    Quantitative determination of active Bowman-Birk isoinhibitors, IBB1 and IBBD2, in commercial soymilks

    Naturally-occurring serine protease inhibitors of the Bowman-Birk family exert their potential chemopreventive and/or therapeutic properties via protease inhibition. In this study, we have quantified the amounts of active BBI isoinhibitors, IBB1 and IBBD2, in six commercial soymilks. By using cation exchange chromatography, the BBI isoinhibitors were isolated and their specific trypsin inhibitory activity was used to estimate their amounts in soymilk samples. IBB1 and IBBD2 concentrations ranged from 0.44 to 5.20 and 0.27 to 4.60 mg/100 ml of soymilk, respectively; total BBI, considered as the sum of both isoinhibitors, ranged from 0.60 to 9.07 mg/100 ml of soymilk. These data show that physiologically relevant amounts of active BBI are present in commercial soymilk and may exert potential health-promoting effects.A.C. acknowledges support by ERDF-co-financed Grant from the Spanish CICYT (AGL2011-26353).     Selective Interactions of Valeriana officinalis Extracts and Valerenic Acid with [3H]Glutamate Binding to Rat Synaptic Membranes

    Although GABA neurotransmission has been suggested as a mechanism for Valeriana officinalis effects, CNS depression can also be evoked by inhibition of ionotropic (iGluR) and metabotropic glutamate receptors (mGluR). In this study, we examined if aqueous valerian extract interacted with glutamatergic receptors. Freshly prepared aqueous valerian extract was incubated with rat cortical synaptic membranes in presence of 20 nM [3H]Glutamate. Aqueous valerian extract increased [3H]Glutamate binding from 1 × 10−7 to 1 × 10−3 mg/mL. In the presence of (2S,1′S,2′S)-2-(Carboxycyclopropyl)glycine (LCCG-I) and (2S,2′R,3′R)-2-(2′,3′-Dicarboxycyclopropyl)glycine (DCG-IV), Group II mGluR agents, valerian extract markedly decreased [3H]Glutamate binding, while (2S)-2-amino-3-(3,5-dioxo-1,2,4-oxadiazolidin-2-yl) propanoic acid) (quisqualic acid, QA), Group I mGluR agonist, increased [3H]Glutamate binding. At 0.05 mg/mL aqueous valerian extract specifically interacted with kainic acid NMDA and AMPA receptors. Valerenic acid, a marker compound for Valeriana officinalis, increased the [3H]Glutamate binding after 1.6 × 10−2 mg/mL, and at 0.008 mg/mL it interacted only with QA (Group I mGluR). The selective interactions of valerian extract and valerenic acid with Group I and Group II mGluR may represent an alternative explanation for the anxiolytic properties of this plant

    A simple scaling analysis of rainfall in Andalusia (Spain) under different precipitation regimes

    A simple scaling analysis was performed in Andalusia (Spain) using daily records from 377 selected stations covering the temporal period between 1870 and 2018. Since Andalusia is a region of considerable climatic variety, with notably wet areas as well as extremely dry zones, this study is useful to investigate the relationship between the simple scaling parameter value and the characteristic rainfall regime of a place. Despite the great correspondence with the average annual precipitation (PRCPTOT), a clear dependence on rainfall irregularity was observed, revealed by the ratio of the maximum daily precipitation and PRCPTOT, as well the wet spells frequency index CWD. The spatial distribution of the simple scaling parameter captured the increasing influence of the Mediterranean Sea towards the East. The easternmost dry areas are clearly influenced by Mediterranean disturbances, with a high proportion of convective rainfall and an irregular rainfall pattern. Using a simple scaling parameter, the generalized equations of the intensity-duration-frequency (IDF) curves, of great hydrological interest were calculated for the eight Andalusian provincial capitals. Moreover, the temporal trends of this parameter in the four past decades were studied in the different areas with the aim of determining if changes in their rainfall patterns due to global warming could be detected.Peer ReviewedPostprint (published version

    A quality control procedure for long-term series of daily precipitation data in a semiarid environment

    The availability of quality precipitation records in the current climate situation is of great importance in the scientifctechnical feld but also for the public institutions that manage the meteorological networks. This work has implemented a comprehensive spatial quality control procedure in the semiarid region of Andalusia (Southern Spain), using precipitation time series from 1947 stations from three meteorological networks: Spanish Meteorological Agency (AEMET), Agroclimatic Information Network of Andalusia (RIA), and Phytosanitary Information Alert Network (RAIF). The method consists of three consecutive steps: basic, absolute, and relative quality control processes. The latter step compares data from neighboring stations taking into account their proximity, height diference, and correlation, leading to a complete evaluation of each daily value. Finally, the quality of each year at each station can be declared as acceptable, good, or excellent. The automatic weather station networks RIA and RAIF gave absolute quality index Q above 85% for almost 87% of their stations, while only 57% of AEMET network reached this percentage. However, one of the longest AEMET datasets, San Fernando-Cádiz, obtained, except for 1 year, Q values over 90% in all available years for more than a century of measurements, since 1870 until 2000. From a total of more than 15 million daily records, almost 82% was fagged as correct. Despite the limitations of Andalusia region (low density of stations and its structural water defcit), the complete quality control procedure has been satisfactorily applied.     Glucose uptake in Prochlorococcus: diversity of kinetics and effects on the metabolism

