    O Tratado de não Proliferação de Armas Nucleares e o Desafio Imposto pelo seu Direito de Retirada: um Estudo do Problemático Caso Norte-Coreano

    The North Korean nuclear issue, identified by many as the greatest post-Cold War diplomatic challenge and considered one of the greatest failures in the history of American diplomacy, offers a challenge to the Treaty on the Non-Proliferation of Nuclear Weapons and jeopardizes the current non-proliferation regime. Despite having exposed the international community to risks since the beginning of its nuclear claims, the case of North Korea really became problematic when the country announced in 2003 its withdrawal from the Treaty. However, this was not the first time that Pyongyang declared withdrawal from the NPT: in 1993 the country had already announced the withdrawal, however, just one day before its effectuation - given that it only enter into force three months after the announcement, Kim Jong-il declared the withdrawal announcement suspended. Due to the sui generis situation introduced to the nuclear scenario by the North Koreans, the objective of this research is to analyze the possible interpretations to the following question: after all, what is the current status of North Korea before the NPT?O caso nuclear norte-coreano, identificado por muitos como o maior desafio diplomático pós- Guerra Fria e considerado um dos mais notáveis fracassos da história da diplomacia norte- americana, oferece um grande desafio ao Tratado de Não Proliferação de Armas Nucleares e coloca em risco o atual regime de não-proliferação. Apesar de ter exposto a comunidade internacional a riscos desde o início de suas pretensões nucleares, o caso da Coréia do Norte se tornou realmente problemático quando o país anunciou, em 2003, a sua retirada do Tratado. Contudo, essa não foi a primeira vez que Pyongyang denunciou o TNP: em 1993 o país já havia anunciado a sua retirada, contudo, apenas um dia antes da sua efetivação  tendo em vista que a mesma somente entra em vigor três meses após o anúncio, Kim Jong-il declarou suspenso o anúncio de retirada. Devido à situação sui generis introduzida ao cenário nuclear pelos norte-coreanos, o objetivo desta pesquisa é analisar as possíveis interpretações ao seguinte questionamento: a final, qual é o atual status da Coréia do Norte perante o TNP

    Genótipo e expressão da proteína fluorescente verde melhorada em ratos da linhagem LEW-Tg (EGFP) F455.5/Rrrc

    The Green fluorescent protein (GFP) was first described after being extracted from Aequorea victoria in 1987. Since then, GFP and its derivatives have been widely used in several experiments as cell and protein marker. In the present study it was verified the genotype of the offspring from crosses between heterozygote Lewis LEW-Tg (EGFP) F455.5/ Rrrc rats and analyzed the expression of the enhanced green fluorescent protein (EGFP) in different cell types and genotypes. The genotype of the offspring was assessed by PCR and analysis of EGFP expression in different cells and genotypes, including mesenchymal stem cells (MSC) derived from adipose tissue and calvarial osteoblast cells. Expression of EGFP was verified by flow cytometry, fluorescence microscopy, and immunostaining. Through these methods, it was identified the genotypes of the offspring and determined the levels of expression of EGFP in two cell types. A difference in expression between the (EGFP +/+) and (EGFP +/-) genotypes was also observed in addition to the presence of autofluorescence. Further studies on the natural fluorescence of cells with the (EGFP +/-) genotype and that induced by presence of the EGFP are necessary.A proteína fluorescente verde (GFP) foi descrita pela primeira vez após ter sido extraída de Aequorea victoria em 1987. Desde então, a GFP e seus derivados têm sido amplamente utilizados em várias experiências como marcador celular e de proteínas. O objetivo do presente estudo foi o de verificar o genótipo dos descendentes de cruzamentos entre ratos Lewis LEW-Tg (EGFP) F455.5/Rrrc heterozigotos e de analisar a expressão da proteína fluorescente verde melhorada (EGFP) em diferentes tipos celulares e genótipos. O genótipo da descendência foi avaliado por PCR e pela análise da expressão da EGFP em diferentes células e genótipos, incluindo-se as células-tronco mesenquimais (MSC) derivadas de tecido adiposo e de osteoblastos de calvária. A expressão da EGFP foi verificada por citometria de fluxo, microscopia de fluorescência e imunocoloração. Foram, identificados os genótipos da descendência e determinados os níveis de expressão de EGFP em dois tipos de células. Foi também constatada uma diferença de expressão entre os genótipos (EGFP +/+) e (EGFP +/-) além da presença de autofluorescência. Mais estudos são necessários para esclarecer a fluorescência natural de células com o genótipo (EGFP +/-) e aquela induzida pela presença da EGFP

    SWCNT-porphyrin nano-hybrids selectively activated by ultrasound: an interesting model for sonodynamic applications

    Sonodynamic therapy (SDT) is an innovative anticancer approach, based on the excitation of a given molecule (usually a porphyrin) by inertial acoustic cavitation that leads to cell deathviathe production of reactive oxygen species (ROS). This study aims to prepare and characterize nanosystems based on porphyrin grafted carbon nanotubes (SWCNTs), to understand some aspects of the mechanisms behind the SDT phenomenon. Three different porphyrins have been covalently linked to SWCNTs using either Diels-Alder or 1,3-dipolar cycloadditions. ROS production and cell viability have been evaluated upon ultrasound irradiation. Despite the low porphyrin content linked on the SWCNT, these systems have shown high ROS production and high tumour-cell-killing ability. The existence of a PET (photoinduced electron transfer)-like process would appear to be able to explain these observations. Moreover, the demonstrated ability to absorb light limits the impact of side effects due to light-excitation

    Dual-regulated lentiviral vector for gene therapy of X-linked chronic granulomatosis

    Regulated transgene expression may improve the safety and efficacy of hematopoietic stem cell (HSC) gene therapy. Clinical trials for X-linked chronic granulomatous disease (X-CGD) employing gammaretroviral vectors were limited by insertional oncogenesis or lack of persistent engraftment. Our novel strategy, based on regulated lentiviral vectors (LV), targets gp91(phox) expression to the differentiated myeloid compartment while sparing HSC, to reduce the risk of genotoxicity and potential perturbation of reactive oxygen species levels. Targeting was obtained by a myeloid-specific promoter (MSP) and posttranscriptional, microRNA-mediated regulation. We optimized both components in human bone marrow (BM) HSC and their differentiated progeny in vitro and in a xenotransplantation model, and generated therapeutic gp91(phox) expressing LVs for CGD gene therapy. All vectors restored gp91(phox) expression and function in human X-CGD myeloid cell lines, primary monocytes, and differentiated myeloid cells. While unregulated LVs ectopically expressed gp91(phox) in CD34(+) cells, transcriptionally and posttranscriptionally regulated LVs substantially reduced this off-target expression. X-CGD mice transplanted with transduced HSC restored gp91(phox) expression, and MSP-driven vectors maintained regulation during BM development. Combining transcriptional (SP146.gp91-driven) and posttranscriptional (miR-126-restricted) targeting, we achieved high levels of myeloid-specific transgene expression, entirely sparing the CD34(+) HSC compartment. This dual-targeted LV construct represents a promising candidate for further clinical development
