    A Radio--Optical Reference Frame VIII. CCD observations from KPNO and CTIO: internal calibration and first results

    In this pilot investigation, precise optical positions in the FK5 system are presented for a set of 16 compact extragalactic radio sources, which will be part of the future radio--optical reference frame. The 0.9 m KPNO and CTIO telescopes equipped with 2K CCD's have been used for this project. The astrometric properties of these instruments are investigated in detail. New techniques of using wide field CCD observations for astrometry in general are developed. An internal precision of 5 to 31 mas in position per single exposure is found, depending on the brightness of the object. The tie to the primary optical reference system is established by photographic astrometry using dedicated astrographs on both hemispheres. An accuracy of 30\approx 30 mas per source is estimated for the multi--step reduction procedure when based on the future Hipparcos catalog, while the FK5--based positions suffer from system errors of 100 to 200 mas as compared to the radio positions. This work provides a contribution to the international effort to link the Hipparcos instrumental coordinate system to the quasi--inertial VLBI radio reference frame. Precise radio and optical astrometry of a large sample of compact extragalactic sources will also contribute to the astrophysics of these objects by comparing the respective centers of emission at the optical and radio wavelengths.Comment: AAS v.4 LaTeX, 2 parts on 1 file (main text + deluxetable), accepted by AJ, Dec.95, fig. with reprint

    Enhanced sensitivity of postsynaptic serotonin-1A receptors in rats and mice with high trait aggression

    Individual differences in aggressive behaviour have been linked to variability in central serotonergic activity, both in humans and animals. A previous experiment in mice, selectively bred for high or low levels of aggression, showed an up-regulation of postsynaptic serotonin-1A (5-HT1A) receptors, both in receptor binding and in mRNA levels, in the aggressive line. The aim of this experiment was to study whether similar differences in 5-HT1A receptors exist in individuals from a random-bred rat strain, varying in aggressiveness. In addition, because little is known about the functional consequences of these receptor differences, a response mediated via postsynaptic 5-HT1A receptors (i.e., hypothermia) was studied both in the selection lines of mice and in the randomly bred rats. The difference in receptor binding, as demonstrated in mice previously, could not be shown in rats. However, both in rats and mice, the hypothermic response to the 5-HT1A agonist alnespirone was larger in aggressive individuals. So, in the rat strain as well as in the mouse lines, there is, to a greater or lesser extent, an enhanced sensitivity of postsynaptic 5-HT1A receptors in aggressive individuals. This could be a compensatory up-regulation induced by a lower basal 5-HT neurotransmission, which is in agreement with the serotonin deficiency hypothesis of aggression.

    Huge decreases in the risk of breast cancer relapse over the last three decades

    Introduction The aim of this study was to evaluate local and systemic breast cancer control by comparing the risk of relapse in breast cancer patients in 2003–2004 with that in 1972–1979 and in 1980–1986. Methods About 8,570 women diagnosed with invasive breast cancer in 2003–2004 were selected from the population-based Netherlands Cancer Registry and compared with 133 patients treated in 1972–1979 and 174 in 1980–1986. Five-year risk of relapse was calculated by the Kaplan–Meier method. Cox-proportional hazard models were applied to adjust for tumour size, nodal status and age at diagnosis. Results Patients diagnosed in 2003–2004 had smaller tumours and a less advanced nodal stage than patients diagnosed in 1972–1986. In 1972–1979, 1980–1986 and 2003–2004, treatment included mastectomy in 94%, 72% and 47%; postmastectomy radiotherapy in 75%, 70% and 30%; chemotherapy in 9%, 14% and 37% and hormonal therapy in 3%, 3% and 42% of patients, respectively. Five-year risk of locoregional and distant recurrence decreased from 37% and 34% to 15%, respectively. The 5-year risk of second primary breast cancer did not differ and was 1%, 4% and 2%, respectively. The improved relapse-free survival in patients diagnosed in 2003–2004 as compared with 1972–1979 hardly changed after adjustment (HR = 0.38, 95% CI = 0.28–0.52). Conclusion Over the last decades, local breast cancer therapies have become less rigorous, whereas systemic therapy use has increased. Simultaneously, the risk of breast cancer relapse has tremendously decreased.     The StarScan plate measuring machine: overview and calibrations

    The StarScan machine at the U.S. Naval Observatory (USNO) completed measuring photographic astrograph plates to allow determination of proper motions for the USNO CCD Astrograph Catalog (UCAC) program. All applicable 1940 AGK2 plates, about 2200 Hamburg Zone Astrograph plates, 900 Black Birch (USNO Twin Astrograph) plates, and 300 Lick Astrograph plates have been measured. StarScan comprises of a CCD camera, telecentric lens, air-bearing granite table, stepper motor screws, and Heidenhain scales to operate in a step-stare mode. The repeatability of StarScan measures is about 0.2 micrometer. The CCD mapping as well as the global table coordinate system has been calibrated using a special dot calibration plate and the overall accuracy of StarScan x,y data is derived to be 0.5 micrometer. Application to real photographic plate data shows that position information of at least 0.65 micrometer accuracy can be extracted from course grain 103a-type emulsion astrometric plates. Transformations between "direct" and "reverse" measures of fine grain emulsion plate measures are obtained on the 0.3 micrometer level per well exposed stellar image and coordinate, which is at the limit of the StarScan machine.Comment: 24 pages, 8 figures, accepted for PAS

    p53 overexpression is a predictor of local recurrence after treatment for both in situ and invasive ductal carcinoma of the breast