    We have previously shown that Prochlorococcus sp. SS120 strain takes up glucose by using a multiphasic transporter encoded by the Pro1404 gene. Here, we studied the glucose uptake kinetics in multiple Prochlorococcus strains from different ecotypes, observing diverse values for the Ks constants (15–126.60 nM) and the uptake rates (0.48–6.36 pmol min-1 mg prot-1). Multiphasic kinetics was observed in all studied strains, except for TAK9803-2. Pro1404 gene expression studies during the 21st Atlantic Meridional Transect cruise showed positive correlation with glucose concentrations in the ocean. This suggests that the Pro1404 transporter has been subjected to diversification along the Prochlorococcus evolution, in a process probably driven by the glucose availabilities at the different niches it inhabits. The glucose uptake mechanism seems to be a primary transporter. Glucose addition induced detectable transcriptomic and proteomic changes in Prochlorococcus SS120, but photosynthetic efficiency was unaffected. Our studies indicate that glucose is actively taken up by Prochlorococcus, but its uptake does not significantly alter the trophic ways of this cyanobacterium, which continues performing photosynthesis. Therefore Prochlorococcus seems to remain acting as a fundamentally phototrophic organism, capable of using glucose as an extra resource of carbon and energy when available in the environment

    Characterization of the platelet phenotype caused by a germline RUNX1 Variant in a CRISPR/Cas9-generated murine model

    RUNX1-related disorder (RUNX1-RD) is caused by germline variants affecting the RUNX1 gene. This rare, heterogeneous disorder has no specific clinical or laboratory phenotype, making genetic diagnosis necessary. Although international recommendations have been established to classify the pathogenicity of variants, identifying the causative alteration remains a challenge in RUNX1-RD. Murine models may be useful not only for definitively settling the controversy about the pathogenicity of certain RUNX1 variants, but also for elucidating the mechanisms of molecular pathogenesis. Therefore, we developed a knock-in murine model, using the CRISPR/Cas9 system, carrying the RUNX1 p.Leu43Ser variant (mimicking human p.Leu56Ser) to study its pathogenic potential and mechanisms of platelet dysfunction. A total number of 75 mice were generated; 25 per genotype (RUNX1WT/WT, RUNX1WT/L43S, and RUNX1L43S/L43S). Platelet phenotype was assessed by flow cytometry and confocal microscopy. On average, RUNX1L43S/L43S and RUNX1WT/L43S mice had a significantly longer tail-bleeding time than RUNX1WT/WT mice, indicating the variant's involvement in hemostasis. However, only homozygous mice displayed mild thrombocytopenia. RUNX1L43S/L43S and RUNX1WT/L43S displayed impaired agonist-induced spreading and α-granule release, with no differences in δ-granule secretion. Levels of integrin αIIbβ3 activation, fibrinogen binding, and aggregation were significantly lower in platelets from RUNX1L43S/L43S and RUNX1WT/L43S using phorbol 12-myristate 13-acetate (PMA), adenosine diphosphate (ADP), and high thrombin doses. Lower levels of PKC phosphorylation in RUNX1L43S/L43S and RUNX1WT/L43S suggested that the PKC-signaling pathway was impaired.     Factores habilitantes y limitantes para el éxito de iniciativas de bioeconomía circular en Andalucía

    El informe presenta los resultados de un estudio sobre factores habilitantes y limitantes que se ha llevado a cabo mediante la realización de entrevistas a responsables de empresas que pueden ser consideradas casos de éxito de la bioeconomía circular (BEC) en Andalucía. La muestra ha consistido en 21 empresas representativas desde el punto de vista sectorial, geográfico, de dimensión y de tipo de empresa. Las respuestas se han recogido en un cuestionario semiestructurado para la caracterización de su actividad y para la identificación y determinación de la importancia relativa de los factores habilitantes y limitantes de sus líneas de negocio. Finalmente, se ha agregado la información cuantitativa y cualitativa de los factores analizados y se ha realizado un análisis descriptivo global y sectorial. El sector con mayor presencia de iniciativas es el del olivar y aceite de oliva (seis), seguido del hortofrutícola (cuatro) y la reutilización de agua (dos), con presencia de sectores emergentes como bioestimulantes, microalgas, alimentos funcionales e insectos.Unión Europea 81835

    Allele and haplotype frequencies of HLA-A, -B, -C, -DRB1, -DQB1 and -DQA1 in Castile and Leon region from North West of Spain

    HLA studies have been used to determine the admixture of different populations within the Iberian Peninsula including neighbouring regions with shared origins, such as Portugal and Castile and Leon.
