    With increased awareness of the diversity of transmissible spongiform encephalopathy (TSE) strains in the ruminant population, comes an appreciation of the need for improved methods of differential diagnosis. Exposure to bovine spongiform encephalopathy (BSE) has been associated with the human TSE, variant Creutzfeldt–Jakob disease, emphasizing the necessity in distinguishing low-risk TSE types from BSE. TSE type discrimination in ruminants such as cattle, sheep, goats and deer, requires the application of several prion protein (PrP)-specific antibodies in parallel immunochemical tests on brain homogenates or tissue sections from infected animals. This study uses in a single incubation step, three PrP-specific antibodies and fluorescent Alexa dye-labelled anti-mouse Fabs on a Western blot. The usual amount of brain tissue needed is 0.5 mg. This multiplex application of antibodies directed towards three different PrP epitopes enabled differential diagnosis of all established main features of classical scrapie, BSE and Nor98-like scrapie in sheep and goats, as well as the currently known BSE types C, H and L in cattle. Moreover, due to an antibody-dependent dual PrP-banding pattern, for the first time CH1641 scrapie of sheep can be reliably discriminated from the other TSE isolate types in sheep

    PrPCWD lymphoid cell targets in early and advanced chronic wasting disease of mule deer

    Up to 15% of free-ranging mule deer in northeastern Colorado and southeastern Wyoming, USA, are afflicted with a prion disease, or transmissible spongiform encephalopathy (TSE), known as chronic wasting disease (CWD). CWD is similar to a subset of TSEs including scrapie and variant Creutzfeldt¿Jakob disease in which the abnormal prion protein isoform, PrPCWD, accumulates in lymphoid tissue. Experimental scrapie studies have indicated that this early lymphoid phase is an important constituent of prion replication interposed between mucosal entry and central nervous system accumulation. To identify the lymphoid target cells associated with PrPCWD, we used triple-label immunofluorescence and high-resolution confocal microscopy on tonsils from naturally infected deer in advanced disease. We detected PrPCWD primarily extracellularly in association with follicular dendritic and B cell membranes as determined by frequent co-localization with antibodies against membrane bound immunoglobulin and CD21. There was minimal co-localization with cytoplasmic labels for follicular dendritic cells (FDC). This finding could indicate FDC capture of PrPCWD, potentially in association with immunoglobulin or complement, or PrPC conversion on FDC. In addition, scattered tingible body macrophages in the germinal centre contained coarse intracytoplasmic aggregates of PrPCWD, reflecting either phagocytosis of PrPCWD on FDC processes, apoptotic FDC or B cells, or actual PrPCWD replication within tingible body macrophages. To compare lymphoid cell targets in early and advanced disease, we also examined: (i) PrPCWD distribution in lymphoid cells of fawns within 3 months of oral CWD exposure and (ii) tonsil biopsies from preclinical deer with naturally acquired CWD. These studies revealed that the early lymphoid cellular distribution of PrPCWD was similar to that in advanced disease, i.e. in a pattern suggesting FDC association. We conclude that in deer, PrPCWD accumulates primarily extracellularly and associated with FDCs and possibly B cells ¿ a finding which raises questions as to the cells responsible for pathological prion productio

    Arctic fox (Alopex lagopus) from the North Sea

    The first record of arctic fox Alopex lagopus (Linnaeus, 1758) from the Eurogeul area (North Sea) is reported based on a distal humerus fragment collected by private collectors from dredged sediments on ‘De Zandmotor’. It was radiocarbon dated to 29,900 + 550/- 490 BP (GrA-69520), which is younger than most Late Pleistocene fossils from this area. Besides adding a new species to the faunal list of the Eurogeul area, this specimen also demonstrates the value of beaches nourished with dredged sediments and highlights the importance of close cooperation between all parties interested in palaeontology: from collector to researcher. The discovery prompted re-examination of the other arctic fox specimen from the North Sea, a femur of which the identification is here confirmed and measurements are provided

    Stormwater quality characteristics in (Dutch) urban areas and performance of settlement basins

    Stormwaters, flowing into storm sewers, are known to significantly increase the annual pollutant loads entering urban receiving waters and this results in significant degradation of the receiving water quality. Knowledge of the characteristics of stormwater pollution enables urban planners to incorporate the most appropriate stormwater management strategies to mitigate the effects of stormwater pollution on downstream receiving waters. This requires detailed information on stormwater quality, such as pollutant types, sediment particle size distributions, and how soluble pollutants and heavy metals attach themselves to sediment particles. This study monitored stormwater pollution levels at over 150 locations throughout the Netherlands. The monitoring has been ongoing for nearly 15 years and a total of 7,652 individual events have been monitored to date. This makes the database the largest stormwater quality database in Europe. The study compared the results to those presented in contemporary international stormwater quality research literature. The study found that the pollution levels at many of the Dutch test sites did not meet the requirements of the European Water Framework Directive (WFD) and Dutch Water Quality Standards. Results of the study are presented and recommendations are made on how to improve water quality with the implementation of Sustainable Urban Drainage Systems (SUDS) devices