    Background. Several biological markers have been related to prognosis in mammary ductal carcinoma. The aim of the study was to determine biological markers that could predict local recurrence following treatment for all stages of primary operable ductal carcinoma of the breast. Materials and methods. A consecutive series of patients treated for pure ductal carcinoma in situ (DCIS, n = 110) and invasive ductal carcinoma (IDC, n = 243) was studied. Twenty-three patients with DCIS were excluded because of lack of original paraffin embedded tissue. All patients had been treated between July 1996 and December 2001. Median follow-up was 49.8 mo. From the original paraffin embedded tumors, tissue microarrays (TMAs) were constructed. On these TMAs, immunohistochemistry was performed for estrogen-receptor (ER), progesterone-receptor (PR), Her2/neu, p53, and cyclin D1. Main outcome was the event of LR. All analyses were stratified for diagnosis (DCIS or IDC) and pathological grade. Results. In univariate analyses, Her2/neu overexpression (hazard ratio [HR] 3.1, 95% confidence interval [CI] 1.1-8.7, P = 0.032) and p53 overexpression (HR 3.5, 95% Cl 1.3-9.3, P = 0.014) were associated with LR in patients treated for both DCIS and IDC. In multivariate analysis, p53 overexpression (HR 3.0, 95% CI 1.1-8.2, P = 0.036 and HR 4.4,95% Cl 1.5-12.9, P = 0.008) and adjuvant radiotherapy (HR 0.2, 95% Cl 0.1-0.8, P = 0.026) were independent common predictors of LR in patients who had received treatment for both DCIS and IDC. Conclusions. p53 overexpression is a common predictor of LR following treatment for all stages of primary operable ductal carcinoma of the breast.     Managing boundaries in multiteam structures:From parochialism to integrated pluralism

    Multiteam structures are increasingly used to coordinate complex tasks between different groups. To realize this potential, however, the members of a multiteam structure must manage a complex set of boundary relations within, between, and beyond the various constituent teams—boundary relations that can be cooperative, competitive, or some combination of both at the same time. This multimethod study provides insight into how multiteam structures can meet this challenge. Specifically, we examined how the different organizations that utilize and support the Dutch railway system learned to manage boundaries as they transitioned from a centralized, arms-length structure to a colocated, multiteam structure for coordinating disruption responses (i.e., the Rail Operations Control Center (ROCC)). In part 1 of our study, qualitative analyses of interview, observational, and archival data suggested that learning to manage boundaries within the ROCC was not simple or linear but evolved through trial and error during various phases. Ultimately, the ROCC developed an approach we call “integrated pluralism,” establishing a dynamic balance that combines both collaborative and competitive approaches to boundary management. In this manner, the ROCC teams were able to attain integrated solutions and coordinated task accomplishment while simultaneously defending internal team operations and home organization interests. In part 2, we employed an interrupted time series analysis to demonstrate that the implementation of the ROCC resulted in significant performance improvements. Consistent with the results of part 1, we found that these improvements emerged gradually over time as teams learned to work out their boundary relations and transitioned to integrated pluralism. These findings provide new insights into how individuals and teams can work together to tackle the unique boundary management challenges presented by multiteam structures and illuminate the dynamic trial and error process by which component teams can learn to both cooperate and compete

    Heeding Supply Chain Disruption Warnings:When And How Do Cross‐Functional Teams Ensure Firm Robustness?

    Firms can adopt several strategies to increase their robustness to potential supply chain (SC) disruptions. One promising strategy is the use of a cross-functional team with representatives from functional departments. Such a team may facilitate sharing relevant information, enabling the firm to respond effectively to SC disruption warnings. However, despite their potential, cross-functional teams also differ in their ability to respond to SC disruption warnings and to ensure firm robustness. Extending insights from information-processing theory and team research to the field of SC management, we propose that a cross-functional team’s ability to handle high numbers of SC disruption warnings depends on the extent to which the team adopts centralized decision-making, with one or two members orchestrating the decision-making process. We also introduce internal integration problems as a mediating mechanism explaining why a cross-functional team lacking centralized decision-making may be unable to handle high numbers of SC disruption warnings. In two independent studies, we use multi-source data on cross-functional teams’ performance in dealing with SC disruption warnings during a realistic SC management simulation; the results support our predictions

    RAB25 expression is epigenetically downregulated in oral and oropharyngeal squamous cell carcinoma with lymph node metastasis

    Oral and oropharyngeal squamous cell carcinoma (OOSCC) have a low survival rate, mainly due to metastasis to the regional lymph nodes. For optimal treatment of these metastases, a neck dissection is required; however, inaccurate detection methods results in under- and over-treatment. New DNA prognostic methylation biomarkers might improve lymph node metastases detection. To identify epigenetically regulated genes associated with lymph node metastases, genome-wide methylation analysis was performed on 6 OOSCC with (pN+) and 6 OOSCC without (pN0) lymph node metastases and combined with a gene expression signature predictive for pN+ status in OOSCC. Selected genes were validated using an independent OOSCC cohort by immunohistochemistry and pyrosequencing, and on data retrieved from The Cancer Genome Atlas. A two-step statistical selection of differentially methylated sequences revealed 14 genes with increased methylation status and mRNA downregulation in pN+ OOSCC. RAB25, a known tumor suppressor gene, was the highest-ranking gene in the discovery set. In the validation sets, both RAB25 mRNA (P = 0.015) and protein levels (P = 0.012) were lower in pN+ OOSCC. RAB25 mRNA levels were negatively correlated with RAB25 methylation levels (P < 0.001) but RAB25 protein expression was not. Our data revealed that promoter methylation is a mechanism resulting in downregulation of RAB25 expression in pN+ OOSCC and decreased expression is associated with lymph node metastasis. Detection of RAB25 methylation might contribute to lymph node metastasis diagnosis and serve as a potential new therapeutic target in OOSCC
