    Role of the protein annexin A1 on the efficacy of anti-TNF treatment in a murine model of acute colitis

    TNF-α is involved in the mechanisms that initiate inflammatory bowel diseases (IBDs). Anti-TNF-α drugs, such as infliximab (IFX), cause non-responsiveness and side effects, indicating the need to investigate alternative therapies for these diseases. The anti-inflammatory protein, annexin A1 (AnxA1), has been associated with the protection of the gastrointestinal mucosa. To further address the role of endogenous AnxA1 on the TNF-α blockade efficacy in a murine model, we assessed colitis induced by Dextran Sulfate Sodium (DSS) in wild-type (WT) and AnxA1−/− Balb/c mice treated with IFX. We consistently observed endogenous AnxA1 prevented clinical and physiological manifestations of experimental colitis treated with IFX, additionally the manifestation of the disease was observed earlier in AnxA1−/− mice. Rectal bleeding, diarrhea, histological score, epithelial damages and collagen degradation caused by DSS were prevented following IFX treatment only in WT mice. IL-6 increased during colitis in WT and AnxA1−/− mice, decreasing under IFX treatment in WT. The influx of neutrophils and TNF-α secretion were largely elevated in AnxA1−/− mice when compared to WT mice. In the group WT/DSS + IFX, phagocytes were more susceptible to apoptosis following treatment with IFX. Endogenous expression of AnxA1 increased after DSS and decreased with IFX treatment, demonstrating an attenuated inflammatory response. The data indicate that AnxA1 contributes to the establishment of intestinal homeostasis after blocking of TNF-α was used as a treatment of IBD, constituting a key molecule in the mechanism of action and a potential biomarker of therapeutic efficacy.Fil: De Paula Silva, Marina.     Preservation of probiotic strains isolated from kefir by spray drying

    Aims: This work aims to investigate the survival of Lactobacillus kefir CIDCA 8348, Lactobacillus plantarum CIDCA 83114 and Saccharomyces lipolytica CIDCA 812, all isolated from kefir, during spray drying and subsequent storage. Methods and Results: Micro-organisms were grown in De Man, Rogosa, Sharpe (MRS) or yeast medium (YM) medium and harvested in the stationary phase of growth. The thermotolerance in skim milk (D and Z values), the survival of spray drying at different outlet air temperatures and subsequent storage in different conditions during 150 days were studied. The resistance to the heat treatments was higher in Lact. plantarum compared to Lact. kefir and S. lipolytica. The three micro-organisms studied varied considerably in their ability to survive to spray drying processes. Lactobacillus plantarum showed the highest survival rate for all the tested outlet air temperatures and also to the further storage in the dried state. The survival rates of Lact. kefir and S. lipolytica through drying and subsequent storage in the dried state decreased when the drying outlet air temperatures increased. Conclusions: Spray drying is a suitable method to preserve micro-organisms isolated from kefir grains. A high proportion of cells were still viable after 80 days of storage at refrigerated temperatures Significance and Impact of Study: It is the first report about spray-dried probiotic strains isolated from kefir grain and contributes to the knowledge about these micro-organisms for their future application in novel dehydrated products.Fil: Golowczyc, Marina Alejandra.     Formyl Peptide Receptors and Annexin A1: Complementary Mechanisms to Infliximab in Murine Experimental Colitis and Crohn's Disease

    Non-responsiveness to anti-TNF-α therapies presents relevant rates in inflammatory bowel disease patients, presenting the need to find biomarkers involved in therapeutic efficacy. Herein, we demonstrate that higher levels of colonic formyl peptide receptor 1 and annexin A1 correlate with histological recovery in Crohn’s disease patients under remission. Using the dextran sulfate sodium colitis model in mice, we suggest that infliximab induces annexin A1 expression and secretion in activated intestinal leukocytes. Conversely, this mechanism might stimulate epithelial formyl peptide receptors, inducing wound healing and consequent histological remission. Our data indicate that assessing intestinal expressions of formyl peptide receptors and annexin A1 might provide precious information on the disease activity and responsiveness to infliximab in inflammatory bowel disease patients

    EDUCAÇÃO NUTRICIONAL COM MULHERES PARTICIPANTES DO CURSO DE APRENDIZAGEM GESTACIONAL NA ASSOCIAÇÃO DE PAIS E AMIGOS DOS EXCEPCIONAIS DE MONTE AZUL PAULISTA - SP

    A educação alimentar da gestante é um quesito muito importante nesta fase da vida, pois uma alimentação adequada é fator de proteção a diversas complicações que podem ocorrer durante e após a gestação. Com advento da má alimentação e tabus alimentares, surgiu a iniciativa de promover uma estratégia, a educação alimentar, levando conhecimentos básicos a gestantes do município de Monte Azul Paulista-SP no intuito de criar o discernimento entre o que é verídico e o que é mito entre os alimentos. Para isso foi realizado como instrumento de avaliação um questionário validado conhecido por Escala de Conhecimento Nutricional, obtendo assim o conhecimento prévio das gestantes quanto ao tema abordado. Em seguida, as participantes receberam uma palestra ministrada sobre os principais objetivos acima citados, utilizando para isso caderneta da gestante e atualizações do Ministério da Saúde. Este trabalho teve por objetivo orientar as gestantes quanto à alimentação saudável, evidenciando práticas para amenizar os sintomas durante a gravidez e discutindo também sobre mitos alimentares. Ao final, o questionário foi reaplicado para comparação dos resultados, onde os dados encontrados mostraram que as mulheres atendidas no grupo de gestante da Associação de Pais e Amigos de Monte Azul Paulista, apresentaram muita dúvida quanto ao valor nutricional dos alimentos. No primeiro momento de avaliação, o grupo apresentou 62,5% de acertos e após a palestra esse mesmo número passou para 87,5%.  Assim é possível concluir que a orientação quanto à alimentação saudável se faz necessária nessa fase da vida e apresenta um resultado eficaz para o conhecimento das gestantes

    The aim of this in vitro study was to evaluate some parameters of dental etching when irradiated with an erbium, chromium:yttrium-scandium-gallium-garnet (Er,Cr:YSGG) laser. One-hundred sound human third molars were selected and randomly distributed into ten groups (n = 10). The class V cavities of group 1 (control) were prepared with a bur and etched with 37% phosphoric acid, while groups G2 to G10, were prepared with laser (5 W, 88.46 J/cm(2), 90/70% air/water) and etched with the following powers: G3 and G4, 0.25 W; G5 and G6, 0.5 W; G7 and G8, 0.75 W; G9 and G10, 1 W. Group G2 received no laser etching. Prior to restoration, G2, G4, G6, G8 and G10 received acid etching. After restoration, all samples were submitted to a microleakage test. According to statistical analysis (Kruskal-Wallis and Dunn`s tests), G10 presented the lowest microleakage values (P < 0.05). The other groups showed no differences between them.     Granulocytic sarcoma of the oral cavity in a chronic myeloid leukemia patient : an unusual presentation

    Intraoral granulocytic sarcoma is an unusual manifestation of chronic or acute leukemia. The oral manifestations often involve enlargements of the gingival and mucosal tissue from direct leukemic cell infiltration. Only 38 cases have been reported in scientific literature to date. We present the case of a 47 year-old female who was diagnosed with chronic myeloid leukemia (CML) in December 2006. She was referred to a dentist for further evaluation, revealing generalized gingival overgrowth as well as periodontal, apical disease, and bleeding of the gums. An oral biopsy was performed and histological features revealed immature blast-like cells
