direct effects of symbiont infection on natural enemies without studying community-wide effects. Here, we explore the effect of a defensive symbiont on population dynamics and species extinctions in an experimental community composed of three aphid species and their associated specialist parasitoids. We found that introducing a bacterial symbiont with a protective (but not a non-protective) phenotype into one aphid species led to it being able to escape from its natural enemy and increase in density. This changed the relative density of the three aphid species which resulted in the extinction of the two other parasitoid species. Our results show that defensive symbionts can cause extinction cascades in experimental communities and so may play a significant role in the stability of consumer-herbivore communities in the field.
