    Sticks, balls or a ribbon? Results of a formative user study with bioinformaticians

    User interfaces in modern bioinformatics tools are designed for experts. They are too complicated for\ud novice users such as bench biologists. This report presents the full results of a formative user study as part of a\ud domain and requirements analysis to enhance user interfaces and collaborative environments for\ud multidisciplinary teamwork. Contextual field observations, questionnaires and interviews with bioinformatics\ud researchers of different levels of expertise and various backgrounds were performed in order to gain insight into\ud their needs and working practices. The analysed results are presented as a user profile description and user\ud requirements for designing user interfaces that support the collaboration of multidisciplinary research teams in\ud scientific collaborative environments. Although the number of participants limits the generalisability of the\ud findings, the combination of recurrent observations with other user analysis techniques in real-life settings\ud makes the contribution of this user study novel

    Correlation between choline level and Gd-DTPA enhancement in patients with brain metastases of mammary carcinoma

    Single voxel 1HH double spin-echo MR spectroscopy was used to examine 15 cases of brain metastasis of mammary carcinoma (18 lesions) in relation to Gd-DTPA enhanced MR imaging. For lesions larger than 50% of MRS voxel size, there was significant correlation between Gd-DTPA-enhanced MRI signal and MRS-detected signal of choline (Cho) containing compounds (r = 0.86, P < 0.01; n = 8). The observed loss of correlation when including the smaller lesions was overcome by correcting for partial volume effects (r = 0.69, P < 0.002; n = 18). Metastasis spectra showed increased Cho compared with control spectra, except for those lesions showing detectable lactate (Lact) signal. The detection of Lact in four of the larger lesions coincided with comparatively low levels of creatine (Cr) and Cho and heterogeneous Gd-DTPA enhancement (ring-enhancement). It was concluded that in brain metastases of mammary carcinoma Lact represents a product of ischemia preceding/during tissue decay resulting in central necrosis, rather than tumor specific metabolism resulting in increas

    Reduce fluctuations in capacity to improve the accessibility of radiotherapy treatment cost-effectively

    This paper is motivated by a case study to reduce the throughput times for radiotherapy treatment. The goal is to find a cost-effective way to meet future throughput targets. A combination of queuing theory and computer simulation was used. First, computer simulation to detect the bottleneck(s) in a multi-step radiotherapy process. Despite, the investment in an additional linear accelerator, the main bottleneck turned out to be the outpatient department (OPD). Next, based on queuing theory, waiting times were improved by reducing the fluctuations in the OPD capacity. Computer simulation was used again to quantify the effect on the total throughput time of a radiotherapy patient. The results showed a reduction in both access times as well as waiting times prior to the consecutive steps: the preparation phase and actual treatment. The paper concludes with practical suggestions on how to reduce the fluctuations in capacity, and seems of interest for other radiotherapy departments or other multi-step situations in a hospital

    Increasing number of long-lived ancestors marks a decade of healthspan extension and healthier metabolomics profiles

    Globally, the lifespan of populations increases but the healthspan is lagging behind. Previous research showed that survival into extreme ages (longevity) clusters in families as illustrated by the increasing lifespan of study participants with each additional long-lived family member. Here we investigate whether the healthspan in such families follows a similar quantitative pattern using three-generational data from two databases, LLS (Netherlands), and SEDD (Sweden). We study healthspan in 2143 families containing index persons with 26 follow-up years and two ancestral generations, comprising 17,539 persons. Our results provide strong evidence that an increasing number of long-lived ancestors associates with up to a decade of healthspan extension. Further evidence indicates that members of long-lived families have a delayed onset of medication use, multimorbidity and, in mid-life, healthier metabolomic profiles than their partners. We conclude that both lifespan and healthspan are quantitatively linked to ancestral longevity, making family data invaluable to identify protective mechanisms of multimorbidity.     Families in comparison: An individual-level comparison of life-course and family reconstructions between population and vital event registers

    It remains unknown how different types of sources affect the reconstruction of life courses and families in large-scale databases increasingly common in demographic research. Here, we compare family and life-course reconstructions for 495 individuals simultaneously present in two well-known Dutch data sets: LINKS, based on the Zeeland province’s full-population vital event registration data (passive registration), and the Historical Sample of the Netherlands (HSN), based on a national sample of birth certificates, with follow-up of individuals in population registers (active registration). We compare indicators of fertility, marriage, mortality, and occupational status, and conclude that reconstructions in the HSN and LINKS reflect each other well: LINKS provides more complete information on siblings and parents, whereas the HSN provides more complete life-course information. We conclude that life-course and family reconstructions based on linked passive registration of individuals constitute a reliable alternative to reconstructions based on active registration, if case selection is carefully considered
